Résumé

The present invention relates to a method for preparing induced hepatic progenitor cells (iHPC) comprising the steps of dedifferentiation of hepatocytes by culture with a culture medium comprising at least one activator of the Wnt signaling, Basic fibroblast growth factor (b-FGF), and Epidermal growth factor (EGF).

Basic fibroblast growth factor

Fibroblast growth factor 2, also known as basic fibroblast growth factor (bFGF) and FGF-β, is a growth factor and signaling protein encoded by the FGF2 gene. It binds to and exerts effects via specific fibroblast growth factor receptor (FGFR) proteins, themselves a family of closely related molecules. Fibroblast growth factor protein was first purified in 1975; soon thereafter three variants were isolated: 'basic FGF' (FGF2); Heparin-binding growth factor-2; and Endothelial cell growth factor-2.

FGF23) . Ce sont des protéines qui activent la migration et la multiplication de cellules cibles. Ces facteurs sont généralement sécrétés par des fibroblastes.

Facteur de croissance épidermique

Le facteur de croissance épidermique (EGF, de l'anglais epidermal growth factor) est une hormone protéique aux multiples actions, principalement trophiques. Son gène est EGF, situé sur le chromosome 4 humain. Son site d'action ne se résume pas au tissu épidermique mais plutôt à l'ensemble des tissus ; l'adjectif épidermique est historique. La fixation de cette hormone sur le récepteur de l'EGF provoque une activité mitotique très rapide au sein des tissus ciblés.
