Bottom-up approaches for organizing nanoparticles with polymers

This thesis describes some of three years'work carried at the Centre Suisse d'Electronique et de Microtechnique (Neuchâtel, Switzerland) under the supervision of Dr. Martha Liley and in collaboration with Prof. Horst Vogel of the Ecole Polytechnique Fédérale of Lausanne (Switzerland). The goal of this work is to contribute to the development of innovative technologies in the treatment of surfaces, for the lateral organization of various materials on the micrometer and nanometer scale. The aim is therefore to offer a valid alternative to common lithographic techniques that often require expensive capital equipment and infrastructures. In spite of the high-quality and precision of their products, these techniques often have a limit of ~70 nm on the resolution of the minimum feature size. In this work, techniques based on the ability of some systems to self-organize were used. In particular, the tendency of immiscible polymer mixtures and of block copolymers to form separate phases was exploited. Films made of different polymer phases formed templates for the organization of various materials, with length scale that varied from some 10 µm to some 10 nm. The materials that were most investigated were particles of the diameter of some nanometer, made of semiconductors (CdSe and ZnS) and of metals (Au, CoPt3 and Co). These nanoparticles have different properties from the corresponding bulk material. This fact is due to two main phenomena: in a nanoparticle the electronic energy levels of electrons are discrete because of their spatial confinement. Also, surface effects arise due to the high surface to volume atom ratio compared to the bulk material. As an example, semiconductor particles of the diameter of some nanometers are fluorescent. Semiconductor nanoparticle fluorescence is usually characterized by long fluorescence lifetimes. This fact promoted the investigation, in the framework of this thesis, of the fluorescence properties of Mn-doped ZnS nanoparticles. These particles turned out to have exceptionally long fluorescence lifetimes compared to the fluorophores commonly used as fluorescent markers in biological techniques. This singular property allowed the construction of a simple and cost effective instrument for time-resolved detection of the nanoparticle fluorescence. This technique allows a remarkable increase of the signal-to-noise ratio compared to conventional detection methods. Two approaches were explored to laterally organize nanoparticles on polymer surfaces. In the first method, the nanoparticles were added into a mixture of immiscible polymers and a film was formed from the solution by spin-coating, typically on silicon or glass slides. Numerical simulations by other authors indicate that if the particles have a higher affinity for one of the two polymers, they will be distributed in the corresponding phase. This behaviour was confirmed by our experiments. The lateral dimensions of the patterns in which the nanoparticles organized could be changed with continuity from some 10 µm to se sub-micrometer range. Their shape was modelled from a stochastic to an ordered type. The second approach explored for the organization of the nanoparticles consisted of the pre-formation of the polymer films and their subsequent decoration with the nanoparticles. Due of the different interactions of the two polymers with the particle-solvent system, different absorption behaviours of the nanoparticles were found on the two polymer phases. This technique allowed nanoparticle organization on homopolymer demixed films in patterns having typical sizes in the micron and in the submicron range. Alternatively, the nanoparticles were organized on block copolymer films in regular patterns having typical periodicities of the order of 100 nm. Nanoparticles organized on thin block copolymer films could be transferred on the hard substrate via removal of the polymer molecules by oxygen plasma etching. This process did not affect the nanoparticle organization. Under particular conditions, an aggregation of 10 nm gold nanoparticles was induced using oxygen plasma. This technique allowed the formation of gold nanowires and nanostructures both on polymer layers and on the hard substrate. Their width varied from about 25 nm to the micrometer, while their length extended for various micrometers. They presented a fingerprint like structure or, alternatively, quasi-parallel nanowires extended for several µm2, the typical periodicities being about 100 nm. The conductivity of these nanowires and nanostructures was demonstrated using SEM.

