Postpolymerization Modification of Poly(glycidyl methacrylate) Brushes: An XPS Depth-Profiling Study

Surface-initiated polymerization represents a versatile strategy to modify a diverse range of materials with thin, functional polymer coatings. While in many cases the desired functional groups can be directly incorporated via (co)polymerization of the appropriate monomer(s), other functional groups are incompatible with the polymerization strategies that are commonly used to grow polymer brushes and can only be introduced by postpolymerization modification. Determining the local concentration and spatial distribution of these functional groups in postmodified brushes is a challenging task but could help to optimize the design and properties of these polymer coatings. This article reports on the use of X-ray photoelectron spectroscopy (XPS) combined with C-60 cluster ion sputtering to address this challenge. Poly(glycidyl methacrylate) (PGMA) brushes prepared via surface-initiated atom transfer radical polymerization (SI-ATRP) were used as a model platform and were postmodified with propylamine and bovine serum albumin (BSA). The XPS depth-profiling experiments showed that the small propylamine molecules were essentially homogeneously distributed throughout the brush, with the exception of the top few nanometers, which were enriched in propylamine moieties. It was also demonstrated that the amount of propylamine introduced within the polymer brush increased with increasing the postpolymerization reaction time, while no concentration gradients could be observed, indicative of a fast diffusion of the propylamine through the polymer brush layer. On the other hand, XPS depth-profiling experiments performed on polymer brushes that were postmodified with BSA revealed that this protein was only localized in the topmost layers of the polymer coating, which reflects the steric hindrance by the dense polymer brush that prevents efficient diffusion of these large molecules. Together, the results of these experiments demonstrate that XPS depth-profiling combined with C-60 cluster ion sputtering is an efficient and powerful means to study the distribution of functionalities incorporated within a polymer brush layer by postpolymerization modification reactions.

