Abstract

For many decades, ventilated cavities in wall assemblies of buildings have been essential for creating moisture-resilient constructions by allowing airflow within the air gap to promote drying. In addition to that, the airflow in the cavity can contribute to the overall performance of the wall structure. Similar to the wall assemblies with traditional claddings, photovoltaic (PV) panels in the Building-integrated photovoltaic (BIPV) façades can be separated from the building envelope by an air gap that is created for ventilation purposes. In this study, the impact of the thermo-hydrodynamic behavior of the airflow in the naturally ventilated cavity behind traditional (passive) and BIPV (active) façades on the performance of the entire wall assembly is numerically and experimentally investigated. Due to the complexity of the problem caused by the contribution of several parameters, as a first step, a comprehensive literature review is performed to specify the effective factors on the air change rate in the ventilated cavity behind common wall assemblies. As the second step, the steady-state analysis is carried out within the framework of a research project ASHRAE 1759-RP funded by the American Society of Heating, Refrigerating and Air-Conditioning Engineers (ASHRAE), in which the plausible definitions of the thermal resistance of a vertical ventilated air-space behind external cladding systems are defined using theoretical and applied formulations. Moreover, based on the uncertainty analysis of measurements, the modifications of the relevant standardized method (ASTM C1363-19) to account for the airflow effects on the thermal performance of the wall assembly are proposed. In the third step, the problem is further explored in the transient conditions by developing a 2D numerical in-house code validated against experimental measurements. Accordingly, the performance of wall structures with different thermal inertia having passive (traditional) and active (BIPV) external claddings are numerically investigated. In the last step of this thesis, the problem is experimentally discovered, and the thermo-hydrodynamic performance of ventilated wall structures of the test facility is investigated using data collected during a full-year measuring campaign. The results of this study show that the thermo-hydrodynamic behavior of the airflow behind external claddings is a complex phenomenon. The results of the steady-state analysis reveal that, under some conditions, the air-space could be able to resist the heat flux passing the wall assembly even more than the external cladding. Moreover, the uncertainty analysis sheds light on the fact that accurate instruments are needed to reduce the uncertainty of measuring the thermal resistance of the cavity. The results of the transient numerical analysis show that the overall performance of a ventilated wall structure dynamically changes depending on the diurnal variation of the outdoor/indoor conditions. Moreover, it is shown that replacing the passive cladding with an active façade could strongly affect the hydrodynamic and thermal performances of the entire wall assembly. The results of the experimental measurements pointed out that the airflow in the ventilated cavity has a major impact on the yearly performance of a wall structure; therefore, further investigations are required in future studies to evaluate the possibility of controlling the air change rate in the cavity using a fan-assisted system.

Uncertainty quantification in unfolding elementary particle spectra at the Large Hadron Collider