Single molecule detection and fluorescence correlation spectroscopy on surfaces

In this thesis a new approach for single molecule detection and analysis is explored. This approach is based on the combination of two well established methods, fluorescence correlation spectroscopy (FCS) and total internal reflection fluorescence microscopy (TIRFM). In contrast to most existing fluorescence spectroscopy techniques, the subject of primary interest in FCS is not the fluorescence intensity itself but the random intensity fluctuation around the mean value. Intensity fluctuations are induced by thermal noise in a minute observation volume, which is in classical FCS the confocal volume of a confocal microscope. E.g. FCS is commonly utilized to investigate diffusion. In this case, diffusing fluorescent molecules entering or leaving the observation volume cause intensity fluctuations, which are analyzed by calculating the temporal autocorrelation of the observed signal. The autocorrelation is a measure for the self-similarity of a signal and contains information about the average fluctuation strength and duration. The confocal observation volume, i.e. the measurement volume that is actually seen by the detector is approximately given by the product of the optical transfer function with the fluorescence exciting intensity distribution of a focused laser beam. To achieve a high signal-to-background ratio a small observation volume is absolutely essential, first of all because the background from e.g. scattered light increases with the size of the observation volume. Second, a small volume assures for a small average number of fluorophores inside the observation volume and therefore for a high fluctuation amplitude i.e. FCS signal. This thesis proposes and discusses an alternative to confocal FCS specially conceived for measurements on surface-bound molecular systems, such as biological receptors or immobilized enzymes. In contrast to confocal FCS, fluorescence is excited within an evanescent field generated by total internal reflection (TIR) of a laser beam at the interface between a microscope coverslip and the sample. This is achieved by focusing the laser beam off-axis at the back focal plane of a high NA oil-immersion objective. The collimated beam that emerges from the objective is incident at an oblique angle at the coverslip-sample interface and totally internal reflected. In contrast to confocal FCS, the generated observation volume is completely confined to the surface and background fluorescence as well as scattered light from the bulk is efficiently suppressed. Our method, called objective-type TIR-FCS in the following, features an increased collection efficiency compared to existing techniques that combine evanescent wave excitation and FCS. Existing techniques use total internal reflection on the surface of a prism to generate an evanescent field. This leads to a configuration where the choice of objectives is limited to air or water-immersion objectives. In our system we use a high NA oil-immersion objective, specially conceived for TIR applications, which collects light efficiently. The collection efficiency is further enhanced by a naturally occurring change of the emission properties of fluorophores close to interfaces between dielectric media. The presence of the interface favors emission into the optically denser medium so that about 60% of the emitted light can be collected. These factors, together with a reduced observation volume lead to a very sensitive method with a high potential for applications in single molecule detection and analysis. The performance of the proposed method was experimentally shown for measurements on molecules subject to Brownian motion and binding to modified coverslips. In particular, it was experimentally shown that objective-type TIR-FCS features high signal-to-background ratio on a single molecule level. In this thesis, concise derivations of analytical expressions for autocorrelation functions for diffusion and most important, the case of ligands reversibly binding to a single and localized binding site are presented. The derived model allows for the quantitative determination of binding rates for a single receptor. We strongly believe that the application of these results in the context of investigations of receptor-ligand binding kinetics will allow for deeper understanding of cellular signaling. Moreover, this thesis discusses the applicability of the proposed method in enzymology. Enzymes, as most proteins are subject to continuous changes of their structure or conformation. These conformational changes are correlated with the function of the enzyme. In the discussed example the enzyme catalyzes an oxidation where the product is fluorescent but the substrate is not. The function of the enzyme i.e. the recurring product formation leads to observable intensity fluctuations. Since function and conformational states are correlated, conformational fluctuations can be investigated by means of FCS as was already shown for confocal FCS. A technique closely related to FCS is fluorescence lifetime spectroscopy (where lifetime refers to the mean lifetime of the electronic excited state). Whereas in FCS the relaxation after random deviations from thermal equilibrium is investigated, relaxation of excited fluorophores towards their electronic ground-state is investigated in lifetime spectroscopy. The technique is used for e.g. discrimination between fluorophores with different lifetimes. Lifetime spectroscopy combined with imaging is used in many domains of life-science, including microarray reading and medical diagnostics. In preliminary work we developed a novel approach to perform lifetime imaging that is based on a multiplexing technique. The proposed method requires no mechanical scanning stage and only a single-point detector. Furthermore, noise is reduced under certain circumstances if the signal is low. Characteristics of this technique as well as advantages and disadvantages are shortly discussed.

