Challenges and Opportunities of Edge AI for Next-Generation Implantable BMIs

Neuroscience and neurotechnology are currently being revolutionized by artificial intelligence (AI) and machine learning. AI is widely used to study and interpret neural signals (analytical applications), assist people with disabilities (prosthetic applications), and treat underlying neurological symptoms (therapeutic applications). In this brief, we will review the emerging opportunities of on-chip AI for the next-generation implantable brain machine interfaces (BMIs), with a focus on state-of-the-art prosthetic BMIs. Major technological challenges for the effectiveness of AI models will be discussed. Finally, we will present algorithmic and IC design solutions to enable a new generation of AI-enhanced and high-channel-count BMIs.

Identifying uncertainty states during wayfinding in indoor environments: An EEG classification Study

The researchers used a machine-learning classification approach to better understand neurological features associated with periods of wayfinding uncertainty. The participants (n = 30) were asked to complete wayfinding tasks of varying difficulty in a virtual reality (VR) hospital environment. Time segments when participants experienced navigational uncertainty were first identified using a combination of objective measurements (frequency of inputs into the VR controller) and behavioral annotations from two independent observers. Uncertainty time-segments during navigation were ranked on a scale from 1 (low) to 5 (high). The machine-learning model, a Random Forest classifier implemented using scikit-learn in Python, was used to evaluate common spatial patterns of EEG spectral power across the theta, alpha, and beta bands associated with the researcher-identified uncertainty states. The overall predictive power of the resulting model was 0.70 in terms of the area under the Receiver Operating Characteristics curve (ROC-AUC). These findings indicate that EEG data can potentially be used as a metric for identifying navigational uncertainty states, which may provide greater rigor and efficiency in studies of human responses to architectural design variables and wayfinding cues.

A system for measuring synchrony between two or more regions, or within a single region, of a subject's brain. The system can include a signal conditioning module in communication with a phase extraction module. The signal conditioning module can receive an oscillating signal, bandpass filter the signal, and convert the signal to its real and imaginary components. The phase extraction module can receive the real and imaginary components, determine the quadrant of the oscillating signal on the complex plane based on the real and imaginary components, and determine the phase of the oscillating signal using a linear arctangent approximation based on the determined quadrant. The system can calculate synchrony metrics based on the determined phase.

Fast and accurate decoding of finger movements from ECoG through Riemannian features and modern machine learning techniques

Objective. Accurate decoding of individual finger movements is crucial for advanced prosthetic control. In this work, we introduce the use of Riemannian-space features and temporal dynamics of electrocorticography (ECoG) signal combined with modern machine learning (ML) tools to improve the motor decoding accuracy at the level of individual fingers. Approach. We selected a set of informative biomarkers that correlated with finger movements and evaluated the performance of state-of-the-art ML algorithms on the brain-computer interface (BCI) competition IV dataset (ECoG, three subjects) and a second ECoG dataset with a similar recording paradigm (Stanford, nine subjects). We further explored the temporal concatenation of features to effectively capture the history of ECoG signal, which led to a significant improvement over single-epoch decoding in both classification (p < 0.01) and regression tasks (p < 0.01). Main results. Using feature concatenation and gradient boosted trees (the top-performing model), we achieved a classification accuracy of 77.0% in detecting individual finger movements (six-class task, including rest state), improving over the state-of-the-art conditional random fields by 11.7% on the three BCI competition subjects. In continuous decoding of movement trajectory, our approach resulted in an average Pearson's correlation coefficient (r) of 0.537 across subjects and fingers, outperforming both the BCI competition winner and the state-of-the-art approach reported on the same dataset (CNN + LSTM). Development of neural interface and brain-machine interface (BMI) systems enables the treatment of neurological disorders including cognitive, sensory, and motor dysfunctions. While neural interfaces have steadily decreased in form factor, recent developments target pervasive implantables. Along with advances in electrodes, neural recording, and neurostimulation circuits, integration of disease biomarkers and machine learning algorithms enables real-time and on-site processing of neural activity with no need for power-demanding telemetry. This recent trend on combining artificial intelligence and machine learning with modern neuralinterfaceswill leadto a new generationof lowpower, smart, and miniaturized therapeutic devices for a wide range of neurological and psychiatric disorders. This paper reviews the recent development of the 'on-chip' machine learning and neuromorphic architectures, which is one of the key puzzles in devising next-generation clinically viable neural interface systems.

Unsupervised Domain Adaptation for Cross-Subject Few-Shot Neurological Symptom Detection

Modern machine learning tools have shown promise in detecting symptoms of neurological disorders. However, current approaches typically train a unique classifier for each subject. This subject-specific training scheme requires long labeled recordings from each patient, thus failing to detect symptoms in new patients with limited recordings. This paper introduces an unsupervised domain adaptation approach based on adversarial networks to enable few-shot, cross-subject epileptic seizure detection. Using adversarial learning, features from multiple patients were encoded into a subject-invariant space and a discriminative model was trained on subject-invariant features to make predictions. We evaluated this approach on the intracranial EEG (iEEG) recordings from 9 patients with epilepsy. Our approach enabled cross-subject seizure detection with a 9.4% improvement in 1-shot classification accuracy compared to the conventional subject-specific scheme.

