Caroline Vandevyver

Caroline Vandevyver received her degree in biochemistry in 1985 from the Katholieke Universiteit Leuven, Belgium and her Ph.D. degree in biotechnology at the same university in 1989.

She joined the Biomedical Research Institute "Dr. L- Willems Instituut" in Hasselt, Belgium, in 1989. Subsequently, she has worked there on T cell mediated autoimmune diseases (Multiple Sclerosis, Rheumatoid Arthritis), genetic association and linkage studies in multifactorial diseases, mutation analysis of genetic disorders (Phenylketonuria, alpha 1 anti-trypsine deficiency, congenital adrenal hyperplasia) and the development of active and passive immune therapies for cancer.

She joined EPFL in 2000 at the Laboratory of Chemical Biotechnology headed by Prof. Ruth Freitag, where she was responsible for the cell culture lab and the (recombinant) protein purification and characterization. She joined the Laboratory of Lanthanide Supramolecular Chemistry (LCSL) in May 2006, where she was responsible for the live cell imaging project with Lanthanide complexes.

In 2005, she joined the Office of the Research Commission. Her duties there were the management of national and international fellowship programs, the management of local research awards and the promotion of research performed at EPFL. In August 2013, she was nominated coordinator of the International Funding, one of the pillars of the Research Office at EPFL. Since January 2017, she is the Head of EPFL's Research Office.

Lanthanide bioprobes and bioconjugates are ideal luminescent stains in view of their low propensity to photobleaching, sharp emission lines and long excited state lifetimes permitting time-resolved detection for enhanced sensitivity. In this paper, we expand our previous work which demonstrated that self-assembled dinuclear triple-stranded helicates[Ln(2)(L-C2X)(3)] behave as excellent cell and tissue labels in immunocytochemical and immunohistochemical assays. The synthetic strategy of the hexadentate ditopic ligands incorporating dipicolinic acid, benzimidazole units and polyoxyethylene pendants is revisited in order to provide a more straightforward route and to give access to further functionalization of the polyoxyethylene arms by incorporating a terminal function X. Formation of the helicates [Ln(2)(L-C2X)(3)] (X= COOH, CH2OH, COEt, NH2, phthalimide) is ascertained by several experimental techniques and their stability tested against diethylenetriaminepentaacetate. Their photophysical properties (quantum yield, lifetime, radiative lifetime and sensitization efficiency) are presented and compared with those of the parent helicates [Ln(2)(L-C2)(3)]. Finally, the cellular uptake of five Eu-III helicates is monitored by time-resolved luminescence microscopy and their localization in HeLa cells established by co-staining experiments.

Fluorescent magnetic bead and cell differentiation/counting using a CMOS SPAD matrix

We present a monolithic silicon chip comprising a matrix of 84 single photon avalanche diodes (SPADs) to detect and discriminate fluorescent beads or fluorescently labeled single cells in a polydimethyl(-siloxane) (PDMS) cartridge that is positioned on top of the chip. Our detection is based on the different photon count when either a fluorescent or non-fluorescent bead or cell is present above a SPAD, due to the additional photons emitted from a fluorescent object. Our technique allows microscope-less fluorescence detection and permits easy exchange of the disposable microfluidic cartridge. We first demonstrate the working principle of our device by counting and discriminating fluorescent from non-fluorescent 3, 6 and 10 μm magnetic beads, which are commonly used as versatile mobile carriers for separating a target analyte from a matrix via magnetic forces in microfluidic lab-on-a-chip systems. We then apply our system to count and discriminate fluorescently-labeled MCF-7 breast cancer cells from unlabeled Jurkat cells mixed in a phosphate buffer saline (PBS) solution. Our device is robust and does not need complex microfluidic handling to achieve cell count without the need of external fluorescence detection bulky equipment.

Chaotic mixing using source-sink microfluidic flows in a PDMS chip

We present an active fixed-volume mixer based on the creation of multiple source–sink microfluidic flows in a polydimethylsiloxane (PDMS) chip without the need of external or internal pumps. To do so, four different pressure-controlled actuation chambers are arranged on top of the 5 μl volume of the mixing chamber. After the mixing volume is sealed/fixed by microfluidic valves made using ‘microplumbing technology’, a virtual source–sink pair is created by pressurizing one of the membranes and, at the same time, releasing the pressure of a neighboring one. The pressurized air deforms the thin membrane between the mixing and control chambers and creates microfluidic flows from the squeezed region (source) to the released region (sink) where the PDMS membrane is turned into the initial state. Several schemes of operation of virtual source–sink pairs are studied. In the optimized protocol, mixing is realized in just a sub-second time interval, thanks to the implementation of chaotic advection.
