Bioactive Lipids Lysophosphatidic Acid and Sphingosine 1-Phosphate Mediate Breast Cancer Cell Biological Functions Through Distinct Mechanisms

Lysophosphatidic acid (LPA) and sphingosine 1-phosphate (S1P) are structurally related bioactive lipids with growth factor-like activities. LPA and SIP are naturally produced in vivo by blood platelets upon platelet aggregation and at least in vitro by fibroblasts, adipocytes, and multiple types of tumor cells. Breast cancer cells respond to LPA and S1P. However, their specific actions on breast cancer cell biological functions remain unclear. We therefore conducted an in vitro side-by-side study of these two lipids on breast cancer cells. LPA mediates human breast cancer MDA-BO2 cell proliferation, migration, and invasion through activation of a G JERK1/2-dependent signaling pathway, whereas activation of G(alpha i)/PI3K predominates upon S I P stimulation. In MDA-BO2 cells, LPA but not S I P activities were dependent on active type I insulin-like growth factor and epithelial growth factor receptors. LPA and SIP act directly on endothelial cells to induce angiogenesis. We demonstrate that LPA and SIP have indirect angiogenic properties as judged by induced secretion of angiogenic factors by breast cancer cells primed with these lysophospholipids. SIP, but not LPA, controlled the expression of VEGF-A by breast cancer cells, while LPA, but not S1P, controlled the expression of GM-CSF, Gro-alpha, MCP-1, and IL-6. According to the secretion of these paracrine osteoclastic factors, LPA, but not S1P, stimulates breast cancer cell-induced osteoclastogenesis. These findings suggest that, in vivo, LPA and SIP can coordinate their action on tumor and surrounding cells to induce breast cancer progression both at primary and bone metastatic sites.

The interplay between the VEGF-C/VEGFR-3 axis and CCL21/CCR7 axis in tumor cell invasion

There are two main routes for tumor spread and metastasis, the blood vasculature and the lymphatic system. A wide range of human carcinomas, including lung, colon and breast cancer, spread to distant sites via the lymphatic system. Preclinical and clinical studies have demonstrated a positive correlation between the incidence of lymph node metastasis and the secretion of vascular endothelial growth factor-C (VEGF-C) by tumor cells. Indeed, VEGF-C is critical for the formation of new lymphatic vessels in a process called lymphangiogenesis, suggesting this mechanism as the main "escape route" for tumor cells. However, many studies have failed to identify functional lymphatic vessels inside tumors, and much research has demonstrated that metastasis can occur in the absence of lymphangiogenesis. Furthermore, there is growing evidence that many tumor cells may express vascular endothelial growth factor receptor-3 (VEGFR-3), the primary receptor for VEGF-C, suggesting that autologous VEGFR-3 signaling loops may enhance the invasive and metastatic potential of tumor cells. Other than lymphangiogenic factors, the lymphatic homing chemokine receptor CCR7 is strongly correlated to lymph node metastasis, suggesting that tumor cells mimic dendritic cells in sensing and homing to lymphatic vessels and lymph nodes. Potential mechanisms underlying these observations have not been elucidated. Using several in vitro 3D models, we examined the interactions between tumor and lymphatic endothelial cells (LECs) to gain insight into the mechanistic roles of VEGF-C and CCR7-mediated tumor cell invasion towards lymphatics as well as elucidate their potential interplay. All the designed experimental setups utilized a 3D matrix for a more physiologically relevant chemokine and growth factor signaling environment. For example, we designed one culture model where tumor cells are seeded in one gel compartment and lymphatic endothelial cells in a horizontally adjacent one, allowing both cell types to establish contact through soluble mediators without being initially mixed while simultaneously allowing direct visualization of their interactions. We first demonstrated that receptor expression patterns by cells are strongly influenced by the extracellular matrix (ECM). Indeed, while we failed to detect tumor expression of VEGFR-3 in 2D conditions, we found both expression and phosphorylation of VEGFR-3 in three of four tumor cell lines tested. Furthermore, in the three VEGFR-3 positive cell lines, invasive capacity and chemoattraction towards lymphatics was demonstrated to be dependent on both VEGF-C secretion and CCR7 expression by the tumor cells. We found that VEGF-C could act in both an autocrine manner, by enhancing the migratory and invasive capacity of tumor cells, and in a paracrine manner where it enhances CCL21 secretion by LECs to promote the chemoinvasion of tumor cells via CCL21 secretion. The same mechanism was also tested in vivo by injecting VEGF-C into mouse skin: VEGF-C, but not saline nor VEGF-A, caused marked upregulation of CCL21 associated with lymphatic vessels. In this way, VEGF-C and CCR7 act synergistically to promote the invasive phenotype of tumor cells. Additionally, we uncovered a new and critical role for VEGFR-3 on mammary epithelial cells. We demonstrated that the receptor is involved in epithelial tubulogenesis in a collagen matrix, and furthermore, it helps to preserve the integrity of a mammary cell layer (since blocking the receptor leads to increased epithelial permeability). In conclusion, this work highlights the importance of the VEGFR-3/VEGF-C and CCR7/CCL21 axis in promoting tumor cell migration and invasion. It also shows that VEGFR-3 may play many roles in the process of tumorigenesis and metastasis.

