New Analytical Methods for Size Fractionated, Quantitative, and Element Specific Analysis of Metallic Engineered Nanoparticles in Aerosols and Dispersions

The application of engineered nanoparticles (ENP) increased almost exponentially during the last decade. Next to carbon based ENP, metal based ENP are the next most commonly employed. Utilization of such particles for medical applications, remediation of toxic substances, consumer products, new materials, and numerous other fields has been reported already. Despite their extraordinary success and implementation, concerns have been raised about possible negative effects in humans and the environment. Once released, ENP are easily distributed via the atmosphere and the aquatic environment where an exposure to organisms can occur. To understand ENP release, life cycle (fate), and exposure as well as to evaluate their toxicological mechanisms, appropriate, media-specific (aerosols and dispersions) analytical methods are required. First, the development of a new set-up to determine ENP in aerosols is presented. For this purpose online (SMPS) and offline (electrostatic sampling on TEM grids) aerosol particle measurement techniques were combined. The aim was to study the release and fate of metallic ENP from commercial available spray products. Method development and verification was performed employing a well characterized aqueous Ag-NP spray product. The investigations have shown that the method offers the possibility to determine size, and element -specific nanoparticle release. Furthermore information about particle morphology and quantity could be obtained. It was revealed, that the release of nanoparticles strongly depends on the type of spray. Pump sprays did not show to produce airborne ENP in the aerosol, whereas propellant gas sprays released nanoparticles in substantial quantities. The release of nanoparticles was correlated with the droplet size distributions generated from the different spray types. Following method development, the setup was applied to determine ENP release of commercial available sprays, indicated to contain ENP. The obtained data was useful to model the consumer exposure of ENP from such products. Second, a set-up to determine ENP in dispersions was developed. For this purpose an asymmetric flow field flow fractionation (A4F) apparatus was coupled to an UV/Vis, light scattering- and inductively coupled plasma mass spectrometric (ICPMS) detector, simultaneously. In the beginning, method development and systematic investigations of prospects and limitations was performed employing well-characterized Au nanoparticles (Au-NP). The investigation revealed that A4F is a versatile separation technique for metallic ENP. In contrast, size determination with the A4F system, derived from the retention times of nanoparticles in the chromatogram, was erroneous. Unspecific interaction of ENP with the A4F channel membrane led to retention time shifts resulting in biased size information. For this reason, online dynamic light scattering (DLS) was employed and it offered a reliable determination of nanoparticle dimensions. Determination of nanoparticle concentration also proved to be to be problematic. Unspecific loss of nanoparticles in the A4F separation channel led to insufficient recovery prevented direct quantification of nanoparticles. To overcome this problem an alternative method was employed. With ICPMS after ultracentrifugation, quantification of the ionic and particulate -fraction of metallic ENP dispersions was possible. Validation of the set-up was performed with certified Au-NP reference materials obtained from NIST and results showed excellent agreement. Combining both of these new methods provided a comprehensive understanding of the metallic ENP systems. After method development with well defined nanoparticle systems, the set-up was tested for its applicability to "real world" samples. First, the characterization of commercial available, polydisperse, commercial Ag nanoparticle (Ag-NP) products was performed. The new developed set-up allowed obtaining an element specific, mass, and number size distribution. Furthermore, Ag-NP concentrations as well as the toxicological relevant ionic Ag concentration could be determined. Figures of merit compared to transmission electron microscopy (TEM) and batch-DLS results were in good agreement and the new method was advantageous in terms of analysis time and reliability. Furthermore the possibility to characterize SiO2 coated Au nanoparticles (Au@SiO2) was investigated. These particles are employed for Shell-Isolated Nanoparticle Enhanced Raman Spectroscopy (SHINERS). It was shown that in terms of particle size characterization, the method provided excellent data. However, determination of Si concentrations to calculate the shell thickness of such particles was impeded by the poor A4F peak shape as well as the high Si background in ICPMS. In conclusion, both setups show great potential for the characterization of metallic ENP either in aerosols or dispersions, respectively.