Polymer Brushes as Substrates for Microarray Applications

The development of innovative bioanalytical technologies that enable the discovery of new therapeutics or the creation of systems for rapid, accurate and parallel analyses has gained an extraordinary interest in the biomedical sector in the last 10-15 years. Since proteins are the key-targets for such essential discoveries and developments, a particular emphasis has been placed on proteomics (study of proteins), including the investigation of protein structures, changes of protein expression upon treatment of cellular systems, the role of post-translational protein modifications on cell signaling events and the validation of selected proteins as disease-specific markers or pharmaceutical targets. These studies usually aim at using crude biological samples as the starting materials, which typically contain a large variety of proteins at concentrations covering a wide dynamic range and where the low abundant proteins are often the biologically most relevant. Therefore, the challenge is to create analytical tools which are able to cope with this large protein concentration range and which provide sufficiently high sensitivity to measure low abundant analytes in the presence of a high abundant protein matrix. The information that is expected from such technologies may soon exert a drastic change on the pace of medical research and considerably impact on the care of patients. As protein microarrays offer a powerful tool for the rapid, parallel analysis of complex samples and only require a minimal amount of reagents, they are currently one of the hot topics of life sciences, which strive to provide such analytical solutions. One of the current limiting factors in the field of protein microarrays for achieving the required analytical sensitivity is the amount of proteins that can be immobilized on the proteomic chip surface. Polymer brushes, which consist of an arrangement of polymer chains that are attached by one end to a substrate in a stretched, well-defined chain conformation, can increase the surface area available for protein binding, and thus act as a new category of 3D protein microarray substrates. With the advent of surface-initiated controlled polymerization techniques, extremely precise control over the thickness, composition, architecture and grafting density can be achieved, which allow to finely tune the properties of the polymer brush. In addition, these polymer chains may contain, natively or after specific post-polymerization reaction of their side chains, a high density of functional groups that can react with proteins. The different approaches available for the preparation of such surface-tethered polymer chains, the strategies allowing control over their architecture and properties as well as examples of applications related to protein binding are presented in Chapter 1. This Thesis describes the use of polymer brushes to create a new type of 3D proteomic chips, with the ultimate goal to improve the protein binding capacity compared to existing chip surface, without altering the accessibility of the detection reagents or increasing the intrinsic fluorescence of the background. The objective of this research work is to prepare polymer brush-based substrates that can outperform the sensitivity (i.e. increase the signal-to-noise ratio) that is obtained with a commercial microarray system, which is used as a benchmark in this Thesis. In this regard, functional polymer brushes containing epoxide groups that can react with the nucleophilic moieties of proteins have been chosen as promising candidates. In a preliminary set of experiments, the potential of glycidyl methacrylate-containing brushes to bind (bio)molecules via a post-polymerization modification reaction under mild conditions, i.e. at room temperature in aqueous medium, has been studied (Chapter 2). To this end, poly(glycidyl methacrylate) brushes, as well as several poly(glycidyl methacrylate)-co-poly(2-(diethylamino)ethyl methacrylate) copolymer brushes of different composition have been prepared by surface-initiated atom transfer radical (co)polymerization. The kinetics of the post-polymerization modification reaction of these (co)polymer brushes were followed using various model primary amines, as well as proteins. The introduction of tertiary amine groups, incorporated in the polymer brushes such as 2-(diethylamino)ethyl methacrylate units, was demonstrated not only to enhance the rate of the epoxide ring-opening reaction but also to induce higher protein binding capacities as compared to poly(glycidyl methacrylate) homopolymer brushes. The improved loading capacities of the (co)polymer brushes were evaluated in Chapter 3 under conditions that more closely resemble real microarray applications, such as the use of automated nanospotters to deposit proteins and fluorescence readers to detect protein immobilization. In this regard, commercially-available proteomic chips were coated with the epoxide-containing (co)polymer brushes and the spotting conditions as well as the proteins were varied. These studies revealed that the loading capacity depends on the pH and on the investigated protein, but also that the (co)polymer brush-coated substrates show far higher loading capacities than the standard, unmodified proteomic chips. This investigation also ascertained the supremacy of brushes containing 2-(diethylamino)ethyl methacrylate units compared to homopolymer brushes, as well as the superiority of thick (co)polymer brushes in comparison to their thinner equivalents. These (co)polymer brushes were subsequently used as platforms for real, although simplified, protein assays. The modified proteomic chips were spotted with unlabeled proteins that were further recognized with detection antibodies via either a one-step or a two-step approach. The clear benefits observed for copolymer brushes as compared to the existing technology under these realistic assay conditions nicely support the fact that the interior volume of the brush remains accessible for the detection reagents. In conclusion, the combination of the high loading capacity and the remaining accessibility for the detection partners make these (co)polymer brushes, and particularly the poly(glycidyl methacrylate)-co-poly(2-(diethylamino)ethyl methacrylate) brushes, excellent candidates for the improvement of the sensitivity in protein microarray applications.

