Low-coherence interferometry based on customized detector arrays

Many biological tissues and a lot of technical objects of interest are to a certain degree transparent to light in the visible and near infrared wavelength range of the electromagnetic spectrum. Nevertheless, it has not been possible for a long time to create clear optical images of these objects due to the multiple scattering of a large part of the employed light. The recent development of low-coherence interferometry (LCI) forms the basis for selecting just the ballistic photons from the back-scattered or transmitted light, and hence enables optical low-coherence reflectometry (OLCR) and optical coherence tomography (OCT) for profiling or imaging with micrometer resolution. All published work until now on OLCR or OCT for cross-sectional imaging relies on point detectors, single-channel electronics and mechanical scanning for 2-dimensional or 3-dimensional imaging, limiting the image acquisition rate to 4-8 per second. This work is devoted to improving the state of the art by enhancing the acquisition speed of OLCR 2D and 3D imaging by systematically exploiting the parallelism offered by custom image sensors with smart pixels based on CMOS technology and by developing a new optical setup without moving parts. We replace the conventional single-channel approach by a multi-channel approach using an array of smart pixels, where each pixel consists of a high-sensitivity photodetector with an offset subtraction circuitry, an amplifier, and an envelope detector. Although dated 2 μm CMOS technology is used for the fabrication, our layout is so compact that a pixel period of only 110 μm is achieved. This makes it possible to integrate linear arrays as well as area arrays of smart pixels. We have successfully demonstrated a 64-pixel line sensor and a 58´58 pixel image sensor in an optical low-coherence reflectometry (OLCR) system, making use of versatile, programmable, high-speed readout electronics we developed. This allows the acquisition of OLCR pixel information in arbitrary sequences for efficient region-of-interest scans. When reading out at a speed of 1 MHz, we can acquire volumetric OLCR data at a repetition rate of 6 Hz, which can be easily improved to the video rate of 25 Hz. The simultaneous photosensing and heterodyne signal detection performed by our smart pixels is carried out close to the photon shot noise limit. In order to simplify OLCR and to reduce system cost, we developed a system based on a novel optical setup with a reflection echelon placed in the reference arm of the interferometer. In combination with the 2D customized high-sensitivity image sensor, this allows high-speed depth cross-sectional measurements without any moving parts, opening the way to cost-effective applications of OLCR in medicine, biology and industry. This thesis has successfully demonstrated that the developed parallel approach to OLCR imaging with CMOS-based smart pixel imagers and a novel optical setup without moving parts is highly promising, representing an important contribution to increasing the practical impact of OLCR imaging.