This thesis studies statistical inference in the high energy physics unfolding problem, which is an ill-posed inverse problem arising in data analysis at the Large Hadron Collider (LHC) at CERN. Any measurement made at the LHC is smeared by the finite resolution of the particle detectors and the goal in unfolding is to use these smeared measurements to make nonparametric inferences about the underlying particle spectrum. Mathematically the problem consists in inferring the intensity function of an indirectly observed Poisson point process. Rigorous uncertainty quantification of the unfolded spectrum is of central importance to particle physicists. The problem is typically solved by first forming a regularized point estimator in the unfolded space and then using the variability of this estimator to form frequentist confidence intervals. Such confidence intervals, however, underestimate the uncertainty, since they neglect the bias that is used to regularize the problem. We demonstrate that, as a result, conventional statistical techniques as well as the methods that are presently used at the LHC yield confidence intervals which may suffer from severe undercoverage in realistic unfolding scenarios. We propose two complementary ways of addressing this issue. The first approach applies to situations where the unfolded spectrum is expected to be a smooth function and consists in using an iterative bias-correction technique for debiasing the unfolded point estimator obtained using a roughness penalty. We demonstrate that basing the uncertainties on the variability of the bias-corrected point estimator provides significantly improved coverage with only a modest increase in the length of the confidence intervals, even when the amount of bias-correction is chosen in a data-driven way. We compare the iterative bias-correction to an alternative debiasing technique based on undersmoothing and find that, in several situations, bias-correction provides shorter confidence intervals than undersmoothing. The new methodology is applied to unfolding the Z boson invariant mass spectrum measured in the CMS experiment at the LHC. The second approach exploits the fact that a significant portion of LHC particle spectra are known to have a steeply falling shape. A physically justified way of regularizing such spectra is to impose shape constraints in the form of positivity, monotonicity and convexity. Moreover, when the shape constraints are applied to an unfolded confidence set, one can regularize the length of the confidence intervals without sacrificing coverage. More specifically, we form shape-constrained confidence intervals by considering all those spectra that satisfy the shape constraints and fit the smeared data within a given confidence level. This enables us to derive regularized unfolded uncertainties which have by construction guaranteed simultaneous finite-sample coverage, provided that the true spectrum satisfies the shape constraints. The uncertainties are conservative, but still usefully tight. The method is demonstrated using simulations designed to mimic unfolding the inclusive jet transverse momentum spectrum at the LHC.

Most tissue engineering approaches include the delivery of reparative cells to a damaged host tissue. Generally these cells are sought to be of a stem cell character since they retain a high potential for proliferation and differentiation into diverse phenotypes. To restrict cell function to the desired phenotype and to avoid anti-immune responses, autologous stem cells are frequently isolated from intact regions of the same (damaged) organ, such as keratinocytes from the basal layer of the skin and hair follicles or from the same germ layer origin such as mesenchymal stem cells (MSC) that are obtained from the bone marrow. This approach faces two major problems: 1) The number of source cells obtained from biopsies is generally too low for direct implantation, which requires cell multiplication in culture. 2) The standard culture conditions, including growth in plastic dishes, passaging, and the composition of the growth medium, never correspond to the natural microenvironment and influence cell differentiation, often resulting in non-desired phenotypes. The objective of my thesis was to provide a solution for both problems that is applicable for the average cell culture laboratory. Our technique reduces traumatic enzymatic passaging, provides constant cell densities to keep cells in the exponential growth phase and at the same time avoids contact inhibition of cell growth. This novel approach comprises the development of a new elastic culture surface, produced from a high extension silicone rubber (HESR) that we here for the first time applied for cell culture. The HESR is enlarged by a novel mechanical approach permitting to visualize and control cells under expansion. Untreated silicone, the polymer component of the HESR, does not promote cell attachment and must therefore be functionalized. In the first part of my thesis (Chapter 2) we establish a surface treatment for silicone membranes that will resist large surface expansions. We compare established coating techniques that have originally been developed to provide static silicone surfaces with cell adhesive molecules by 1) hydrophobic, 2) electrostatic and 3) covalent interactions. Cell attachment, spreading and proliferation was evaluated between these three treatments as a function of the coating in static and stretched conditions. We show that covalent immobilization of collagen type I is most suitable to stretch cells cultured on elastic silicone substrates. We applied this technique to functionalize the HESR culture surface in all later applications. The second central part of my thesis (Chapter 3) develops a new culture method consisting of a motorized device that isotropically expands the coated HESR surface to yet unmatched 1000% of its initial surface. Proof-of-principle of this method is given by culturing human MSC, which are sensitive to passaging during expansion in vitro and that are therapeutically relevant cells to repair bone, cartilage, and cardiovascular tissue. MSC were cultured three consecutive passages on the culture surface that has been expanded in steps of 5% every 6 h for 20 days, respectively. After nine weeks of expansion culture, MSC exhibited consistently high proliferation rates, fully retained their pluripotent stem cell character and were inducible to differentiate into adipogenic, chondrogenic, osteogenic, and myogenic lineages. The cell surface expansion culture surface produced ∼10-times higher cell numbers compared with nine weeks of standard culture, which required nine events of passaging to attain the same culture surface. We propose our method to produce MSC for clinical cell therapy. In the third part of my thesis (Chapter 4) we implement our novel surface expansion method to culture fibroblasts with the aim to: 1) rapidly expand primary fibroblasts, 2) achieve higher cell yields than obtained with conventional culture, and 3) obtain homogeneous cell populations with non-fibrogenic character. The expansion of primary autologous dermal fibroblasts in culture is a pivotal step to obtain sufficiently high cell numbers for application in skin esthetics and for the treatment of large burn wounds. Dynamically enlarging the culture surface by 800% over 20 d, yielded ∼1.5 higher fibroblasts numbers than the equivalent three standard passages. By further optimizing the surface expansion speed according to the cell growth rates, the same high cell numbers were produced after only 9 d of culture. The highly compliant nature of the HESR surface had the beneficial effect of completely abolishing the formation of 'fibrotic' myofibroblasts. Hence, our novel method for fibroblast culture is faster and yields higher numbers of more homogeneous cells than standard culture passaging. It delivers a fibroblast quality and quantity that is appealing for cell therapeutic purposes. The aim of the last part of my thesis (Chapter 5) was to provide highly compliant silicone elastomers, such as the HESR, with a micropattern of cell adhesive proteins. In particular we aim to produce arrays of extracellular matrix proteins with a size and morphology to promote formation of single focal adhesions on highly elastic and expandable surfaces. This will allow future studies on cell mechanosensing through focal adhesions by discriminating between effects of focal adhesion size changes and stretch. An existing method to pattern rigid surfaces with proteins, so-called microcontact printing, is not suitable to transfer proteins onto surfaces that are softer than the stamp. We here establish a new transfer technique that is based on a stencil with micron-sized openings. The new stencil is used to expose the surface of compliant silicone substrates with Young's modulus of 10-100 kPa to a protein solution of fibronectin, collagen I and protein-repellent polymers, leaving behind the desired protein pattern. Fibroblasts were successfully attached onto the resulting focal islet arrays on compliant substrates.

