An assay is an investigative (analytic) procedure in laboratory medicine, mining, pharmacology, environmental biology and molecular biology for qualitatively assessing or quantitatively measuring the presence, amount, or functional activity of a target entity. The measured entity is often called the analyte, the measurand, or the target of the assay. The analyte can be a drug, biochemical substance, chemical element or compound, or cell in an organism or organic sample. An assay usually aims to measure an analyte's intensive property and express it in the relevant measurement unit (e.g. molarity, density, functional activity in enzyme international units, degree of effect in comparison to a standard, etc.). If the assay involves exogenous reactants (the reagents), then their quantities are kept fixed (or in excess) so that the quantity and quality of the target are the only limiting factors. Study of loading and delivery of drugs from various hydrogel matrices for orthopaedic applications

Nowadays, there are still several unmet medical needs in the field of orthopaedic and trauma surgery leading to the incentive of finding alternative and innovative clinical therapeutics for the treatment of bone regeneration. For example, the entrapment of osteoinductive growth factors in a polymeric matrix with subsequent gradual release and local delivery to the site of trauma. This new technology inspired the activities relating to my project in which I investigated the potential use of synthetic hydrogels for the sustained in situ release of bioactives. The tasks relating to my project were divided into two main experiments; release assays and bioactivity assays. During the release assays, the in vitro release of three osteoinductive growth factors from five different synthetic hydrogels was investigated. The goal was to characterise the release of the protein drugs from the different polymers, selected on the basis of their swelling and degradation behaviour. The quantification of the released bioactives was assessed by performing an enzyme-linked immunosorbent assay (ELISA), for which different parameters were optimised. As a result, release profiles of the different protein drugs from the different hydrogel formulations were obtained. Then, it was investigated if the growth factors were still biologically active or if incubation in synthetic matrices influenced their properties. This was assessed by performing bioassays during which the alkaline phosphatase (ALP) activity of a mouse myoblast cell line (C2C12 cells) was evaluated. Indeed, ALP is a marker of osteoblastic activity which indicates increased levels of osteogenesis, implying biological activity of the tested osteoinductive bioactives. These results gave a good representation of the effect of the synthetic hydrogels and their incubation in release buffer on the bioactivity of the incorporated protein drugs

New optical tools for non-invasive imaging and quantification of glucose and nicotinamide riboside in living cells and animals

Glucose is one of the most important metabolites that plays a key role in living organisms, as it is the primary energy source. Glucose metabolism is also an integral part in the pathogenesis of diabetes and cancer. Despite this tremendous impact of glucose, there are no tools for imaging of glucose uptake non-invasively and in a real-time manner working in both living cells and animals without utilization of hazardous radiation and suitable for convenient preclinical use. The first part of this thesis is describing the development of a probe for imaging glucose uptake based on a highly sensitive and versatile bioluminescence technique. We set a goal to design a probe, which should be reliable, robust, easy to use and simply represent the glucose uptake in both living cells and animals. In addition the synthesis of the probe should be straightforward and high-yielding. This part of the thesis is composed of two projects utilizing different approaches to the probe design and probeâs mechanism of action. The first project was based on the Staudinger Ligation for imaging of glucose uptake by modifying the glucose molecule with an azide moiety and using luciferin caged with a reactive phosphine as a counter-partner for the assay. The second project describes the development of a cleavable disulfide glucose-conjugate probe. For the first project we aimed to synthesize a library of glucose pro bioluminescent probes, validate them in a cell-free and cell-based assay and identify the hit compound. Then we proved the GLUT-specificity of the uptake of our hit compound in living cells and animals. The robust and reproducible assay method for measurement of glucose uptake was established. The probe was applied for imaging of antidiabetic drugsâ influence on glucose uptake. We have also demonstrated its superior properties over 2-NBDG that is currently the most used probe in preclinical research. However, the probe utilized in the second project namely cleavable disulfide glucose conjugate probe was not so sensitive as glucose-azide. Nevertheless, we showed a successful application of this probe for imaging of glucose uptake in living cells and animals. The second part of this thesis describes the design and evaluation of the bioluminescent probe for imaging of nicotinamide riboside (NR) uptake. NR is a recently discovered NAD+ precursor that is naturally present in cow's milk. Despite its importance in modern research and a high potential as a new medicine for treatment of various types of diseases, including diabetes, there are no tools to visualize NR uptake in preclinical research. Also, there is a lack of knowledge on where and how NR can be absorbed in mammals. Here we established the Staudinger Ligation-based approach to develop an imaging tool for NR uptake. After the successful synthesis of azido-nicotinamide riboside molecule, we demonstrated its specificity and utility for imaging in living cells and animals. Findings described in this part cover not only the imaging of NR uptake, but also the possibility of existence of specific NR transporter in mammalian cells, which has not been discovered and described yet.

Identification of genetic insulator elements to increase the safety of viral gene therapy vectors

Haematopoeitic system diseases, both acquired and inherited, can be currently cured by allogeneic haematopoietic stem cell transplantation. This treatment provides highly successful immune function recovery for patients receiving grafts of HLA-compatible donors but has still a great risk of complications and even failure if no suitable donor is available. Among the alternative therapeutic options, ex vivo retrovirus-mediated gene transfer into haematopoietic progenitor cells has been shown to be an efficient strategy for a substantial number of severe combined immunodeficiency-suffering patients. A recent gene therapy trial has been remarkably effective for the immunological reconstitution of patients suffering from X-linked severe combined immunodeficiency. This treatment was able to provide full correction of disease phenotype and thus, clinical benefit. However, the appearance of leukemia in several patients has put in question the safety of the procedure. This severe adverse event has been attributed to the integration of the therapeutic transgene-carrying viral vector into a known T-cell oncogene, LMO2, thereby contributing to the development of T-cell leukemia by causing aberrant expression of LMO2. Further studies mentioned the possible retroviral-mediated cis-activation of the LMO2 promoter underlying the potential ability of retroviral regulatory elements to influence neighboring gene transcription. This project aimed at decreasing the risk associated with the use of viral vectors for gene therapy through the identification of genetic insulator elements capable of isolating the vector regulatory elements in order to prevent the activation of chromosomal genes by the viral enhancers. We have established a standardized screening procedure whereby the potency of insulators can be assessed quantitatively on relevant vector elements. This assay system consists of a series of plasmids containing two reporter genes: one mimicking a therapeutic gene under the control of strong viral long terminal repeat (LTR) enhancer, and the other one standing for an endogenous gene close to the chromosomal vector integration site. The assay allowed the quantification of the enhancer blocking activity of the well-characterized chicken beta-globin 5'HS4 insulator (cHS4) in cultured cells. We assessed the insulator activity of novel synthetic elements, constructed from optimized binding sites for the insulator protein CTCF. In addition, we demonstrated the enhancer blocking activity of the nuclear factor 1 protein family (CTF/NF1) and showed that it also displays barrier properties, protecting transgene expression from silencing. We showed that both CTCF and CTF/NF1 binding sites act as insulators that mediate potent enhancer-blocker activity, resulting in a commensurate reduction of genotoxicity when implemented in viral gene therapy vectors. Finally, we used the same approach to analyze the enhancer blocking activity of well-known chromatin domain delimiters, matrix attachment regions (MAR), mainly characterized by their barrier properties. We could demonstrate the enhancer blocking ability of the 1-68 MAR that is mainly harbored by an A-T rich core sub-region.