Radiation-induced modifications of the tumor microenvironment promote metastasis

Radiotherapy is successfully used to treat cancer. Emerging evidence, however, indicates that recurrences after radiotherapy are associated with increased local invasion, metastatic spreading and poor prognosis. Radiation-induced modifications of the tumor microenvironment have been proposed to contribute to increased aggressive tumor behavior, an effect also referred to as tumor bed effect, but the putative mechanisms involved have remained largely elusive. We have recently demonstrated that irradiation of the prospective tumor stroma impairs de novo angiogenesis through sustained inhibition of proliferation, migration and sprouting of endothelial cells. Experimental tumors growing within a pre-irradiated field have reduced tumor angiogenesis and tumor growth, increased hypoxia, necrosis, local invasion and distant metastasis. Mechanisms of progression involve adaptation of tumor cells to local hypoxic conditions as well as selection of cells with invasive and metastatic capacities. The matricellular protein CYR61 and integrin alpha V beta 5 emerged as molecules that cooperate to mediate lung metastasis. Cilengitide, a small molecular inhibitor of alpha V integrins prevented lung metastasis formation. These results represent a conceptual advance to the understanding of the tumor bed effect and indicate that alpha V integrin inhibition might be a potential therapeutic approach for preventing metastasis in patients at risk for post-radiation recurrences.

Low Doses of Ionizing Radiation Promote Tumor Growth and Metastasis by Enhancing Angiogenesis

Radiotherapy is a widely used treatment option in cancer. However, recent evidence suggests that doses of ionizing radiation (IR) delivered inside the tumor target volume, during fractionated radiotherapy, can promote tumor invasion and metastasis. Furthermore, the tissues that surround the tumor area are also exposed to low doses of IR that are lower than those delivered inside the tumor mass, because external radiotherapy is delivered to the tumor through multiple radiation beams, in order to prevent damage of organs at risk. The biological effects of these low doses of IR on the healthy tissue surrounding the tumor area, and in particular on the vasculature remain largely to be determined. We found that doses of IR lower or equal to 0.8 Gy enhance endothelial cell migration without impinging on cell proliferation or survival. Moreover, we show that low-dose IR induces a rapid phosphorylation of several endothelial cell proteins, including the Vascular Endothelial Growth Factor (VEGF) Receptor-2 and induces VEGF production in hypoxia mimicking conditions. By activating the VEGF Receptor-2, low-dose IR enhances endothelial cell migration and prevents endothelial cell death promoted by an anti-angiogenic drug, bevacizumab. In addition, we observed that low-dose IR accelerates embryonic angiogenic sprouting during zebrafish development and promotes adult angiogenesis during zebrafish fin regeneration and in the murine Matrigel assay. Using murine experimental models of leukemia and orthotopic breast cancer, we show that low-dose IR promotes tumor growth and metastasis and that these effects were prevented by the administration of a VEGF receptor-tyrosine kinase inhibitor immediately before IR exposure. These findings demonstrate a new mechanism to the understanding of the potential pro-metastatic effect of IR and may provide a new rationale basis to the improvement of current radiotherapy protocols.
