Specific SIRT1 activation mimics low energy levels and protects against diet-induced metabolic disorders by enhancing fat oxidation

The NAD(+)-dependent deacetylase SIRT1 controls metabolic processes in response to low nutrient availability. We report the metabolic phenotype of mice treated with SRT1720, a specific and potent synthetic activator of SIRT1 that is devoid of direct action on AMPK. SRT1720 administration robustly enhances endurance running performance and strongly protects from diet-induced obesity and insulin resistance by enhancing oxidative metabolism in skeletal muscle, liver, and brown adipose tissue. These metabolic effects of SRT1720 are mediated by the induction of a genetic network controlling fatty acid oxidation through a multifaceted mechanism that involves the direct deacetylation of PGC-1alpha, FOXO1, and p53 and the indirect stimulation of AMPK signaling through a global metabolic adaptation mimicking low energy levels. Combined with our previous work on resveratrol, the current study further validates SIRT1 as a target for the treatment of metabolic disorders and characterizes the mechanisms underlying the therapeutic potential of SIRT1 activation.

Role of the bile acid membrane receptor TGR5 in macrophages and metabolism

Activation of the bile acid responsive G protein-coupled receptor TGR5 improves metabolic homeostasis via induction of energy expenditure and release of glucagon-like peptide-1 (GLP-1). The goal of my PhD work was to explore the role of macrophage TGR5 with respect to inflammation and to evaluate its impact on chronic low-grade inflammatory and metabolic disorders, such as atherosclerosis and insulin resistance. In the first part of this work, we have focused on the role of TGR5 in the progression of atherosclerotic lesions. I showed that activation of TGR5 in macrophages by 6"-ethyl-23(S)-methyl-cholic acid (S-EMCA, INT-777), a semi-synthetic bile acid, inhibits pro-inflammatory responses via a cAMP-NF-!# pathway. Furthermore, INT-777 treatment of chimeric Ldlr knockout mice that were transplanted with bone marrow from either Tgr5 wild-type or Tgr5 knockout mice, revealed that TGR5 in leukocytes largely contribute to the protective effects on atherosclerotic lesion formation. In the second part of this work, I have determined the function of TGR5 in macrophage polarization and evaluated the relative contribution of macrophage TGR5 in the onset of diet-induced insulin resistance. I demonstrated that deficiency of TGR5 results in exacerbated insulin resistance in diet-induced obesity (DIO) mice with severe impairment of insulin responsiveness in adipose tissue. This was associated with an increase in M1 macrophages and a reduction of M2 macrophages in adipose tissue of Tgr5 knockout mice. In line with this, TGR5 activation by INT-777 in DIO mice increased M2 macrophages, attenuated adipose tissue inflammation and improved insulin resistance. Taken together, the research that is described in this thesis reveals a significant immune modulatory function of TGR5, which has a major impact on the prevention of atherosclerosis and insulin resistance in mouse models. Future studies in humans will be required to underscore the importance of TGR5 as a potential target to counteract the chronic low-grade inflammation that goes hand-in hand with many disorders of the metabolic syndrome.

Growth differentiation factor 15 is a myomitokine governing systemic energy homeostasis

Reduced mitochondria! electron transport chain activity promotes longevity and improves energy homeostasis via cell-autonomous and non-autonomous factors in multiple model systems. This mitohormetic effect is thought to involve the mitochondrial unfolded protein response (UPRmt), an adaptive stress-response pathway activated by mitochondria' proteotoxic stress. Using mice with skeletal muscle specific deficiency of Crifl (muscle-specific knockout [MKO]), an integral protein of the large mitoribosomal subunit (39S), we identified growth differentiation factor 15 (GDF15) as a UPRmt-associated cell non-autonomous myomitokine that regulates systemic energy homeostasis. MKO mice were protected against obesity and sensitized to insulin, an effect associated with elevated GDF15 secretion after UPRmt activation. In ob/ob mice, administration of recombinant GDF15 decreased body weight and improved insulin sensitivity, which was attributed to elevated oxidative metabolism and lipid mobilization in the liver, muscle, and adipose tissue. Thus, GDF15 is a potent mitohormetic signal that safeguards against the onset of obesity and insulin resistance.

Differential roles of GDF15 and FGF21 in systemic metabolic adaptation to the mitochondrial integrated stress response

Perturbation of mitochondrial proteostasis provokes cell autonomous and cell non-autonomous responses that contribute to homeostatic adaptation. Here, we demonstrate distinct metabolic effects of hepatic metabokines as cell non-autonomous factors in mice with mitochondrial OxPhos dysfunction. Liver-specific mitochondrial stress induced by a loss-of-function mutation in Crif1 (LKO) leads to aberrant oxidative phosphorylation and promotes the mitochondrial unfolded protein response. LKO mice are highly insulin sensitive and resistant to diet-induced obesity. The hepatocytes of LKO mice secrete large quantities of metabokines, including GDF15 and FGF21, which confer metabolic benefits. We evaluated the metabolic phenotypes of LKO mice with global deficiency of GDF15 or FGF21 and show that GDF15 regulates body and fat mass and prevents diet-induced hepatic steatosis, whereas FGF21 upregulates insulin sensitivity, energy expenditure, and thermogenesis in white adipose tissue. This study reveals that the mitochondrial integrated stress response (ISRmt) in liver mediates metabolic adaptation through hepatic metabokines.
