Benzene is an organic chemical compound with the molecular formula C6H6. The benzene molecule is composed of six carbon atoms joined in a planar ring with one hydrogen atom attached to each. Because it contains only carbon and hydrogen atoms, benzene is classed as a hydrocarbon. Benzene is a natural constituent of petroleum and is one of the elementary petrochemicals. Due to the cyclic continuous pi bonds between the carbon atoms, benzene is classed as an aromatic hydrocarbon. Benzene is a colorless and highly flammable liquid with a sweet smell, and is partially responsible for the aroma of gasoline. It is used primarily as a precursor to the manufacture of chemicals with more complex structure, such as ethylbenzene and cumene, of which billions of kilograms are produced annually. A laboratory study of heterogeneous reactions relevant to the atmospheric boundary layer

The present work deals with two subjects. The interaction of NO2 and HONO with different types of soot are examined in the first part whereas in the second part an experimental set-up is presented which has been built in order to measure the kinetics of the degradation of organic compounds by OH radicals. Both soot particles as well as NO2 are mainly produced by fossil fuel and biomass burning. The two species are therefore ubiquitous in the atmospheric boundary layer where they may react with each other. It has been shown that one possible product of this heterogeneous interaction is nitrous acid (HONO). HONO is an important trace gas in atmospheric chemistry because it is easily photolysed resulting in OH radical and NO. Thus, the photolysis of HONO significantly enhances photooxidation processes early in the morning. In the present study two different types of decane soot have been produced. It has been shown that the air/fuel ratio of the diffusion flame is a key parameter influencing the reactivity of soot towards NO2. Whereas soot from a rich flame ("grey" decane soot) leads to HONO with yields up to 100% upon interaction with NO2, only small amounts of HONO, but significant amounts of NO are formed in the presence of soot originating from a lean flame ("black" decane soot). A reaction mechanism has been developed showing that NO2 is initially converted to HONO by a redox reaction. HONO produced in this way is subsequently either quantitatively desorbed into the gas phase on "grey" decane soot or it decomposes to a large extent in a disproportionation reaction on "black" decane soot. The products of disproportionation are NO, which is instantaneously desorbed into the gas phase and NO2, which undergoes secondary reactions. In addition to the HONO producing pathway there is a reaction channel that leads to adsorption of NO2 for both types of soot which is irreversible on the time scale of our standard experiments. Saturation effects that are occurring on the time scale of our experiments are primarily caused by saturation of adsorption sites and not by the depletion of the reducing agent. It seems that the fraction of NO2 that is irreversibly adsorbed is blocking part of the surface sites where NO2 is initially adsorbed. Uptake coefficients γ take initial values of up to 0.1 for both types of soot. However, with increasing uptake of NO2 γ is decreasing. After a NO2 consumption of approximately 8×1013 molecule cm-2, which corresponds to 13% of a formal monolayer, γ drops to values of 3×10-7 for "grey" decane soot and 6×10-7 for "black" decane soot, respectively. Furthermore, our results show that the rate of initial uptake is the rate limiting step of the NO2/soot interaction mechanism. Experiments where "black" decane soot has been exposed to a flow of HONO revealed that it readily decomposes into NO and NO2. For HONO concentrations above 3.3×1011 molecule cm-3 a total product yield of 50%, whereof 40% NO and 10% NO2, has been observed. The initial uptake coefficient ( γ0) for HONO on "black" decane soot did not show clear saturation effects but varied randomly in the concentration range from 1.2×1012 to 5.9×1012 molecule cm-3. The average value measured at these concentrations was equal to 0.027. The strong interaction of HONO with "black" decane soot shows that soot is not only a potential source, but also a potential sink of HONO. On the other hand, no significant uptake could be observed when "grey" decane soot was exposed to HONO. This is consistent with the observation that NO2 is converted to HONO at yields of up to 100%. Acetylene soot is an additional type of soot that has been examined. The results for the initial uptake coefficients were basically the same as for "grey" and "black" decane soot, that is maximum values of γ0 of 0.1 for NO2 uptake decreasing with increasing concentrations of NO2. This similarity holds only for the first few seconds of interaction because acetylene soot saturates much faster than "grey" and "black" decane soot with continuous exposure. The acetylene samples are generally almost completely saturated after only three minutes of exposure to NO2. Soxhlet extractions of "grey" decane soot which have been performed using tetrahydrofuran as a solvent also produced HONO upon interaction with NO2. This shows that the active reactants are not part of the graphitic soot backbone but are extractable, rather polar compounds of the organic fraction of soot. This assumption is supported by the observation that the corresponding benzene extraction is not reactive in relation to NO2 uptake and HONO formation. In the second part of the present work an experimental set-up is described which has been built in order to measure the rate constants of the reaction of organic compounds with the OH radical in the gas phase using a relative rate technique at atmospheric pressure. The system may be called a "mini smog chamber" as the reaction cell has a volume of only 480 cm3. The system was validated by measuring the relative rate constants of the reactant pair cyclohexane/toluene. These measurements have been performed for temperatures ranging from 300 to 360 K and led to results which are in excellent agreement with literature data. However, the experimental set-up also had some important limitations. Biphenyl which has a boiling point of 529 K was the compound with the lowest volatility that could be examined in our reaction cell. Compounds with lower vapor pressures or compounds with polar groups adsorbed to a large extent onto the glass walls of the reaction cell even at temperatures of up to 373 K. Additional problems occurred in the closed ion source of the mass spectrometer leading to non linear and/or unstable MS signals. These phenomena were most probably caused by secondary ion-molecule reactions in the closed ion source.