Characterization of Silver Nanoparticle Products Using Asymmetric Flow Field Flow Fractionation with a Multidetector Approach - a Comparison to Transmission Electron Microscopy and Batch Dynamic Light Scattering

Due to the already prevalent and increasing use of silver-nanoparticle (Ag-NP) products and the raised concerns in particular for the aquatic environment, analytical techniques for the characterization of such products are of need. However, because Ag-NP products are of different compositions and polydispersities, analysis especially of the size distribution is challenging. In this work, an asymmetric flow field flow fractionation (A4F) multidetector system (UV/vis, light scattering, inductively coupled plasma mass spectrometry - ICPMS), in combination with a method to distinguish and quantify the particle and dissolved Ag fractions (ICPMS after ultracentrifugation), for the characterization of Ag-NP products with different degrees of polydispersities is presented. For validation and to outline benefits and limitations, results obtained from batch dynamic light scattering (batch-DLS) and transmission electron microscopy (TEM) were compared. With the developed method a comprehensive understanding in terms of dissolved Ag and Ag-NP concentration as well as an element selective, mass- and number particle size distribution (PSD) was obtained. In relation to batch-DLS, the reliability of the data was improved significantly. In comparison to TEM, faster measurement times and the ability to determine the samples directly in dispersions are clearly advantageous. The proposed setup shows potential for a rapid- and reliable characterization method of virtually any polydisperse metallic NP dispersion, many of them available on the market already.

Finite element modeling and experimental study of brittle fracture in tempered martensitic steels for thermonuclear fusion applications

In this work we have studied brittle fracture in high-chromium reduced activation tempered martensitic steels foreseen as structural materials for thermonuclear fusion reactors. Developing the adequate materials that can withstand the severe irradiation conditions of the burning plasma in a future fusion reactor is one of the major challenges to be solved in order to make profit from the great advantages of thermonuclear fusion as an energy source. High-chromium tempered martensitic steels such as F82H and the most advanced version Eurofer97 are among the main candidate materials for structural applications in future fusion power plants due to low irradiation-induced swelling, good mechanical and thermal properties, and reasonably fast radioactive decay. The most concerning drawback of these kind of steels is irradiation embrittlement, which is manifested by a ductile-to-brittle transition temperature shift to higher temperatures after irradiation whose amplitude depends on the irradiation conditions (temperature, neutron flux, neutron fluence, etc). The aim of this work was to study and model brittle fracture in the ductile-brittle transition region of this kind of steels in the as-received unirradiated conditions. It is necessary to be able to transfer laboratory specimen fracture data to real components and structures in order to assess the performance of these steels in the different operating and transient conditions they could find during the operation life of a fusion reactor. In order to do so, the specimen geometry effects and specimen size effects on measured fracture toughness need to be properly understood, taken into account and predicted with an appropriate model. In particular, specimen size effect on measured toughness is a major concern for the nuclear materials research community owing to the limited irradiation volume in current and planed materials irradiation facilities. The main results of this PhD work are summarized below. The microstructure of Eurofer97 and F82H has been characterized and compared by means of optical microscopy, scanning electron microscopy, transmission electron microscopy and energy-dispersive X-ray spectroscopy in order to identify microstructural features that could play a role in the measured fracture toughness. Both steels have similar but slightly different chemical composition and final heat-treatments but the prior austenitic grain size measured in F82H is approximately 8 times larger than in Eurofer97. It was shown that the alloying element Tantalum, added to stabilize the austenite grain size, played a different role in both steels. After a careful analysis of the particles present in both steels, it was found that Tantalum in Eurofer97 formed carbides of an average size around 100 nanometers. In contrast in F82H it did not form small carbides but formed big oxide inclusions with a size up to 30 µm. These large particles do not effectively pin the grain boundaries. The different behavior of Tantalum in these steels is believed to be mainly a consequence of the larger content of Oxygen present and the smaller amount of Aluminum in F82H compared to Eurofer97. The Master-Curve ASTM-E1921 standard is a method initially developed to determine the ductile-to-brittle transition reference temperature, T0, in fission reactor ferritic steels from a small number of experiments. In this work the applicability of the Master-Curve method to reduced activation tempered martensitic steels such as Eurofer97 and F82H was studied in detail. Fracture tests with pre-cracked sub-sized compact tension specimens (three different sizes, 0.18T, 0.35T and 0.87T of Eurofer97) were carried out in the temperature range [-196 °C, -40 °C]. The toughness-temperature behavior and scatter were shown to deviate from the ASTM-E1921 standard predictions near the lower shelf. Using the method of maximum likelihood, the athermal component of the Master-Curve was calculated to better fit our fracture toughness data from the lower to the middle transition region. We showed that these Master-Curve adjustments are necessary to make the T0 values obtained near the lower shelf with 0.35T size C(T) specimens consistent with those obtained in the middle transition region with 0.87T C(T) specimens. The ASTM-E1921 specimen size limitations, setting the maximum toughness measurable with a given specimen size, were found to be too lenient for this kind of steels. This problem was especially evident in fracture toughness data of Eurofer97 and F82H obtained in the upper transition temperature range with two different specimen sizes. Thus a more stringent specimen size requirement was proposed to avoid inconsistent transition temperature determinations. A promising local fracture model with the potential of predicting cleavage fracture toughness was studied. Finite element simulations were undertaken for compact specimens, notched specimens and tensile specimens of Eurofer97 steel tested from 20 °C down to -197 °C. Three and two dimensional as well as axisymmetric simulations were run in order to calculate the stress and strain fields at the onset of brittle fracture. For each tested temperature, the calculated load-displacement curves were found to reproduce very well those of the experiments. A local approach fracture criterion was studied. This criterion states that when the maximum principal stress is larger than a critical stress within a critical volume ahead of the crack tip, notch or neck, cleavage is triggered leading to macroscopic fracture of the specimen. It was shown that this model is able to predict the minimum fracture load of the notched specimens with the same values of critical stress and critical volume that were calibrated to predict the lower bound of fracture in compact specimens. This local approach model was also successfully used to predict the strong size effect observed experimentally in pre-cracked compact tension specimens in the upper transition region. The critical fracture stress determined in the standard tensile specimens was found higher than that of the fracture specimens. It was suggested that this difference stems from a significant difference in the stress state between the different specimens (triaxiality). Finally, the comparison between the overall fracture behavior in the transition between F82H and Eurofer97 steels indicated that these two materials are quite similar. A difference in the reference temperature T0 of about 20 °C was found, with a nominal value of -100 °C and -80 °C for F82H and Eurofer97 respectively. However, some excessive scatter in the toughness data was found in F82H with more data points than expected lying below the 1% tolerance bound; this was not observed in Eurofer97. From an engineering point of view, the entire fracture databases of these two steels are encompassed practically by the same lower tolerance bound.