Multiple scattering in wide-field optical coherence tomography

Optical Coherence Tomography (OCT), a well-established imaging method based on low-coherence interferometry, provides cross-sectional images of the internal structure of biological samples with a resolution in the micrometer range. OCT was successfully applied on various tissues such as for instance the retina, the skin or a tooth. In highly scattering tissues like the skin, probing depth is limited to approximately 2mm, mainly due to insufficient rejection of multiply scattered light. Presently, the contribution of multiple scattering in OCT is not fully understood. Therefore, there is a strong and urgent need to develop models allowing a reliable evaluation of the system's limitations as well as the improvement of the imaging capabilities. It is generally believed that a relevant model should account for loss of correlation between the reference and the sample field due to multiple scattering. We developed a new comprehensive model of OCT. Our preliminary study revealed that the reference and sample fields are actually fully correlated. This important result allowed us to model the OCT signal as a sum of stationary random phasors and treated it as a statistical signal. The mean of this signal can be calculated thanks to classical results of statistical optics and to a Monte Carlo simulation. Unlike other existing models, our model accounts for the source autocorrelation function. The model proved to be in excellent agreement with a whole range of experimental data gathered in a comprehensive study of cross-talk in wide-field OCT. Moreover, our results put in question the applicability of widely used models of OCT based on the "extended Huygens-Fresnel principle", which assume a partial correlation between interfering fields due to multiple scattering. The construction of conventional OCT images is based on lateral scanning of a beam focused within the sample. To increase image acquisition speed and eliminate the need for lateral scanning, wide-field OCT was recently developed. Our experimental and theoretical investigations of the potential and limitations of wide-field OCT revealed the crucial role played by the spatial coherence of the light source. Spatially coherent illumination generates considerable coherent optical cross-talk, which prevents shot-noise-limited detection and diffraction-limited imaging in scattering samples. The dependence on several parameters of the optical system and of the sample properties was investigated in a comprehensive study. Cross-talk increases with the wide-field diameter, numerical aperture, source coherence length, and sample optical density; and strongly depends on sample anisotropy. We showed that spatially incoherent illumination realized with a thermal light source permits cross-talk suppression in wide-field OCT, i.e. rejection of multiply scattered light to a level comparable to that of point scanning OCT. We performed a theoretical study which revealed that the power per spatial mode radiated by thermal light sources is too low to permit a high signal-to-noise ratio while maintaining a fast acquisition speed. Therefore, wide-field OCT realized with either spatially coherent or spatially incoherent illumination suffers from inherent fundamental limitation. This led us to investigate the possibility of exploiting a spatially incoherent light source brighter than a thermal light source. We came to the conclusion that such a "pseudothermal" light source can potentially lead to wide-field OCT systems devoid of cross-talk and an image acquisition speed higher than that of a point scanning OCT system. However, the attractive properties of pseudothermal light sources could be gained at the expense of the simplicity and the economical advantages offered by thermal light sources. Furthermore, fast acquisition speed also relies on a performing "smart pixel detector array". Presently, such detectors do not have sufficient sensitivity and their frequency readout is too low as shown in our feasibility study. We performed a theoretical investigation of the potential of thermal light sources in terms of axial resolution and power per mode. The former revealed that the maximum power per mode is radiated at a wavelength higher than the spectral peak of a blackbody radiator. This led to the important practical conclusion that, at 6000 K, the maximum power is collected in the therapeutic spectral window in OCT (600 - 1300 nm), while at 3000 K this peak is shifted out of the therapeutic window leading to significant power losses. More generally, our work provides a design tool for choosing the optimal thermal light source for a given therapeutic window in terms of signal-to-noise ratio. Currently available sources at 6000 K consist of high pressure gas arc lamps providing a spectrum endowed with spectral lines deleterious for OCT. By suppressing a portion of the spectrum devoid of spectral lines of a mercury arc lamp, we obtained amongst the highest axial resolution so far reported in OCT. Furthermore, the importance of the speckle statistics in OCT incited us to clarify the origin of a difference between two theoretical results reported in the literature. Indeed, two calculations of the amplitude distribution of speckles in OCT, each of them based on a different mathematical formulation, yield different results. We showed that a modification of an initial assumption in one of the formulation leads to equivalent results. In conclusion, this thesis provides a deeper understanding of the potential and limitations of widefield OCT, leading to important design rules. Moreover, it presents a new comprehensive model of OCT putting in question other widely used models.

Mid-infrared surface plasmon resonance sensing applied to refractive index measurements of liquids

