Identification of genetic insulator elements to increase the safety of viral gene therapy vectors

Haematopoeitic system diseases, both acquired and inherited, can be currently cured by allogeneic haematopoietic stem cell transplantation. This treatment provides highly successful immune function recovery for patients receiving grafts of HLA-compatible donors but has still a great risk of complications and even failure if no suitable donor is available. Among the alternative therapeutic options, ex vivo retrovirus-mediated gene transfer into haematopoietic progenitor cells has been shown to be an efficient strategy for a substantial number of severe combined immunodeficiency-suffering patients. A recent gene therapy trial has been remarkably effective for the immunological reconstitution of patients suffering from X-linked severe combined immunodeficiency. This treatment was able to provide full correction of disease phenotype and thus, clinical benefit. However, the appearance of leukemia in several patients has put in question the safety of the procedure. This severe adverse event has been attributed to the integration of the therapeutic transgene-carrying viral vector into a known T-cell oncogene, LMO2, thereby contributing to the development of T-cell leukemia by causing aberrant expression of LMO2. Further studies mentioned the possible retroviral-mediated cis-activation of the LMO2 promoter underlying the potential ability of retroviral regulatory elements to influence neighboring gene transcription. This project aimed at decreasing the risk associated with the use of viral vectors for gene therapy through the identification of genetic insulator elements capable of isolating the vector regulatory elements in order to prevent the activation of chromosomal genes by the viral enhancers. We have established a standardized screening procedure whereby the potency of insulators can be assessed quantitatively on relevant vector elements. This assay system consists of a series of plasmids containing two reporter genes: one mimicking a therapeutic gene under the control of strong viral long terminal repeat (LTR) enhancer, and the other one standing for an endogenous gene close to the chromosomal vector integration site. The assay allowed the quantification of the enhancer blocking activity of the well-characterized chicken beta-globin 5'HS4 insulator (cHS4) in cultured cells. We assessed the insulator activity of novel synthetic elements, constructed from optimized binding sites for the insulator protein CTCF. In addition, we demonstrated the enhancer blocking activity of the nuclear factor 1 protein family (CTF/NF1) and showed that it also displays barrier properties, protecting transgene expression from silencing. We showed that both CTCF and CTF/NF1 binding sites act as insulators that mediate potent enhancer-blocker activity, resulting in a commensurate reduction of genotoxicity when implemented in viral gene therapy vectors. Finally, we used the same approach to analyze the enhancer blocking activity of well-known chromatin domain delimiters, matrix attachment regions (MAR), mainly characterized by their barrier properties. We could demonstrate the enhancer blocking ability of the 1-68 MAR that is mainly harbored by an A-T rich core sub-region.

Transgenic SOD1 mice as a model for developing amyotrophic lateral sclerosis therapies

Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative disease characterized by a loss of motor neurons in the brain (brainstem and cortex) and the spinal cord that leads to a motor neurological symptomatology. Approximately 10% of ALS cases have a familial form of ALS. Among the familial cases, 20% are caused by dominantly inherited mutations (around 100 different mutations) in the protein Cu/Zn superoxide dismutase (SOD1). To date, genetic ALS models on mutated SOD1 (SOD1G93A, SOD1G85R and SOD1G37R) transgenesis in rodents have been successful in recapitulating the main features of the human pathology, especially the loss of spinal or facial motoneurons, the increased astrogliosis, the activation of microglia and the cellular and molecular disruptions in motoneurons. The therapies toward a treatment or cure for ALS have been met with limited success. In the present study, SOD1G93A transgenic mice have been used to test new gene therapies to slow disease progression and pharmacological approach to obtain a better comprehension of the pathological process, more precisely, to define the involvement of microglial activation in the spread of the disease. For the approach by gene therapy, injection of viral vectors in a localized region of central nervous system (CNS) constitutes an excellent tool for the development of new therapies. In the present study, the viral-mediated expression of potential neuroprotective factors was explored in the lumbar spinal cord or facial nucleus of SOD1G93A transgenic mice. As HIV-1-derived lentiviral vectors can efficiently transduce neurons in CNS, these retroviral vectors were used to over-express the neurotrophic factor GDNF (glial cell line-derived neurotrophic factor) to supply a trophic support to lumbar and facial motoneurons, or RNAi molecules specifically targeting the SOD1G93A gene to inhibit the mutant SOD1 expression, the cause of the disease, in the lumbar spinal cord of SOD1G93A transgenic mice. For the study of the level of microglial involvement in the pathology, SOD1G93A transgenic mice had a daily pharmacological treatment with an inhibitor of microglial activation, the minocycline. The present thesis demonstrates that, on the one hand, at the difference of laboratories that delayed the disease progression by intramuscular injection of adenoviral vectors or adenoassociated vectors encoding for GDNF, the intraspinal injection of lentivirus encoding for GDNF did not lead to a protection of lumbar motoneurons and a delay of the disease. But this study showed a significantly rescue of facial motoneurons by the injection of lentivirus encoding for GDNF directly in facial nucleus. A selective vulnerability between motoneurons in facial nucleus and these in lumbar spinal cord has been therefore underlined. On the other hand, we showed that the therapeutic approach to inhibit the mutant SOD1 expression look as the most effective to delay the progression of the ALS pathology. Both studies of gene therapy, compared to others studies using the same therapeutic factors, demonstrated clearly that to optimize a therapeutic treatment, it seems better to act at muscular junction level with a retrograde transport form muscle to motoneuronal bodies of the therapeutic molecule or viral vector. Nevertheless, motoneuron-restricted silencing of mutant SOD1 would be useful for better understanding of neuronal and glial cellular mechanism mediating ALS-linked mutated SOD1 toxicity. Regarding the involvement of microglia in the pathological process, the inhibition of microglial activation by the minocycline treatment, suggests, on the one hand, that microglial activation do not initiate the pathological process and that this microgliosis might be a consequence and triggered by multiple disruptions. On the other hand, that minocycline could protect completely facial motoneurons and partially spinal motoneurons by its anti-apoptotic property. Therapeutic approaches aiming to act directly on the cause of the disease as well as a better comprehension of the role of non-neuronal cells in the pathological process may therefore open new perspectives for the treatment of ALS pathology.