Distributed Constraint Optimization

Distributed Constraint Satisfaction (DisCSP) and Distributed Constraint Optimization (DCOP) are formal frameworks that can be used to model a variety of problems in which multiple decision-makers cooperate towards a common goal: from computing an equilibrium of a game, to vehicle routing problems, to combinatorial auctions. In this thesis, we independently address two important issues in such multi-agent problems: 1) how to provide strong guarantees on the protection of the privacy of the participants, and 2) how to anticipate future, uncontrollable events. On the privacy front, our contributions depart from previous work in two ways. First, we consider not only constraint privacy (the agents' private costs) and decision privacy (keeping the complete solution secret), but also two other types of privacy that have been largely overlooked in the literature: agent privacy, which has to do with protecting the identities of the participants, and topology privacy, which covers information about the agents' co-dependencies. Second, while previous work focused mainly on quantitatively measuring and reducing privacy loss, our algorithms provide stronger, qualitative guarantees on what information will remain secret. Our experiments show that it is possible to provide such privacy guarantees, while still scaling to much larger problems than the previous state of the art. When it comes to reasoning under uncertainty, we propose an extension to the DCOP framework, called DCOP under Stochastic Uncertainty (StochDCOP), which includes uncontrollable, random variables with known probability distributions that model uncertain, future events. The problem becomes one of making "optimal" offline decisions, before the true values of the random variables can be observed. We consider three possible concepts of optimality: minimizing the expected cost, minimizing the worst-case cost, or maximizing the probability of a-posteriori optimality. We propose a new family of StochDCOP algorithms, exploring the tradeoffs between solution quality, computational and message complexity, and privacy. In particular, we show how discovering and reasoning about co-dependencies on common random variables can yield higher-quality solutions.
