Résumé

The invention relates to a technique to detect small molecules using Bioluminescence imaging (BLI) to image and quantify non-invasively, in vitro and in vivo,intracellular metabolite fluxes and which can be applied to azido-modified compounds, such as azido-modified biomolecules.

Bioluminescence imaging

Bioluminescence imaging (BLI) is a technology developed over the past decades (1990's and onward). that allows for the noninvasive study of ongoing biological processes Recently, bioluminescence tomography (BLT) has become possible and several systems have become commercially available. In 2011, PerkinElmer acquired one of the most popular lines of optical imaging systems with bioluminescence from Caliper Life Sciences. Bioluminescence is the process of light emission in living organisms.

Les luciférines sont des molécules dont l'oxydation, sous le contrôle d'une enzyme, la luciférase, aboutit à la formation d'oxyluciférine et à l'émission de photons. La luciférine a été découverte chez plus de 300 espèces capables de bioluminescence. Il existe cinq types de luciférine, chacun étant lié à une luciférase spécifique. Luciférine de luciole L'espèce la plus étudiée est Photinus pyralis (le lampyre), dont la réaction de bioluminescence nécessite la présence d'ATP et de magnésium.
