This lecture covers the principles and applications of Zernike phase-contrast imaging, a technique complementary to propagation-based phase-contrast imaging. It explores the ability to provide high-resolution images of internal structures with high contrast, particularly valuable for studies of weakly absorbing complex biological systems. The working principle of Zernike phase-contrast imaging is explained, along with techniques to optimize contrast. An example involving Zernike microscopy on Thale cress seed is presented.
