Building optical matter with binding and trapping forces

Very high frequency oscillations of intense light fields interact with micron-size dielectric objects to exert dc optical forces that allow polarizable particles to levitate, to be trapped and to be bound. Such optical forces are also suitable to arrange cold atoms in optical lattices. Various assemblages of optical traps, including periodic arrays, can be constructed either with independent lasers, or with a single laser beam split into different parts later recombined by interference, as well as through the use of diffractive elements. These optical-well arrays serve as templates for writing and erasing dynamic two-dimensional and three-dimensional "optical crystals", composed of mono-dispersed polystyrene spheres in water. Subsequently, the crystals become diffractive structures themselves. The association of micro-fluidics and optical trapping allows for the formation of optical traps into micro-channels. This leads to perform microchemistry experiments, such as fluorescence detection, on individual bodies attached to trapped particles. Self-trapping due to the optical binding force relates to the interaction between different dielectric objects located in an electromagnetic field; each one reacts not only to the field of the incident beam, but also to the induced fields radiated coherently by all other particles. Optical binding strongly influences the equilibrium state and the behavior of optical crystals. It must have the potential for creating collective effects.

Light structuring for massively parallel optical trapping

Optical trapping, discovered in the 70's, allows moving and stabilizing small objects which sizes varies from atoms to particles of several microns. This technique, based on momentum conservation, is particularly well suited for manipulating biological matter (cells, organelles, vesicles, functionalized particles, etc.) and offers interesting potentialities for research in biotechnologies and biochemistry. The possibility to individually immobilize large numbers of microscopic objects opens new ways for the downscaling of analysis tools for drug screening, particles sorting or assessing statistical data. The combination of optical trapping with microfluidics greatly increases the prospect of the method. This PhD work takes place in a research aiming at creating large arrays of optical traps compatible with microfluidic devices in order to realize so-called lab-on-a-chip. These miniaturized systems allow recreating at smaller time scale, reduced resources and lower cost, experiments usually performed in a macroscopic environment. This study proposes solutions based on light interference and on landscaping of light intensity. Setups combining several laser beams are proposed to create interference patterns and various configuration of light potential wells. Increasing the number of interfering beams, in particular by using a multiple beams interferometer (Fizeau-Tolansky interferometer) leads to a raise of the light intensity gradient, further increasing the trapping efficiency. The quality of the optical traps is studied and discussed in comparison with conventional laser tweezers. More complex and original solutions using interference of electromagnetic fields are suggested. Namely, the light diffracted by the objects themselves is used to form new potential wells. Diffractive structures are devised to generate three-dimensional arrays of traps. The periodicity of those planar structures creates a self-imaging phenomenon, known as Talbot effect. The modulation of the field in the Fresnel zone, i.e. some tens of micrometers behind the diffractive element, reveals interesting properties for optical trapping, in particular local intensity amplification and gradient enhancement. When several particles are simultaneously immersed in an electromagnetic field, interaction effects arise, that link the particles. This phenomenon of optical binding, is studied and demonstrated here in the case of bidimensional optical crystals.

Developing luminescent Brownian probes for near-field investigations of the intracellular environment

