Georges Wagnières received his diploma degree (MSc) in physics from the University of Lausanne, Switzerland, in 1986. He obtained his doctorate in science (PhD) in physics (Biomedical optics) from the Swiss Federal Institute of Technology at Lausanne (EPFL) in 1992 and did a postdoctoral work in the Wellman Laboratories of Photomedicine (Harvard Medical School), Boston, USA, from 1993 to 1994. He also obtained a Master degree in Management of Technology delivered by the Ecole des Hautes Etudes Commerciales (HEC) of Lausanne University and the EPFL in 2001. Georges Wagnières manages a research group active in the fields of: - Detection of early superficial cancers by fluorescence imaging. - Characterization of early superficial cancers by high magnification narrow band imaging. - In vivo and in vitro measurement of the vascular and tissular oxygen concentration by time-resolved luminescence spectroscopy and imaging. - Preclinical and clinical study of new photosensitizers for photodynamic therapy (PDT). - Treatment of neurodegenerative conditions (Alzheimer, Parkinson's diseases) by photobiomodulation. - Preclinical and clinical photodynamic therapy of inflammatory bowel diseases and atheroscerotic plaque. - Improvement of the selective vascular extravasation of chemotherapeutic agents by PDT. - Monitoring the light dosimetry during PDT by fluorescence spectroscopy and imaging. - Monitoring laser treatments of the retina by reflectance imaging. - Light dosimetry and tissue optical spectroscopy. - Radiometry. - Development of light delivery systems for biomedical applications. Georges Wagnières is also co-founder and was chairman of one spin-off companies: - Medlight SA, founded in July 1997, which develops, produces and commercializes light distributors for photodynamic therapy. Georges Wagnières has currently authored more than 235 papers (more than 150 in international journals with review board) and is inventor of 18 patents. He supervised 12 PhD students up to now, and currently teaches biomedical optics and photomedicine in master programs and doctoral schools. In addition, he gives the course entitled "Physique Générale I" to biology first year students registered to the Biology School of the Lausanne's University. Porphyrin metalla-assemblies coupled to cellulose nanocrystals for PDT and imaging applications

Photodynamic therapy (PDT) is an interesting and promising approach to tackle a broad spectrum of cancer. With the combination of a photosensitizer, light and oxygen, PDT achieves a unique selectivity by the production of localized reactive oxygen species (ROS) inside cells, which leads to their destruction. In addition, the luminescence properties of photosensitizers can be exploited to develop imaging tools. Unfortunately, the cancer selectivity and homogeneity of most photosensitizers are frequently limiting the performances of PDT and cancer detection/characterization by luminescence imaging. Consequently, our study aims to use cellulose nanocrystals to transport and deliver radiolabeled photo-responsive metalla-assemblies to create a new generation of theranostic agents for PDT and imaging applications. The synthesis, structural characterization, cytotoxicity evaluation, and in vivo biodistribution imaging of the compounds are presented. The best candidates show excellent biological activity and selectivity towards ovarian carcinoma cell line (A2780), cisplatin-resistant ovarian carcinoma cell line (A2780cis) versus normal human embryonic kidney cells (HEK293T), as well as efficient imaging properties, suggesting a potential use as multimodal theranostic agents.

Optimization of the Distance between Cylindrical Light Distributors Used for Interstitial Light Delivery in Biological Tissues

Cylindrical light diffusers (CLDs) are often employed for the treatment of large tumors by interstitial photodynamic therapy (iPDT) and photoimmunotherapy (PIT), which involves careful treatment planning to maximize therapeutic dose coverage while minimizing the number of CLDs used. There is, however, a lack of general guidelines regarding optimal positioning of CLDs, in particular when they are inserted in parallel to treat head and neck squamous cell cancer (HNSCC). Therefore, the purpose of this study is to determine the CLD-CLD distances maximizing the necrosis for different geometries of CLD positions and shed light on the influence of different optical parameters on this distance, in particular when HNSCCs are treated by interstitial PIT with cetuximab-IR700 using up to seven CLDs. To that end, Monte-Carlo simulations of the light propagation around CLDs inserted perpendicularly in a semi-infinite tumor were performed to determine the volume receiving a fluence larger than a therapeutic threshold. An optimization algorithm was then used to calculate and maximize the necrosed tumor volumes. Tumor optical properties were derived from published data. Our findings suggest that optimal CLD positioning maximizing the volume of necrosed tumor during interstitial PIT for typical HNSCC optical properties corresponds to a CLD-CLD distance between 11.5- and 13-mm. Variations of the absorption and reduced scattering coefficients have the greatest influence on CLD placements, while tissue anisotropy factor, CLD insertion geometry, CLD length, and the angular dependence of the radiance emitted by the CLDs have minimal influence. At first approximation the influence of these optical parameters on optimal CLD-CLD distance are independent. Our data also suggests it is possible to derive new treatment plans using knowledge of previous treatment plans.

Sodium thiosulfate, a source of hydrogen sulfide, stimulates endothelial cell proliferation and neovascularization

Therapies to accelerate vascular repair are currently lacking. Pre-clinical studies suggest that hydrogen sulfide (H2S), an endogenous gasotransmitter, promotes angiogenesis. Here, we hypothesized that sodium thiosulfate (STS), a clinically relevant source of H2S, would stimulate angiogenesis and vascular repair. STS stimulated neovascularization in WT and LDLR receptor knockout mice following hindlimb ischemia as evidenced by increased leg perfusion assessed by laser Doppler imaging, and capillary density in the gastrocnemius muscle. STS also promoted VEGF-dependent angiogenesis in matrigel plugs in vivo and in the chorioallantoic membrane of chick embryos. In vitro, STS and NaHS stimulated human umbilical vein endothelial cell (HUVEC) migration and proliferation. Seahorse experiments further revealed that STS inhibited mitochondrial respiration and promoted glycolysis in HUVEC. The effect of STS on migration and proliferation was glycolysis-dependent. STS probably acts through metabolic reprogramming of endothelial cells toward a more proliferative glycolytic state. These findings may hold broad clinical implications for patients suffering from vascular occlusive diseases.
