X-ray and Cryo-electron Microscopy Structures of Monalysin Pore-forming Toxin Reveal Multimerization of the Pro-form

beta-Barrel pore-forming toxins (beta-PFT), a large family of bacterial toxins, are generally secreted as water-soluble monomers and can form oligomeric pores in membranes following proteolytic cleavage and interaction with cell surface receptors. Monalysin has been recently identified as a beta-PFT that contributes to the virulence of Pseudomonas entomophila against Drosophila. It is secreted as a pro-protein that becomes active upon cleavage. Here we report the crystal and cryo-electron microscopy structure of the pro-form of Monalysin as well as the crystal structures of the cleaved form and of an inactive mutant lacking the membrane-spanning region. The overall structure of Monalysin displays an elongated shape, which resembles those of beta-poreforming toxins, such as Aerolysin, but is devoid of a receptorbinding domain. X-ray crystallography, cryo-electron microscopy, and light-scattering studies show that pro-Monalysin forms a stable doughnut-like 18-mer complex composed of two disk-shaped nonamers held together by N-terminal swapping of the pro-peptides. This observation is in contrast with the monomeric pro-form of the other beta-PFTs that are receptor-dependent for membrane interaction. The membrane-spanning region of pro-Monalysin is fully buried in the center of the doughnut, suggesting that upon cleavage of pro-peptides, the two disk-shaped nonamers can, and have to, dissociate to leave the transmembrane segments free to deploy and lead to pore formation. In contrast with other toxins, the delivery of 18 subunits at once, nearby the cell surface, may be used to bypass the requirement of receptor-dependent concentration to reach the threshold for oligomerization into the pore-forming complex.

Progress in the analysis of membrane protein structure and function

Structural information on membrane proteins is sparse, yet they represent an important class of proteins that is encoded by about 30% of all genes. Progress has primarily been achieved with bacterial proteins, but efforts to solve the structure of eukaryotic membrane proteins are also increasing. Most of the structures currently available have been obtained by exploiting the power of X-ray crystallography. Recent results, however, have demonstrated the accuracy of electron crystallography and the imaging power of the atomic force microscope. These instruments allow membrane proteins to be studied while embedded in the bi-layer, and thus in a functional state. The low signal-to-noise ratio of cryo-electron microscopy is overcome by crystallizing membrane proteins in a two-dimensional protein-lipid membrane, allowing its atomic structure to be determined. In contrast, the high signal-to-noise ratio of atomic force microscopy allows individual protein surfaces to be imaged at subnanometer resolution, and their conformational states to be sampled. This review summarizes the steps in membrane protein structure determination and illuminates recent progress. 3D reconstruction of two-dimensional crystals

Electron crystallography of two-dimensional (2D) crystals determines the structure of membrane proteins in the lipid bilayer by imaging with cryo-electron microscopy and image processing. Membrane proteins can be packed in regular 2D arrays by their reconstitution in the presence of lipids at low lipid to protein weight-to-weight ratio. The crystal quality depends on the protein purity and homogeneity, its stability, and on the crystallization conditions. A 2D crystal presents the membrane protein in a functional and fully lipidated state. Electron crystallography determines the 3D structure even of small membrane proteins up to atomic resolution, but 3D density maps have a better resolution in the membrane plane than in the vertical direction. This problem can be partly eliminated by applying an iterative algorithm that exploits additional known constraints about the 2D crystal. 2D electron crystallography is particularly attractive for the structural analysis of membrane proteins that are too small for single particle analyses and too unstable to form 3D crystals. With the recent introduction of direct electron detector cameras, the routine determination of the atomic 3D structure of membrane-embedded membrane proteins is in reach. Synthesis, crystal growth and physical properties of the frustrated spin ladder material BiCu2PO6

In recent years quantum antiferromagnets with an intrinsically disordered (“spin liquid”) ground state and an energy gap in the spin excitation spectrum have received a great deal of attention. In search for new experimental realizations of spin-ladder and related models, we have embarked upon a new interesting family of low-dimensional materials with the general formula BiCu2AO6 (A = P, V and As). Among them, BiCu2PO6 shows a structure where Cu spins appear to form a two-leg zigzag ladder, which is believed to be a valuable model of a quantum magnet. However, for detailed studies of quantum magnetism, single crystals are indispensible. Therefore, in this thesis, synthesis and characterization of the newly-discovered spin ladder material BiCu2PO6 (BCPO) and its derivatives in the form of powder and single crystal are presented. Polycrystalline samples of Bi(Cu1-xZnx)2PO6 (x= 0.01, 0.02, 0.03, 0.04, 0.05 and 1), Bi1-y(Ca, Sr, La )yCu2PO6 (y = 0.15, 0.5 and 1) and BiCu2(P1-zVz)O6 (z = 1) were synthesized in this work. The solid state synthesis reactions, phase purity and thermal properties of the samples, substitution solubility, and preliminary magnetic susceptibility have been studied by thermal analysis (TG-DTA), X-ray powder diffraction (XRD), and Scanning Electron Microscope (SEM) as well as DC magnetization measurements, respectively. The BiCu2PO6 structure shows a remarkable chemical flexibility and interesting substitution effects. For detailed studies of the magnetic properties, well characterized large single crystals are indispensable. Thus in this thesis, a number of single crystal growth experiments have been performed using the Travelling Solvent Floating Zone method (TSFZ). Under the elaborated crystal growth conditions (growth rate of ~1 mm/h, gas mixture of 20% O2 in Argon, total 3 pressure (6~6.5 bar), high quality cm -size single crystals of Bi(Cu1-xZnx)2PO6 (x = 0, 0.01, 0.03, 0.05) have been successfully grown for the first time. For these crystals phase purity, crystal morphology/orientation, defect density, chemical composition distribution, crystal structure (room temperature and 1.5 K) and single crystal quality were thoroughly studied based on X-ray and neutron powder diffraction, optical and scanning electron microscopy, X-ray/neutron Laue diffraction, ICP-OES/X-ray fluorescence and two-axis neutron diffraction. Moreover, the specific heat Cp (H,T), AC susceptibility χ(H,T) and DC magnetization M(H,T) measurements on well-characterized single crystal samples of the undoped BCPO and 5% Zn-doped BCPO with the field applied along main crystallographic axes were performed in order to study the unique low temperature magnetic behaviour exhibited in this system, such as anisotropy of the magnetic properties, impurity-induced magnetism by the substitution of non-2+ magnetic cations (e.g. Zn ) and frustration. The magnetic phase diagram (H-T) under low field has been constructed. Further results obtained by close collaborators, based on the measurements made on single crystal samples grown in this thesis, including field-induced quantum phase transitions, NMR, Raman studies are attached directly at the end of this thesis.