There has been a very strong development of the sensors based on surface plasmon resonance during the last thirty years, mostly for biological and biomedical applications. If the first experiments in this field were carried out at the beginning of the 20t century, it would have been necessary to wait for the end of the sixties to see the first devices performing accurate measurements of the refractive indices of gas and liquid samples. Indeed, the surface plasmon resonance phenomenon allows to measure refractive index variations with a high sensitivity that is hardly obtained with other optical measurement techniques. The basic principle consists of exciting the free electrons of a metallic layer with light, which induces the formation of a surface wave called surface plasmon. This resonant phenomenon depends on the wavelength of the excitation light and the refractive indices of the metallic layer and the material located close to the metallic layer. A variation of the refractive index of the latter induces a variation of the resonance condition, which allows to realize optical sensors designed to measure determined samples. An appropriate calibration allows to set a relationship between the variation of the concentration of the sample and the measured refractive index value. Among the various parameters that describe the performances of the sensor, one of the most important is the sensitivity. It is possible to demonstrate that the excitation wavelength has an impact on the sensitivity of the system : the latter increases if the measured sample has an absorption peak at a wavelength close to the excitation wavelength. The main goal of this thesis is to explore this sensitivity enhancement technique based on the absorption properties of the probed medium. A proof-of-principle is experimentally demonstrated with an optical setup constituted of laser sources working in the visible range of the electromagnetic spectrum. However, many materials have absorption peaks located in the mid-infrared range due to their fundamental molecular vibrations. Therefore, the realization of a plasmonic sensor working in the mid-infrared could allow to measure these materials with an increased sensitivity. To the best of our knowledge, surface plasmon resonance based sensors have never been developed above the near-infrared. This thesis presents for the first time the design and realization of a plasmonic sensor working in the mid-infrared range able to measure refractive index variations of gas and liquids samples. Even if the optical sources, components and detectors designed for the mid-infrared are clearly different from the ones designed for the visible range, it has been possible to successfully realize two systems working in the mid-infrared range during this project. The first is able to perform gas measurements. As the excitation wavelength depends on the measured sample, it was necessary to select a laser source that had a wavelength located close to the absorption peak of the gas under measurement. For this purpose, we designed a plasmonic sensor working at a 4.4 µm wavelength able to measure carbon dioxide (CO2) absorbing at 4.3 µm. It has been demonstrated that the experimental sensitivity of this system is five times higher than the theoretical sensitivity obtained with a system working in the visible range at a 633 nm wavelength. The second has been conceived to measure liquid samples in order to measure variations of concentration of CO2 dissolved in water. If measurements of refractive index variations (without using the sensitivity enhancement technique) could be performed successfully with variations of concentration of alcohol in water, further work is necessary to get decisive results for CO2 dissolved in water. This project was carried out in collaboration with an industrial partner in order to take into account some industrial constraints during the development of the sensor. Therefore, particular care has been taken in order to design a system that is compact, mechanically robust, without any mechanical moving parts, cheap and easily reproducible. The technological development presented in this thesis finds many applications mainly in the chemical, biological and biomedical fields. Indeed, many proteins show absorption peaks in the mid-infrared. It could be possible to measure their concentration or the modification of their properties – like their composition or structure – with an increased sensitivity. The interest of surface plasmons is that the measurement is not a transmission-based technique, but rather a "reflection-based technique", as the light does not go through the gas or liquid sample. This measurement technique is also adapted for high volume samples that are present in industrial production processes like water or soft drink quality control.

Design and characterization of a CMOS 3-D image sensor based on single photon avalanche diodes

The design and characterization of an imaging system is presented for depth information capture of arbitrary three-dimensional (3-D) objects. The core of the system is an array of 32 × 32 rangefinding pixels that independently measure the time-of-flight of a ray of light as it is reflected back from the objects in a scene. A single cone of pulsed laser light illuminates the scene, thus no complex mechanical scanning or expensive optical equipment are needed. Millimetric depth accuracies can be reached thanks to the rangefinder's optical detectors that enable picosecond time discrimination. The detectors, based on a single photon avalanche diode operating in Geiger mode, utilize avalanche multiplication to enhance light detection. On-pixel high-speed electrical amplification can therefore be eliminated, thus greatly simplifying the array and potentially reducing its power dissipation. Optical power requirements on the light source can also be significantly relaxed, due to the array's sensitivity to single photon events. A number of standard performance measurements, conducted on the imager, are discussed in the paper. The 3-D imaging system was also tested on real 3-D subjects, including human facial models, demonstrating the suitability of the approach.
