Résumé

Background In factor XI (FXI) deficiency, bleeding cannot be predicted by routine analyses. Since FXI is involved in tissue factor (TF)-independent propagation loop of coagulation, we hypothesized that investigating the spatiotemporal separated phases of coagulation (TF-dependent and -independent) could improve diagnostics.

Coagulation sanguine

La coagulation sanguine est un processus complexe aboutissant à la formation de caillots sanguins. C’est une partie importante de l’hémostase où la paroi endommagée d’un vaisseau sanguin est couverte d’un caillot de fibrine, ce qui a pour conséquence d'arrêter l’hémorragie. Les troubles de la coagulation qui mènent à des risques de saignements plus importants sont appelés hémophilie. D'autres troubles de la coagulation peuvent mener à un plus grand risque de thrombose.

Facteur Rosenthal

Le facteur Rosenthal ou facteur XI est une enzyme intervenant dans la coagulation sanguine. Il est un des (le plus souvent des enzymes, numérotés en chiffres romains) de la cascade de coagulation. Le dosage est exprimé en pourcentage du temps de céphaline du plasma du patient par rapport à celui du plasma d'un témoin sain, la valeur normale est donc de 100% avec des variations normales attendues entre 70 et 120%. Il existe des variations physiologiques normales à la baisse chez la femme enceinte et le nouveau-né.

Haemophilia C (also known as plasma thromboplastin antecedent (PTA) deficiency or Rosenthal syndrome) is a mild form of haemophilia affecting both sexes, due to factor XI deficiency. It predominantly occurs in Ashkenazi Jews. It is the fourth most common coagulation disorder after von Willebrand's disease and haemophilia A and B. In the United States, it is thought to affect 1 in 100,000 of the adult population, making it 10% as common as haemophilia A.