Highly filled polymer nanocomposite films derived from novel nanostructured latexes

The overall aim of this thesis has been to assess the potential of latex-based technologies for the preparation of polymer/clay nanocomposites. The key feature of latex-based technologies is that they offer the possibility of improved control of the final nanocomposite morphology at significantly higher clay loadings than can be obtained with more conventional processing techniques, such as melt blending or in situ polymerization. The idea is to exploit swelling of the clay in either the aqueous or the monomer phase of a water-based latex, depending on the clay surface functionalization, to produce hybrid polymer/clay latex particles with controlled diameters of the order of 100 nm, which may then be consolidated to produce solid nanocomposite films. The materials considered in this work were based on styrenic matrices, considered to be a model system, and acrylics, which are of more interest for commercial coating applications. Two different polymerization techniques were investigated, namely conventional emulsion polymerization and miniemulsion polymerization. The thermal and mechanical properties of films produced from the resulting latexes were then studied in detail. Conventional emulsion polymerization was found to be particularly suitable for the preparation of particles with a well-defined "armoured" morphology, in which the clay formed a more or less complete shell around a matrix core, providing the focus for the remainder of the project. Clay contents of up to about 50 wt % were obtained for both the styrenic and the acrylic latexes using this approach, with excellent degrees of dispersion, the average clay aggregate thickness not exceeding 10 nm. The armoured morphology of the latex particles resulted in a cellular arrangement of the clay in the consolidated films, which became better defined as the clay content increased. The reinforcing effect of the clay on mechanical properties varied according to the physical state of the matrix. Increases in Young's modulus by a factor of 3 to 4 were observed in styrenic films with the cellular morphology in the glassy state, and the degree of exfoliation of the clay was found to be a critical parameter under these conditions, samples containing 5 to 7 wt % of clay showing increased moduli with respect to those obtained at somewhat higher clay contents, for which aggregation was more apparent. In the rubbery state, on the other hand, the Young's modulus increased by more than 2 orders of magnitude for clay contents above 20 wt % and was strongly correlated with the overall filler content. Thermal analysis showed that a significant proportion of the matrix remained immobilized in the rubbery state, i.e. did not contribute to the glass transition. This was argued to be due to strong physical confinement of regions of the matrix intercalated in the clay aggregates. While the increases in Young's modulus in the glassy state could be accounted for in terms of classical micromechanical models, such as those of Halpin-Tsai and Mori-Tanaka, the same models failed to predict the behaviour in the rubbery state. Models based on foam mechanics were therefore developed incorporating an immobilized matrix fraction in the cell walls, whose elastic properties were treated as fitting parameters. Although somewhat different values of the Young's modulus for this immobilized matrix fraction were required to fit the data, depending on the details of the model, they were consistently found to be two to three orders of magnitude greater than that of the neat matrix in the rubbery state (but not to exceed the Young's modulus of the matrix in the glassy state), providing direct evidence for the importance of this interphase for the overall nanocomposite properties. The importance of the cellular arrangement of the clay was also confirmed through comparison with nanocomposites containing non-cellular morphologies resulting from other preparation techniques or the use of mechanical deformation to break-up the initial cellular structure. Finally, it was demonstrated that the results obtained for the styrenic systems could be extended to acrylic-based nanocomposites with comparable morphologies, underlining their broader significance for formulations of potential commercial interest. A further goal of this thesis was to study the effect of the clay on the microdeformation and fracture mechanisms of the nanocomposites, and it was also of interest to compare these results with those obtained for conventional isotactic polypropylene (PP)/clay nanocomposites prepared by melt blending, whose macroscopic properties have been studied previously in our institute. In situ TEM investigation of deformation in glassy styrenic nanocomposite films of about 200 nm in thickness containing the cellular structure revealed a decrease in local matrix drawability at clay contents above 10 wt %, and extensive coarse cavitation, thought to be associated with the particle cores, which replaced crazing as the dominant deformation mechanism, accounting for the observed decrease in macroscopic tensile strength at intermediate clay contents. Moreover, at the highest clay contents, these mechanisms were replaced by failure of the particle-particle interfaces, leading to an extremely brittle macroscopic response. Above Tg, localized deformation zones were also observed, indicating the network formed by the clay and the immobilized regions of matrix to show yielding behaviour, again consistent with the macroscopic response. The conventional PP/clay nanocomposites also showed a decrease in matrix ductility and an increase in coarse cavitation with increasing clay content. In this latter case, however, the zones of cavitation were clearly identified with breakdown of the clay aggregates or the interfaces between the clay and the matrix, underlining the important role of the clay-matrix interface for the properties of the styrenics, and by inference, those of the acrylics. To provide further insight into the role of the cellular structure of the styrenics, finite element (FE) simulations were used to investigate the distribution in hydrostatic stress in the nanocomposite films under finite deformations. These showed the principal stress concentrations to appear in the clay aggregates aligned in the direction of the applied deformation, while the hydrostatic stress in the matrix remained relatively uniform, suggesting that as long as the interface and clay aggregates remain stable, cavitation may initiate anywhere within the matrix, as in the unmodified polymer. The FE simulations were also used to model the elastic properties in the rubbery state, confirming the need to assume the presence of an immobilized matrix fraction in order to account for the observed Young's moduli. The results of these calculations were found to be consistent with those obtained from the foam-based micromechanical models, confirming the applicability of these latter, and suggesting the local anisotropy of the clay aggregates to play a limited role in the overall low strain elastic properties. The main outcome of this thesis has been the establishment of a general physical basis for predicting structure-mechanical property relationships in a new range of latex-based nanocomposite materials with exceptional properties, thanks to the possibility of incorporating very high clay loadings without compromising processability, and vast potential for fine tuning of stiffness and stiffness related properties. Moreover, the work has highlighted the important role of the "nano" effect, particularly at temperatures above the glass transition of the matrix, where matrix immobilization at the clay-matrix interface is clearly demonstrated to contribute to both thermal and mechanical properties. It is consequently expected to provide solid guidelines for the choice of morphological and materials parameters for the optimization of basic mechanical properties in such materials via controlled synthesis. Moreover, with further advances in synthesis and processing and hence in film quality, it should ultimately be possible to extend the basic physical model developed here to account for the structure dependence of other important applicative properties, such as permeability and fire resistance.