Life sciences have a constantly growing need for novel methodological approaches suitable to investigate the intracellular environment with increased temporal and spatial resolutions. Recently, optical near-field probes, such as laser-irradiated pointed metal, uncoated/metal-coated tapered optical fibers, as well as nano-emitters, such as single molecules or nanoparticles, have attracted increasing attention as key components of high-resolution microscopes. In parallel, super-resolution fluorescence microscopy, operating in far-field regime and overcoming the light diffraction limit, has markedly improved imaging resolution. Although both near- and far-field optical approaches drastically improve image resolution, they still represent numerous drawbacks, such as, technical difficulties concerning probe preparation (near-field) or potential damaging through very high light intensities (far-field). The aforementioned shortcomings of high-resolution optical microscopy can be overcome to a great extent by photonic force microscopy (PFM). PFM employs a strongly focused near-infrared (NIR) laser light to hold a dielectric or metallic particle as a local probe. Such an optical trap enables one to ‘cage’ a mesoscopic particle and track its three-dimensional (3D) thermal fluctuations in the surrounding environment, e.g. a viscous liquid. Therefore, besides offering near-field 3D imaging, PFM reports also on other important quantities concerning local mechanical properties, including force and viscosity as well as dynamical properties of the surrounding medium. This additional information is derived from the careful analysis of the jittery motion (so-called Brownian motion) of the trapped single-particle probe that collides with thermally-activated surrounding molecules. In this thesis, we first study in detail the Brownian motion of a single spherical particle that is confined within the harmonic potential of the optical trap. To this end, we optimize the NIR light path and electronic noise floor of our custom-built PFM set-up for detecting and quantifying resonances in the Brownian motion. These resonances arise from the coupling between the hydrodynamic memory and strong strength of the optical trap. Due to the high sensitivity of the short-time dynamics, the size of the particle can be measured by simultaneous fitting of the velocity autocorrelation function and power spectral density of its thermal fluctuations. In order to choose the best-suitable spherical probe for a given experiment in PFM, computational modeling based on a Matlab toolbox is performed. The generalized Lorenz-Mie theory is computed using the T -matrix method for various experimental conditions, including changes in the size and refractive index of the sphere, as well as different laser polarization states. The axial equilibrium position is examined to predict its location compared with the position of the laser focus. Optical forces acting on the sphere are investigated in 3D to highlight, in particular, the limits of the perfectly harmonic potential assumption. These limits are quantified and discussed. Subsequently, we identify different types of inorganic nano/micro-sized particles that can be trapped by the NIR laser of the PFM and used simultaneously as mechanical and near-field light sources. To this end, we take advantage of the visible light emission from single probes confined in the optical trap and excited by the NIR laser light. The emphasis is on particles revealing nonlinear optical properties. In particular, nonlinear optical field enhancement resulting from excitation of surface plasmon resonance (SPR) on trapped gold particles of 200 nm is demonstrated by measuring the emission of that second-harmonic generation (SHG). Furthermore, we show, under optical trapping conditions, SHG emission from single potassium niobate (KNbO3) nano/micro-sized particles. We also report on the multicolor upconversion luminescence (UCL), at the single particle level, for randomly shaped crystals of sodium yttrium fluoride codoped with ytterbium and erbium, NaYF4:Yb,Er (UCC), when they are held in the optical trap. In parallel, 3D Brownian fluctuations of a single trapped particle are quantified. Moreover, luminescence resonance energy transfer (LRET) between a KNbO3 particle or an UCC and molecules of an organic dye (rose bengal) adsorbed on their surface is highlighted and analyzed. Finally, randomly shaped UCCs and hexagonal nanoplates are characterized in detail at the single particle level. The hexagonal β phase in the polycrystalline and monocrystalline crystals is identified, in both randomly and hexagonally shaped particles by electron microscopy. UCL emission from the trapped crystal is quantified in terms of power and found to be dependent on the laser power density, size and shape of the particle as well as its orientation within the trap. Most importantly, the different colors in the UCL spectrum do not vary with the same trend for different orientations of the randomly or hexagonally shaped crystal upon trapping. This suggests the existence of crystalline anisotropy-dependent UCL. Thereby, we should be able to design the best-suited particle probe for LRET experiments with subwavelength resolution.

Micro-optics for multiple laser trapping in microfluidics

Optical trapping allows manipulating very small objects – varying from Angstöms to micron size particles – without physical contact by taking advantage of laser light. This technique, relying on light momentum transfer, allows manipulating biological matter (such as cells, organelles, vesicles or chemically functionalized artificial particles, etc.) and offers promising potentialities for research in biotechnology and biochemistry. Individually confining a large number of microscopic objects opens new ways for the downscaling of analysis tools for drug screening, particles sorting and the assessment of statistical data. In particular, the combination of optical trapping with microfluidics greatly enlarges the possibilities offered by both techniques. This PhD work takes place in a research aiming at developing novel bio-analytical instrumentation relying on large arrays of optical traps compatible with microfluidic systems. The dissertation focuses on the use of micro-optics for generating extended matrices of three-dimensional optical traps capable of capturing a large number of particles within microfluidic environments. The stability of optical traps is first studied with special emphasis on the conditions present in microfluidic flows, both from a theoretical and an experimental point of view, and trapping forces achievable on biological cells are investigated. A multiple optical trapping system relying on microlens arrays, adapted to work on commercial microscopes, is shown to be capable of generating arrays of more than 500 three-dimensional traps. Simultaneously trapping in multiple planes is also evidenced using matrices of microlenses, thanks to a self-imaging effect of the array of traps. Furthermore, possibilities offered by multiple optical trapping within microfluidic environments are explored. Polystyrene spheres as well as biological particles, such as native vesicles, can be trapped in arrays, manipulated inside microfluidic devices and analyzed in parallel through fluorescence microscopy while extremely small quantities of chemical reagents are flown past the array. Finally, for the sake of system miniaturization and further extension of the number of traps, multiple three-dimensional optical trapping based on arrays of miniaturized high numerical aperture parabolic mirrors is proposed, allowing the optics necessary for optical trapping, fluorescence excitation and collection to be integrated at the level of a microfluidic chip. Such miniaturized mirror matrices did not even exist before this thesis; their fabrication and characterization are detailed in this dissertation.