Programmable chiral nanocolloids

Nanoparticles promise a variety of application in energy, medicine, and biology. However, most nanoparticlesâ material composition and shape cannot be tuned and so functions have thus far been limited. Moreover they are often also chemically unstable in solution. Therefore, the overall goal of this thesis is to develop nanoparticles whose function and shape can be programmed and that can be corrosion protected, and to then apply these nanoparticles to sensing tasks in complex biological fluids. A special focus of this thesis is chiral nanostructures. A promising technique to address this is physical vapour shadow growth, namely nano glancing angle depo-sition (nanoGLAD). This scheme allows the design of new three-dimensional (3D) hybrid nanoparticles as it permits the control over both the shape and material composition of the nanoparticles. Although this method offers the possibility to grow nanoparticles that are functionally programmed, it has so far not al-lowed the use of many materials that are chemically unstable in solution, which limits the scope of poten-tial applications. This thesis starts with describing the nanoGLAD growth procedure. A first application is the wafer-scale patterning of unconventional nanoshapes, e.g. tri-layer particles, holes, rings, and hollow domes, which are not possible using state of the art 3D nanofabrication methods (chapter 2). Then, in conjunction with atom-ic layer deposition (ALD), this thesis shows how the nanoGLAD scheme can be adapted for the fabrication of â3D core-shell nanoparticles and nanocolloidsâ using unstable and reactive materials. The key concept here is that the core consists of the unstable material which is grown such that the shell contains no voids or defects (chapter 3). Notably, the shapes that can be grown include symmetry-broken chiral nanoparti-cles, which possess unique spectral properties that make them useful for sensing applications. This thesis uses chiral nanocolloids to realise extremely sensitive plasmonic nanosensors and nanocolloids that can also be used as a nanomechanical probes for active nanorheology. By forming a composite of two materials during growth â one that gives a strong plasmonic response and the other to tune the compositeâs dielec-tric function â plasmonic nanoparticles are presented that show record local surface plasmon resonance (LSPR) sensitivities to date (chapter 4). With the same alloying principle a plasmonic and a ferromagnetic material are combined. The resulting âchiral + plasmonic + ferromagneticâ particles can be actuated using a magnetic field and this is used to measure the viscosity of blood plasma in the presence of blood cells. The viscosity of blood serum is an important disease indicator, but the measurement using commercial rheome-ters requires the separation of the blood cells. This is not needed in the nanorheological measurements shown in this thesis (chapter 5).