Chemistry and Analysis of Nanoparticle-Modified Living Cells

Conventional therapeutics are often limited by their targeting ability, resulting in harmful and potential fatal side-effects for the patients. Recently, new strategies have been developed to improve target specificity of drugs in order to generate more efficient therapeutics. An innovative concept, examined in this thesis, is the use of cells as therapeutic carriers. This concept termed cell-mediated drug delivery, relies on the intrinsic targeting properties of cells to transport the therapeutic drug to the target location. The conjugation of nanoparticles, loaded with drugs or adjuvants, to the surface of cells enables the transport to the desired location and increases the local drug concentration, reducing the required therapeutic leading to further attenuation of side effects. Chapter 1 introduces the concept of cell-mediated drug delivery. It highlights the advantages of using cells decorated with nanoparticles as carriers for therapeutics as opposed to free (polymer) nanoparticles. Examples of various covalent and non-covalent nanoparticle conjugations strategies are discussed and an overview of all approaches, which have been used previously to attach nanoparticles to the cell surface, is presented. The importance of a thorough in vitro characterization of nanoparticle-modified cells is emphasized. The use of common characterization techniques such as flow cytometry, UV-Vis and fluorescence spectroscopy, fluorescence and electron microscopy, and radiolabeling is discussed in detail and examples of qualitative and quantitative in vitro analysis of nanoparticle- modified cells are reviewed. Chapter 2 contains an extensive in vitro analysis of cell-nanoparticle conjugates using fluorescent- and fluorescent label-free methods. The stability of nanoparticles loaded with cargo attached to cells was investigated by encapsulating three different green fluorescent dyes into nanoparticles and tracking the fluorescence signal over 24 h. Two of the dyes revealed a complete loss of fluorescence of a time period of 24 h, whereas a third entrapped dye and a covalently attached dye did not have a decrease in fluorescence over 24 h. Finally, we developed a fluorescent label-free method as an alternative to characterize nanoparticle-decorated cells without the need for any fluorescent label. Chapter 3 systematically explores conjugation chemistries to immobilize two platforms of functional biodegradable polymer nanoparticles (PEG-PLA (poly(ethylene glycol) - poly(lactide)) nanoparticles and carboxylic PLA nanoparticles) on T cells as potential carrier across the blood-brain barrier (BBB). We compared a series of covalent and non-covalent immobilization strategies by flow cytometry (FACS), confocal microscopy, and impact on cell proliferation and cellular viability. We found that T-cell surface coupling of nanoparticles is dependent on the ratio of nanoparticles to cells and that higher ratios led to more particles attached to cells. Furthermore, the most efficient strategy to attach nanoparticles was via ligand-receptor interactions (lectin - sialic acid and biotin - NeutrAvidin). In addition, nanoparticle-decorated T cells were investigated for their ability to cross an in vitro mouse BBB model under static conditions in a two-chamber assay. Decorating the cells with nanoparticles does not affect their ability to bind ICAM-1 (a protein involved in the transmigration process) or to cross the model biological barrier.