Assessment of microbial community changes and limiting factors during bioremediation of hydrocarbon-polluted soil with new miniaturized physiological methods

Due to human activities, organic pollutants are spilled to the environment where they threaten public health, often as contaminants of soil or groundwater. Living organisms are able to transform or mineralize many organic pollutants, and bioremediation techniques have been developed to remove pollutants from a contaminated site. However, fast and easy methods to document both the efficacy of bioremediation and the changes in soil microbial communities during bioremediation are not well developed. The major aim of this thesis was to develop miniaturized methods targeting the physiology of microbial communities during pollutant degradation in soil and to assess pollution-induced community changes. Furthermore, the methods should identify factors limiting efficient pollutant biodegradation. Petroleum hydrocarbons have been chosen as pollutants because these compounds are frequently spilled, readily water-soluble, partly toxic and carcinogenic, and therefore undesirable in soils and drinking water supplies. The influence of petroleum hydrocarbons on a microbial community in the vadose zone was assessed under field conditions. An artificial hydrocarbon mixture consisting of volatile and semi-volatile compounds similar to jet-fuel was emplaced in a previously uncontaminated vadose zone in nutrient-poor glacial melt water sand. The experiment included monitoring of microbial parameters and CO2 concentrations in soil gas over 3 months, in- and outside the hydrocarbon vapor plume that formed around the buried petroleum. Microbial and chemical analyses of vadose zone samples were performed on material from 9 cores drilled on 3 days at 3 distances from the buried petroleum mass to 3.3 m depth. Significantly elevated CO2 concentrations were observed after contamination. Total cell numbers as determined by fluorescence microscopy were strongly correlated with soil organic carbon and nitrogen content but varied little with contamination. Redundancy analysis (RDA) allowed direct analysis of effects of selected environmental variables or the artificial contamination on microbiological parameters. Variation in biomass and CO2 production was to 46 % explained by soil parameters and to 39.8 % by the duration of contamination. The microbial community structure was assessed by community-level physiological profile analysis using BiologTM EcoPlates. Only 35.9 % of variation in BiologTM data could be attributed to soil parameters and contamination, however, the samples with greatest exposure to hydrocarbons grouped together on RDA plots. It is concluded that at this nutrient-poor site, the microbial community was dominated by the natural heterogeneity and that the influence of petroleum hydrocarbon vapor was weak. BiologTM Eco-Plates combined with RDA were able to distinguish between samples with high and low hydrocarbon exposure. However, the method was not sensitive enough to produce consistent patterns in three replicates when extracts from subsurface soils with low cell numbers were inoculated. A more sensitive, MPN-based system was therefore developed and its ability to provide information about specific pollutant degraders was tested in alluvial sand. Eight dilutions of sand extracts were inoculated on medium- and substrate-containing polypropylene deep-well plates. To keep the concentrations of the volatile substrates constant over several days of incubation without intoxicating the soil community, a substrate-containing organic carrier phase was applied to each vial. The biochemically inert 2,2,4,4,6,8,8-heptamethylnonane (HMN) was appropriate for this purpose. Thus, the volatile substrates partition from HMN into the microorganism-containing soil water where they achieve relatively low concentrations. The sensitive watersoluble tetrazolium dye (WST-1) was added to each well after 7 days incubation to detect dehydrogenase activity. A dark yellow signal was developed for the substrates toluene, n-octane, n-dodecane, 1,2,4-trimethylbenzene and methylcyclohexane but not for isooctane, being strongest in n-alkane amended wells. With this method, more hydrocarbon degrading bacteria were detected in an alluvial sand exposed to kerosene for 72 hours compared to the community in the pristine sand. Both communities were mainly composed of n-alkane degraders. Although encouraging results were achieved, we noticed that the tetrazolium reduction was inhibited in some cases. Wells with high cell numbers, in which obvious growth occurred, did not produce a signal. In a next step, the two-liquid phase system with HMN was applied to modify a miniaturized respiration detection system using whole soil samples instead of soil extracts (Campbell et al., AEM 69, p. 3593). Soil was incubated in deep-well plates in presence of single volatile organic hydrocarbons. The carrier phase was added to the interstitial space in a bead layer on the bottom of the well (below the soil sample), and the hydrocarbons diffused into the soil-water. The soil activity was determined by means of an agar plate containing a pH indicator, which changes color as a function of the produced CO2. Physiological profiles specific to petroleum hydrocarbons in pristine and contaminated soils were assessed. The substrate concentrations, which induced highest microbial activity, as well as concentrations causing inhibition, were determined with effect-concentration curves. Community-level physiological profiles based on hydrocarbon degradation were obtained by applying multivariate analysis on the CO2 yields. The first measurement period of 0-6 hours after addition of relatively high hydrocarbon concentrations to the system yields values, which best separated soil types as well as pristine and contaminated soils. If low concentrations were applied, we obtained only significant separations after 24 hours incubation, probably after a growth step. The same micro-respiration system equipped with a 14CO2 detection plate was applied to study N and P limitation of aerobic mineralization of 14C-toluene in a soil previously exposed to petroleum. Significant nutrient limitation during short-term tests (24-48 hours) was identified and the optimized combination of nutrient addition was determined. Conclusively, miniaturized physiological methods have been proven to detect soil microbial community changes in petroleum hydrocarbon-contaminated soils. Adding an organic carrier phase to the miniaturized systems allowed assuring a volatile organic hydrocarbon supply in non-toxic concentrations in the soil water or the medium for several days. Thus simple, cost-effective systems to investigate the soil community during bioremediation and to address questions of nutrient shortage in soils have been provided with this thesis.

