Development and Characterization of a Microfluidic Chip to Capture Circulating Prostate Cancer Cells

The analysis of circulating tumor cells (CTCs) from the blood of cancer patients promises a better understanding of metastasis formation and tumor phenotypes. CTCs are rare (there is 1 CTC in 1 billion of circulating blood cells) and therefore challenging to isolate. Recently developed highly sensitive microfluidic platforms made a leap forward in trapping these cells efficiently. This work focused on the development of a microfluidic platform that specifically captures prostate cancer cells from a whole blood sample. The versatility of the system was exploited by tethering different capturing antibodies on the microchip surface. Circulating prostate tumor cells were successfully recognized with the microfluidic chip. Furthermore, the platform was enhanced to carry out proliferation assays of captured cells directly on the chip, using hydrogel as a three-dimensional cell culture matrix. Although further improvements regarding the on-chip proliferation assay need to be done, the potential benefit of a functional readout promises new insights in cancer progression that can be translated to advanced cancer staging or prognostic tools.

Toward Microfluidic Label-Free Isolation and Enumeration of Circulating Tumor Cells from Blood Samples

The isolation, analysis, and enumeration of circulating tumor cells (CTCs) from cancer patient blood samples are a paradigm shift for cancer patient diagnosis, prognosis, and treatment monitoring. Most methods used to isolate and enumerate these target cells rely on the expression of cell surface markers, which varies between patients, cancer types, tumors, and stages. Here, we propose a label-free high-throughput platform to isolate, enumerate, and size CTCs on two coupled microfluidic devices. Cancer cells were purified through a Vortex chip and subsequently flowed in-line to an impedance chip, where a pair of electrodes measured fluctuations of an applied electric field generated by cells passing through. A proof-of-concept of the coupling of those two devices was demonstrated with beads and cells. First, the impedance chip was tested as a stand-alone device: (1) with beads (mean counting error of 1.0%, sizing information clearly separated three clusters for 8, 15, and 20 um beads, respectively) as well as (2) with cancer cells (mean counting error of 3.5%). Second, the combined setup was tested with beads, then with cells in phosphate-buffered saline, and finally with cancer cells spiked in healthy blood. Experiments demonstrated that the Vortex HT chip enriched the cancer cells, which then could be counted and differentiated from smaller blood cells by the impedance chip based on size information. Further discrimination was shown with dual high-frequency measurements using electric opacity, highlighting the potential application of this combined setup for a fully integrated label-free isolation and enumeration of CTCs from cancer patient samples. In vitro fate mapping of stem cell development at the single cell level

Hematopoietic stem cells (HSCs) are responsible for the continuous production of all blood cells. This unique ability has made it possible to successfully use HSCs in the clinical setting to remedy various blood disorders. However, despite six decades of research, the full potential of HSCs to treat patients with hematological malignancies is still restricted by the low number of functional stem cells that can be isolated from different sources. In vitro HSC expansion has been widely considered a potential solution but has proven to be extremely difficult due to the rapid loss of functional potential of cells in the culture outside of their natural microenviron-mental niche. In order to achieve robust HSC expansion in vitro, it is crucial to better under-stand the mechanisms that regulate HSC fate choices. Moreover, the absence of reliable mark-ers for identifying functional HSCs on a culture dish necessitates the use of expensive and time-consuming in vivo assays. The discovery of predictive phenotypic markers for HSC potential could address this problem. Many platforms for analyzing HSC fate at the single-cell level have been reported, but they generally suffer from poor cell viability, limited throughput, unreliable phenotyping by immunocytochemistry, and the difficulty of performing long-term cell cultures. Additionally, these platforms are usually fabricated from polydimethylsiloxane (PDMS), a poly-mer known to have several disadvantages for cell-based applications. This thesis addresses these shortcomings through the development of a robust microchip sys-tem for the high-resolution, live-cell analysis of hundreds to thousands of single stem cells and their daughter cells. The platform combines key advantages of microfluidic technology (for cell lineage analysis) and hydrogel microwell array technology (for gentle cell capture and long-term culture). The platform is fabricated from two different layers of hydrogel to create a closed ar-ray of grooves, where cellsâ movements are restricted and grow along one axis. This design en-ables an unambiguous tracking of a single cell and all of its progeny over several generations. These two layers are produced from different biocompatible hydrogels: poly (2-hydroxyethyl methylacrylate) / trimethylolpropane trimethacrylate (pHEMA-TMPTMA), a copolymer acting as a topographically structured cellular substrate that can be closed with a polyethylene glycol (PEG) hydrogel lid. These two parts are combined together and covalently bonded to each oth-er after the seeding of live cells into the grooves of the bottom part. To assess the potential of the platform for single stem-cell analyses, HSCs were cultured and their behavior (including cell fate, proliferation kinetics, and functionality) were thoroughly characterized. To analyze imaging data obtained from single-cell time-lapse experiments performed on the platform, an algorithm was developed to automatically identify and track single HSCs and their daughter cells, efficiently extracting various parameters, such as single-cell proliferation kinet-ics, cell fate choices, and lineage relationships. Mitochondria have recently been identified as key modulators of HSC function; however, their specific role in fate regulation has remained obscure. Therefore, the single-cell analysis plat-form was applied to measure the distribution of mitochondrial mass in dividing HSCs and their progeny to assess a potential correlation with HSC fate choices. Expression of E-Cadherin in Epithelial Cancer Cells Increases Cell Motility and Directionality through the Localization of ZO-1 during Collective Cell Migration

Collective cell migration of epithelial tumor cells is one of the important factors for elucidating cancer metastasis and developing novel drugs for cancer treatment. Especially, new roles of E-cadherin in cancer migration and metastasis, beyond the epithelial-mesenchymal transition, have recently been unveiled. Here, we quantitatively examined cell motility using micropatterned free edge migration model with E-cadherin re-expressing EC96 cells derived from adenocarcinoma gastric (AGS) cell line. EC96 cells showed increased migration features such as the expansion of cell islands and straightforward movement compared to AGS cells. The function of tight junction proteins known to E-cadherin expression were evaluated for cell migration by knockdown using sh-RNA. Cell migration and straight movement of EC96 cells were reduced by knockdown of ZO-1 and claudin-7, to a lesser degree. Analysis of the migratory activity of boundary cells and inner cells shows that EC96 cell migration was primarily conducted by boundary cells, similar to leader cells in collective migration. Immunofluorescence analysis showed that tight junctions (TJs) of EC96 cells might play important roles in intracellular communication among boundary cells. ZO-1 is localized to the base of protruding lamellipodia and cell contact sites at the rear of cells, indicating that ZO-1 might be important for the interaction between traction and tensile forces. Overall, dynamic regulation of E-cadherin expression and localization by interaction with ZO-1 protein is one of the targets for elucidating the mechanism of collective migration of cancer metastasis.
