Impact Assessment of Pumping Parameters on Groundwater Bacterial Communities

Groundwater microbial community samples are traditionally collected using pumping techniques optimized for groundwater chemistry assessment, although the impact of groundwater pumping parameters on apparent bacterial community structures (BCSs) is not really known. We therefore studied the impact of pumping lift, flow regime, and tubing material on BCS, which were analyzed by terminal-restriction fragment length polymorphism (T-RFLP). Ruzicka dissimilarity coefficients were calculated between T-RFLP profiles to assess disparities between BCS. Variations in pumping lift, flow regime, and tubing material did not affect the apparent BCS in experiments using a homogenous water system under laboratory conditions showing that the conditions within the tube had no detectable effect on BCS. However, pumping groundwater from aquifer monitoring wells at different flow rates in the field revealed a significant impact on the apparent BCS. Water samples collected from fine sediment were the most affected by the pumping flow rate.

Study of the microbial communities of core and water samples in a chloroethenes contaminated aquifer

Widely used in industrial activities, chloroethenes have become common groundwater contaminants. Dehalorespiration is an anaerobic bacterial respiration coupling their sequential degradation to energy production. The interactions between the dehalorespiring bacteria and the other guilds and environmental variables are presumed to influence the dehalorespiration efficiency. Here, water and core samples were extracted from the PCE-contaminated site of Lyss (BE, Switzerland). Bacterial DNA was extracted and the 16S rRNA gene amplified by PCR. Terminal restriction fragment length polymorphism (T-RFLP) was used to assess the diversity of the bacterial communities. Core and groundwater environmental variables were analyzed as well. The T-RFLP profiles and the environmental data obtained were processed using statistical multivariate analysis. Within a period of two months, the bacterial communities remained relatively specific to their respective sampling origins. Communities from the oxic layer shared a similar set of bacterial species whereas piezometers from the anoxic layer had a specific one. The T-RFLP profiles obtained from single depth and multi-level piezometers were clearly distinct, probably due to differences in the pumping flow rates and their associated transport regimes, which can have an impact on i) the sediments extracted from the aquifer, ii) the hydraulic zone reached and iii) a potential selective bacterial adsorption onto the pumping tube wall. Environmental factors related to the groundwater redox state appeared as the major variables influencing the bacterial populations. Bacterial communities from core samples clustered into two distinct sets: i) samples from the strongly reductive silt layer, and ii) samples from the coarser and less reductive layer. Grain size distribution and redox state both had a major influence on the bacterial communities of core samples. Finally, it was shown that microbial populations extracted from groundwater differed from the ones extracted from core material, and thus were not representative of the overall aquifer microbial diversity.

Hydroxylated Metabolites of β- and δ-Hexachlorocyclohexane: Bacterial Formation, Stereochemical Configuration, and Occurrence in Groundwater at a Former Production Site

Estrogenic substances discharged from wastewater treatment plants have been detected in surface sediments of receiving waters, but little is known of their vertical migration through buried sediments and their potential to contaminate subsurface waters. The vertical profiles of estrogenic chemicals were investigated in sediment cores at an alluvial freshwater site (Ditchling) and a clay-rich estuarine site (Lewes), both of which are downstream of wastewater discharges into the River Ouse (Sussex, U.K.). Estrone (E1) was the predominant estrogen detected in surface and buried sediments at both sites and was detected in undisturbed clay sediments >120 years old. Profiles of E1 at Ditchling were characterized by a prominent subsurface peak of E1 at the alluvium/clay interface (-15 cm) at a concentration (28.8 ± 6.0 ng/g of dry wt) that was 9-fold higher than in the surface sediment. In contrast, a steady downcore decline in E1 concentrations was observed in the clay-rich Lewes core. This work provides the first in situ evidence of estrogen migration through river bed sediments and reveals that movement of estrogens through unconsolidated sediment can result in penetration to the underlying substrata and therefore the potential for groundwater contamination.

Design approach for the improvement of the economic and environmental performances of potable water supply

Triggered by the Kyoto Protocol and by the aggravation of freshwater scarcity, environmental impacts should soon become key decision criteria for the planning of potable water supply projects, especially when advanced systems such as seawater desalination are at stake. In order to foster the transition of the water industry towards sustainable practices, the present work proposes an integrated design approach dedicated to potable water supply that targets both economic and environmental objectives. At first, the current industrial practices (Chapter I) and the technical characteristics of potable water supply systems (Chapter II) are analyzed via the modeling of the process units used for potable water supply: pumping systems, conventional water treatment processes (e.g. clarification, filtration, disinfection) and advanced water treatment processes (e.g. membrane processes, thermal processes). Using the ISO 14040 standardized Life Cycle Assessment (LCA) method, Chapter III presents the development of a performance indicator that assesses the environmental impacts generated through all stages of the life cycle of potable water supply (i.e. from "cradle to grave"): from the construction of the potable water treatment plant, to its operation and decommissioning. This LCA-based indicator provides a holistic overview of all potential environmental impacts (e.g. green house gases (GHG) emissions, impacts on ecosystems and on human health). It allows to stress out the impact sources and the penalizing steps within potable water supply (Chapter IV): The production of electricity and chemicals required by the potable water treatment plant is highlighted to generate respectively 75% and 15% of the total impacts generated during the life cycle of potable water supply. Different potable water supply scenarios (e.g. potable water supply from ground water, from surface water, seawater desalination, water import from distant water resources) are benchmarked, in order to identify the best solutions as a function of the local context (e.g. type of electricity supply, topographic conditions, feed water quality). Based on the results of the environmental assessment, Chapter V proposes measures for the improvement of the industrial practices of the water sector, targeting energy and chemicals management, electricity sourcing and effluent disposal. Within the same perspective, Chapter VI details an optimization method for the design of the reverse osmosis (RO) membrane process, i.e. the key treatment process for desalination and wastewater reclamation. This method systematically synthesizes RO process configurations and evaluates their performances on economical (total annualized costs), technical (energy requirement, water conversion rate) and environmental criteria (GHG emissions). Evolutionary algorithms are then used to optimize the design of these configurations (process layout and operating conditions) and to identify those featuring the best trade-offs between economical costs and environmental impacts. As case study, the optimization method is applied on a brackish water reverse osmosis (BWRO) desalination project. The characteristics of the optimal BWRO process configurations are calculated as a function of the project constraints (e.g. economical and technical settings, minimum potable water quality), in order to illustrate how this method may support process engineers for the design of desalination plants.