Developing Reactions of Strained Rings

Carbo- and heterocyclic structures containing nitrogen-substituted stereocenters are recurrent structural motifs in natural and bioactive compounds, therefore constituting a privileged class of synthetic targets. In this regard, substituted three- and four-membered carbocycles are powerful building blocks. Activation by a catalyst allows for an efficient exploitation of their ring strain, enabling a large variety of transformations. In particular, activation via oxidative addition by transition metals has been widely applied in annulations of these rings. Another common mode of activation consists in the use of Lewis acids to promote heterolytic ring opening of electron-poor strained rings, among which donor-acceptor cyclopropanes and cyclobutanes stand out for their well-established and broad chemistry. In this thesis, intermolecular annulations of three-membered carbocycles via oxidative addition by transition metals were first investigated. We mainly focused on the use of aminocyclopropanes as well as more strained aminomethylidenecyclopropanes. Despite a broad screening of transition metal complexes and modifications of the cyclopropane structure, no desired product could be formed. The use of aminomethylidene cyclopropanes in Lewis acid catalyzed intermolecular annulations was also studied. Employing aldehydes as reaction partners delivered highly functionalized 2-aminotetrahydrofurans. Instead, the use of all-carbon dipolarophiles for the synthesis of carbocycles was not successful. Subsequently, we explored enantioselective transformations of donor-acceptor cyclopropanes. We first investigated a new Lewis acid catalyzed DYKAT of donor-acceptor aminocyclopropanes with 2-siloxyfurans as dipolarophiles. After a thorough optimization, the desired product could be obtained in high er. However, the yield was not satisfactory, and any attempt to improve it by modifying the structure of the aminocyclopropane or by screening additives and reaction conditions was unsuccessful. On the other hand, we succeeded in the development of the first Lewis Acid catalyzed enantioselective desymmetrization of donor-acceptor meso-diaminocyclopropanes. The methodology consists in a copper(II) catalyzed Friedel-Crafts alkylation of indoles and a pyrrole, and delivers enantioenriched urea derivatives, which are recurrent structures in natural and bioactive compounds. The reaction tolerates electronically and sterically diverse substituents on the benzene ring of the indole. The choice of appropriate protecting groups on the cyclopropane was required to guarantee its solubility and to maintain good levels of enantioselectivity. The modification of an underexplored subclass of bisoxazoline ligands, bearing bulky diarylmethanol groups in alpha to the nitrogen atoms, was essential for obtaining high enantiomeric ratios, and led to the synthesis of several novel ligands. Finally, we developed Lewis acid catalyzed [4+2] annulations of donor-acceptor aminocyclobutanes with aromatic aldehydes, which afforded useful 2-aminotetrahydropyrans. A scandium catalyst was employed for the annulation of unsubstituted aminocyclobutanes, which delivered products in yields up to 95% and diastereoselectivities up to 17:1. On the other hand, one equivalent of a cheap and non-toxic iron Lewis acid was required for substituted aminocyclobutanes, which afforded densely functionalized six-membered heterocycles bearing three stereocenters in diastereoselectivities up to 5:1.