Characterizing molecular recognition principles of specific odorant molecule interactions with an olfactory receptor and a nuclear hormone receptor

Olfaction, the detection of odorous chemicals in the environment, is one of the oldest mammalian sensory systems and involves large number (up to 1000) of distinct G protein coupled olfactory receptors (OR). The chemical interaction of volatile molecules with specific ORs marks the primary step in odor perception and is of critical importance for elucidating the molecular basis of detecting and discriminating thousands of odorous chemicals. Despite the fact that a number of putative ORs have been cloned, only limited progress has been made in assigning odorant molecules to specific receptors. Furthermore, the analysis of OR–ligand interactions at a cellular level is complicated by combinatorial recognition principles and thus requires large scale odorant and receptor screening to establish receptor-specific ligand profiles. Described herein are efforts towards the development of reliable and efficient assays for detecting and quantifying the responses of heterologously expressed ORs to odorants. Three strategies were employed for the optical or electrophysiological detection of ORs responses with functional assays in HEK293 cells. In two approaches, OR activation was monitored via the endogenous cAMP pathway, which was coupled either to the cAMP response element-mediated expression of different reporter genes, or to a cyclic nucleotide gated ion channel. In a third approach, OR activation was measured using Ca2+ imaging in the presence of a promiscuous Gαq protein. ORs of different species were then functionally analyzed using the different assays. A library of 250 odorant compounds representative of the chemical functional groups relevant in perfumery was screened; 16 novel specific ligands for the mouse eugenol odorant receptor (EGOR) were identified. Ten of these compounds are structurally unrelated to previously identified ligands and thus provide an extended molecular basis for describing the ligand-specificity and selectivity of EGOR in new detail. Interestingly, some ORs exhibited odorant responses in the cAMP-based assays, but not in Ca2+ imaging assays and vice-versa, which indicates a possible preferential OR coupling to specific G proteins. Such preferential coupling might be dependent on the cell type or influenced by specific odorant molecules and could potentially increase the complexity in odorantmediated signaling. A second part of this thesis describes investigations into other potential activities of odorants apart from their role in olfaction. An odorant compound library was screened to determine possible xenoestrogenic activity on human ERα expressed in cultured mammalian cells. An image-based assay to observe the nuclear redistribution processes of a functional YFP-ERα fluorescent chimera allowed the identification of seven odorant molecules that interact with the receptor. As the optical detection of YFP-ERα redistribution processes did not distinguish between agonistic and antagonistic effects, we tested the active substances in a reporter assay using luciferase under transcription control of two estrogen response elements (ERE). Two active odorants identified in the screening exhibited agonist activity as indicated by a dose-dependent activation of ERα mediated transcription of the luciferase reporter gene.

A conserved transcriptional enhancer that specifies Tyrp1 expression to melanocytes

Pigment cells of mammals originate from two different lineages: melanocytes arise from the neural crest, whereas cells of the retinal pigment epithelium (RPE) originate from the optic cup of the developing forebrain. Previous studies have suggested that pigmentation genes are controlled by different regulatory networks in melanocytes and RPE. The promoter of the tyrosinase-related family gene Tyrp1 has been shown to drive detectable transgene expression only to the RPE, even though the gene is also expressed in melanocytes as evident from Tyrp1-mutant mice. This indicates that the regulatory elements responsible for Tyrp1 gene expression in the RPE are not sufficient for expression in melanocytes. We thus searched for a putative melanocyte-specific regulatory sequence and demonstrate that a bacterial artificial chromosome (BAC) containing the Tyrp1 gene and surrounding sequences is able to target transgenic expression to melanocytes and to rescue the Tyrp1b (brown) phenotype. This BAC contains several highly conserved non-coding sequences that might represent novel regulatory elements. We further focused on a sequence located at -15 kb, which we identified as a melanocyte-specific enhancer as shown by cell culture and transgenic mice experiments. In addition, we show that the transcription factor Sox10 can activate this conserved enhancer. The presence of a distal Tyrp1 regulatory element, which specifies melanocyte-specific expression, supports the idea that separate regulatory sequences can mediate differential gene expression in melanocytes and RPE.
