Christian Depeursinge

Christian Depeursinge is the leader of the Microvision and Micro-Diagnostics (MVD) group at the Advanced Photonics Laboratory of the Institute of Microengineering at EPFL (Ecole Polytechnique Fédérale de Lausanne), Switzerland (http://apl.epfl.ch/muvision). His research and expertise in biomedical engineering and optics is internationally acknowledged. His current research topics include coherent and incoherent Imaging applied to diagnostics in biology, His research group pioneered in the development of DHM technology. He worked on several projects developed in cooperation with European and international partners. He is author and co-author of over 100 papers published in peer reviewed journals, several book chapters and more than 30 patents. He has given more than 20 invited lectures and plenaries in the last five years. He developed many projects in cooperation with national and international industries. He is co-founder of a start-up company (Lyncée Tec SA: www.Lynceetec.com). He is currently teaching at EPFL and occasionally in foreign universities and institutes.

Simultaneous dual-contrast three-dimensional imaging in live cells via optical diffraction tomography and fluorescence

We report a dual-contrast method of simultaneously measuring and visualizing the volumetric structural information in live biological samples in three-dimensional (3D) space. By introducing a direct way of deriving the 3D scattering potential of the object from the synthesized angular spectra, we obtain the quantitative subcellular morphology in refractive indices (RIs) side-by-side with its fluorescence signals. The additional contrast in RI complements the fluorescent signal, providing additional information of the targeted zones. The simultaneous dual-contrast 3D mechanism unveiled interesting information inaccessible with previous methods, as we demonstrated in the human immune cell (T cell) experiment. Further validation has been demonstrated using a Monte Carlo model. (C) 2019 Chinese Laser Press.

Quantitative phase imaging in biomedicine

Quantitative phase imaging (QPI) has emerged as a valuable method for investigating cells and tissues. QPI operates on unlabelled specimens and, as such, is complementary to established fluorescence microscopy, exhibiting lower phototoxicity and no photobleaching. As the images represent quantitative maps of optical path length delays introduced by the specimen, QPI provides an objective measure of morphology and dynamics, free of variability due to contrast agents. Owing to the tremendous progress witnessed especially in the past 10-15 years, a number of technologies have become sufficiently reliable and translated to biomedical laboratories. Commercialization efforts are under way and, as a result, the QPI field is now transitioning from a technology-development-driven to an application-focused field. In this Review, we aim to provide a critical and objective overview of this dynamic research field by presenting the scientific context, main principles of operation and current biomedical applications.

Quantitative phase-digital holographic microscopy: a new imaging modality to identify original cellular biomarkers of diseases

Quantitative phase microscopy (QPM) has recently emerged as a powerful label-free technique in the field of living cell imaging allowing to non-invasively measure with a nanometric axial sensitivity cell structure and dynamics. Since the phase retardation of a light wave when transmitted through the observed cells, namely the quantitative phase signal (QPS), is sensitive to both cellular thickness and intracellular refractive index related to the cellular content, its accurate analysis allows to derive various cell parameters and monitor specific cell processes, which are very likely to identify new cell biomarkers. Specifically, quantitative phase-digital holographic microscopy (QP-DHM), thanks to its numerical flexibility facilitating parallelization and automation processes, represents an appealing imaging modality to both identify original cellular biomarkers of diseases as well to explore the underlying pathophysiological processes.