Interactive Surfaces Based on Polymer Brushes

While the bulk structure of a material determines its mechanical properties, its surface plays a key role when interactions with the environment are important. Nowadays materials in bio- and nanotechnology are required to possess very complex functionalities and to interact specifically to the stimuli they receive from their surroundings. Thus, their surface modifications are becoming very demanding. In recent years surface initiated controlled radical polymerization techniques have emerged as very versatile strategies that provide a powerful toolbox to tackle the challenges that are set to material scientists. The attractiveness of these methods lies in the possibility to create well defined surface arrangements of polymer chains, which we refer to as polymer brushes, with precise control over thickness, composition and architecture. In addition, these techniques can be applied to very challenging substrates with respect to the chemical composition and geometry, which also includes porous materials. This Thesis, therefore, exploits the possibilities that surface-initiated controlled radical polymerization techniques offer to finely tune interfacial properties of materials and render them capable to interact with chemical and biological species in a desired way. Its paramount aim is to demonstrate how the functionalities and geometry of a polymer brush matrix can be employed to influence inorganic film deposition using synthetic, biomimetic and purely biological mechanism or enhance polymer brushed stability in aqueous media. This Thesis is divided into five distinct chapters, which will be briefly summarized in following paragraphs. Chapter 1 describes the preparation of polymer brushes and strategies that are used to control their chemical composition and architecture. Additionally, it will give an overview on the use of brushes as cell adhesive surfaces as well as templates for the production of nanoparticles and thin inorganic films. Chapter 2 explores a new strategy to guide the chemical solution deposition of thin, microstructured metal films. The proposed strategy is based on the use of poly(2-(methacryloyloxy)ethyl ammonium chloride) (PMETAC) brushes grown via surface-initiated atom transfer radical polymerization as a template. Thin films are prepared by first loading the PMETAC brushes with an appropriate precursor, followed by its reduction by NaBH4 and finally an oxygen plasma treatment to remove the stabilizing polymer brush matrix and generate the desired thin metallic film. Of particular interest is the question whether the thickness and lateral dimensions of the deposited metallic films can be controlled by micropatterning the polymer brush template and varying the brush thickness. In addition, this concept will be extended to the preparation of bimetallic (gold/palladium) films using a single polymer brush matrix. A similar strategy is explored in Chapter 3 to control the deposition of thin calcium carbonate films using a biomimetic process. The process is characterized by the stabilization of amorphous calcium carbonate, the geometry and localication of which can be controlled by the poly(methacrylic acid) brush template. A special focus of this Chapter is to examine the influence of the polymer brush thickness on the mineral deposition and to determine to which extent is the mineral phase is integrated into the polymer brush film. The aim of Chapter 4 is to develop a general approach for the modification of implantable materials with a polymer brush based coating that promotes bone formation. The proposed coatings are prepared via surface-initiated copolymerization of 2-hydoxyethyl methacrylate and 2-(methacryloyl)ethyl phosphate, which results in a thin hydrogel layer that mimics the negatively charged matrix macromolecules involved in natural bone formation. To further emulate the properties of the natural bone extracellular matrix, the coatings will be functionalized with the cell adhesive GGGRGDS peptide. The ability of the proposed coatings to induce bone formation is investigated in in vitro experiments on MC3T3-E1 pre-osteoblasts and evaluated using an ALP and alizarin red assays. Finally, Chapter 5 focuses on studying the parameters that could prevent the hydrolytic cleavage of Si-O bond, which tethers the polymer brushes grafted from SiOx in aqueous media. In the first part of the study, the length of the organosilane initiator's alkyl spacer will be varied, in order to evaluate to which extent it can insulate the site that anchors the hydrophilic polymer brush to the substrate. In the second part, a short, hydrophobic block will be introduced between the silicon oxide substrate and the hydrophilic polymer in order to more efficiently protect the underlying Si-O bond from the surrounding aqueous media and prevent the chain detachment.

Polymer Brush Gradients Grafted from Plasma-Polymerized Surfaces

A new method for generating a surface density gradient of polymer chains is presented. A substrate-independent polymer deposition technique was used to coat materials with a chemical gradient based on plasma copolymerization of 1,7-octadiene and allylamine. This provided a uniform chemical gradient to which initiators for atom transfer radical polymerization (ATRP) were immobilized. After surface-initiated atom transfer radical polymerization (SI-ATRP), poly(2-hydroxyethyl methacrylate) (PHEMA) chains were grafted from the surface and the measured thickness profiles provided direct evidence for how surface crowding provides an entropic driving force resulting in chain extension away from the surface. Film thicknesses were found to increase with the position along the gradient surface, reflecting the gradual transition from collapsed to more extended surface-tethered polymer chains as the grafting density increased. The method described is novel in that the approach provides covalent linkages from the polymer coating to the substrate and is not limited to a particular surface chemistry of the starting material.
