The neural code between neocortical pyramidal neurons depends on neurotransmitter release probability

Although signaling between neurons is central to the functioning of the brain, we still do not understand how the code used in signaling depends on the properties of synaptic transmission. Theoretical analysis combined with patch clamp recordings from pairs of neocortical pyramidal neurons revealed that the rate of synaptic depression, which depends on the probability of neurotransmitter release, dictates the extent to which firing rate and temporal coherence of action potentials within a presynaptic population are signaled to the postsynaptic neuron. The postsynaptic response primarily reflects rates of firing when depression is slow and temporal coherence when depression is fast. A wide range of rates of synaptic depression between different pairs of pyramidal neurons was found, suggesting that the relative contribution of rate and temporal signals varies along a continuum. We conclude that by setting the rate of synaptic depression, release probability is an important factor in determining the neural code.

Synaptic transmission in the neocortex is dynamic, such that the magnitude of the postsynaptic response changes with the history of the presynaptic activity. Therefore each response carries information about the temporal structure of the preceding presynaptic input spike train. We quantitatively analyze the information about previous interspike intervals, contained in single responses of dynamic synapses, using methods from information theory applied to experimentally based deterministic and probabilistic phenomenological models of depressing and facilitating synapses. We show that for any given dynamic synapse, there exists an optimal frequency of presynaptic spike firing for which the information content is maximal; simple relations between this optimal frequency and the synaptic parameters are derived. Depressing neocortical synapses are optimized for coding temporal information at low firing rates of 0.5-5 Hz, typical to the spontaneous activity of cortical neurons, and carry significant information about the timing of up to four preceding presynaptic spikes. Facilitating synapses, however, are optimized to code information at higher presynaptic rates of 9-70 Hz and can represent the timing of over eight presynaptic spikes.

Spike-Based Reinforcement Learning in Continuous State and Action Space: When Policy Gradient Methods Fail

Changes of synaptic connections between neurons are thought to be the physiological basis of learning. These changes can be gated by neuromodulators that encode the presence of reward. We study a family of reward-modulated synaptic learning rules for spiking neurons on a learning task in continuous space inspired by the Morris Water maze. The synaptic update rule modifies the release probability of synaptic transmission and depends on the timing of presynaptic spike arrival, postsynaptic action potentials, as well as the membrane potential of the postsynaptic neuron. The family of learning rules includes an optimal rule derived from policy gradient methods as well as reward modulated Hebbian learning. The synaptic update rule is implemented in a population of spiking neurons using a network architecture that combines feedforward input with lateral connections. Actions are represented by a population of hypothetical action cells with strong mexican-hat connectivity and are read out at theta frequency. We show that in this architecture, a standard policy gradient rule fails to solve the Morris watermaze task, whereas a variant with a Hebbian bias can learn the task within 20 trials, consistent with experiments. This result does not depend on implementation details such as the size of the neuronal populations. Our theoretical approach shows how learning new behaviors can be linked to reward-modulated plasticity at the level of single synapses and makes predictions about the voltage and spike-timing dependence of synaptic plasticity and the influence of neuromodulators such as dopamine. It is an important step towards connecting formal theories of reinforcement learning with neuronal and synaptic properties

The precise times of occurrence of individual pre- and postsynaptic action potentials are known to play a key role in the modification of synaptic efficacy. Based on stimulation protocols of two synaptically connected neurons, we infer an algorithm that reproduces the experimental data by modifying the probability of vesicle discharge as a function of the relative timing of spikes in the pre- and postsynaptic neurons. The primary feature of this algorithm is an asymmetry with respect to the direction of synaptic modification depending on whether the presynaptic spikes precede or follow the postsynaptic spike. Specifically, if the presynaptic spike occurs up to 50 ms before the postsynaptic spike, the probability of vesicle discharge is upregulated, while the probability of vesicle discharge is downregulated if the presynaptic spike occurs up to 50 ms after the postsynaptic spike. When neurons fire irregularly with Poisson spike trains at constant mean firing rates, the probability of vesicle discharge converges toward a characteristic value determined by the pre- and postsynaptic firing rates. On the other hand, if the mean rates of the Poisson spike trains slowly change with time, our algorithm predicts modifications in the probability of release that generalize Hebbian and Bienenstock-Cooper-Munro rules. We conclude that the proposed spike-based synaptic learning algorithm provides a general framework for regulating neurotransmitter release probability.