Predicting task performance from biomarkers of mental fatigue in global brain activity

Objective. Detection and early prediction of mental fatigue (i.e. shifts in vigilance), could be used to adapt neuromodulation strategies to effectively treat patients suffering from brain injury and other indications with prominent chronic mental fatigue. Approach. In this study, we analyzed electrocorticography (ECoG) signals chronically recorded from two healthy non-human primates (NHP) as they performed a sustained attention task over extended periods of time. We employed a set of spectrotemporal and connectivity biomarkers of the ECoG signals to identify periods of mental fatigue and a gradient boosting classifier to predict performance, up to several seconds prior to the behavioral response. Main results. Wavelet entropy and the instantaneous amplitude and frequency were among the best single features across sessions in both NHPs. The classification performance using higher order spectral-temporal (HOST) features was significantly higher than that of conventional spectral power features in both NHPs. Across the 99 sessions analyzed, average F1 scores of 77.5% +/- 8.2% and 91.2% +/- 3.6%, and accuracy of 79.5% +/- 8.9% and 87.6% +/- 3.9% for the classifier were obtained for each animal, respectively. Significance. Our results here demonstrate the feasibility of predicting performance and detecting periods of mental fatigue by analyzing ECoG signals, and that this general approach, in principle, could be used for closed-loop control of neuromodulation strategies.

Migraine is a disabling neurological disorder that can be recurrent and persist for long durations. The continuous monitoring of the brain activities can enable the patient to respond on time before the occurrence of the approaching migraine episode to minimize the severity. Therefore, there is a need for a wearable device that can ensure the early diagnosis of a migraine attack. This brief presents a low latency, and power-efficient feature extraction and classification processor for the early detection of a migraine attack. Somatosensory Evoked Potentials (SEP) are utilized to monitor the migraine patterns in an ambulatory environment aiming to have a processor integrated on-sensor for power-efficient and timely intervention. In this work, a complete digital design of the wearable environment is proposed. It allows the extraction of multiple features including multiple power spectral bands using 256-point fast Fourier transform (FFT), root mean square of late HFO bursts and latency of N20 peak. These features are then classified using a multi-classification artificial neural network (ANN)-based classifier which is also realized on the chip. The proposed processor is placed and routed in a 180nm CMOS with an active area of 0.5mm(2). The total power consumption is 249 mu W while operating at a 20MHz clock with full computations completed in 1.31ms.

Neural interfaces capable of multi-site electrical recording, on-site signal classification, and closed-loop therapy are critical for the diagnosis and treatment of neurological disorders. However, deploying machine learning algorithms on low-power neural devices is challenging, given the tight constraints on computational and memory resources for such devices. In this paper, we review the recent developments in embedding machine learning in neural interfaces, with a focus on design trade-offs and hardware efficiency. We also present our optimized tree-based model for low-power and memory-efficient classification of neural signal in brain implants. Using energy-aware learning and model compression, we show that the proposed oblique trees can outperform conventional machine learning models in applications such as seizure or tremor detection and motor decoding.

ResOT: Resource-Efficient Oblique Trees for Neural Signal Classification

Classifiers that can be implemented on chip with minimal computational and memory resources are essential for edge computing in emerging applications such as medical and IoT devices. This paper introduces a machine learning model based on oblique decision trees to enable resource-efficient classification on a neural implant. By integrating model compression with probabilistic routing and implementing cost-aware learning, our proposed model could significantly reduce the memory and hardware cost compared to state-of-the-art models, while maintaining the classification accuracy. We trained the resource-efficient oblique tree with power-efficient regularization (ResOT-PE) on three neural classification tasks to evaluate the performance, memory, and hardware requirements. On seizure detection task, we were able to reduce the model size by 3.4x and the feature extraction cost by 14.6x compared to the ensemble of boosted trees, using the intracranial EEG from 10 epilepsy patients. In a second experiment, we tested the ResOT-PE model on tremor detection for Parkinson's disease, using the local field potentials from 12 patients implanted with a deep-brain stimulation (DBS) device. We achieved a comparable classification performance as the state-of-the-art boosted tree ensemble, while reducing the model size and feature extraction cost by 10.6x and 6.8x, respectively. We also tested on a 6-class finger movement detection task using ECoG recordings from 9 subjects, reducing the model size by 17.6x and feature computation cost by 5.1x. The proposed model can enable a low-power and memory-efficient implementation of classifiers for real-time neurological disease detection and motor decoding.
