Rescue of a severe mouse model for spinal muscular atrophy by U7 snRNA-mediated splicing modulation

In spinal muscular atrophy (SMA), the leading genetic cause of early childhood death, the survival motor neuron 1 gene (SMN1) is deleted or inactivated. The nearly identical SMN2 gene has a silent mutation that impairs the utilization of exon 7 and the production of functional protein. It has been hypothesized that therapies boosting SMN2 exon 7 inclusion might prevent or cure SMA. Exon 7 inclusion can be stimulated in cell culture by oligonucleotides or intracellularly expressed RNAs, but evidence for an in vivo improvement of SMA symptoms is lacking. Here, we unambiguously confirm the above hypothesis by showing that a bifunctional U7 snRNA that stimulates exon 7 inclusion, when introduced by germline transgenesis, can efficiently complement the most severe mouse SMA model. These results are significant for the development of a somatic SMA therapy, but may also provide new means to study pathophysiological aspects of this devastating disease.

Gyrokinetic global three-dimensional simulations of linear ion-temperature-gradient modes in Wendelstein 7-X

Using a global approach for solving an ion gyrokinetic model in three-dimensional geometry the linear stability and structure of ion-temperature-gradient (ITG) modes in the configuration of the stellarator Wendelstein 7-X (W7-X) [G. Grieger , in Plasma Physics and Controlled Nuclear Fusion Research 1990 (International Atomic Energy Agency, Vienna, 1991), Vol. 3, p. 525.] is studied. The time evolution of electrostatic perturbations is solved as an initial value problem with a particle-in-cell deltaf method. The vacuum magnetohydrodynamic equilibrium is calculated by the code VMEC [S. P. Hirshman and D. K. Lee, Comput. Phys. Commun. 39, 161 (1986)]. In this work the most unstable ITG mode in W7-X is presented. This mode has a pronounced ballooning-type structure; however, it is not tokamak-like. A driving mechanism analysis using the energy transfer shows that the contribution of curvature effects is non-negligible. The growth rate and the mixing-length estimate for transport are compared with those for ITG modes found in axisymmetric geometries. (C) 2004 American Institute of Physics.

Step training reinforces specific spinal locomotor circuitry in adult spinal rats

Locomotor training improves function after a spinal cord injury both in experimental and clinical settings. The activity-dependent mechanisms underlying such improvement, however, are sparsely understood. Adult rats received a complete spinal cord transection (T9), and epidural stimulation (ES) electrodes were secured to the dura matter at L2. EMG electrodes were implanted bilaterally in selected muscles. Using a servo-controlled body weight support system for bipedal stepping, five rats were trained 7 d/week for 6 weeks (30 min/d) under quipazine (0.3 mg/kg) and ES (L2; 40 Hz). Nontrained rats were handled as trained rats but did not receive quipazine or ES. At the end of the experiment, a subset of rats was used for c-fos immunohistochemistry. Three trained and three nontrained rats stepped for 1 h (ES; no quipazine) and were returned to their cages for 1 h before intracardiac perfusion. All rats could step with ES and quipazine administration. The trained rats had higher and longer steps, narrower base of support at stance, and lower variability in EMG parameters than nontrained rats, and these properties approached that of noninjured controls. After 1 h of stepping, the number of FOS+ neurons was significantly lower in trained than nontrained rats throughout the extent of the lumbosacral segments. These results suggest that training reinforces the efficacy of specific sensorimotor pathways, resulting in a more selective and stable network of neurons that controls locomotion.
