Optimization of in vitro cord blood hematopoietic stem cell expansion through the identification of secreted factors mediating inter-cellular communication

Robust and large-scale expansion of umbilical cord blood stem cells in vitro is necessary for widening the usage of transplantation therapies for the treatment of hematological and immune diseases. The lack of understanding of the complex inter-cellular networks regulating stem cell fate in culture explains the low success met so far for the ex vivo expansion of hematopoietic stem cells. The development of a mathematical model of in vitro hematopoiesis coupled with gene expression profiling led to predictions about the secreted factors that play a crucial role in regulating hematopoietic stem cell self-renewal in culture. We tested 18 putative molecules predicted to display effects on primitive progenitor (Long-Term Culture-Initiating Cell; LTC-IC) output, functionally validating three stimulators (VEGF, PDGF, EGF) and three inhibitors (TGFβ, CCL4, CXCL10). Combinatorial studies with the stimulatory molecules showed less-than additive effects, perhaps related to redundant signaling mechanisms. Small molecule-mediated inhibition of the downstream signaling pathways activated by VEGF, PDGF, and EGF led to a decreased expansion of primitive cell compartments, confirming the endogenous activity of these growth factors for the stimulation of blood stem and progenitor cells. Blocking TGFβ-mediated negative feedback signaling in culture enhanced mature cell outputs but had no effect on primitive cell expansion, consistent with the role of TGFβ as a proliferation inhibitor, and underlying the complexity and multi-parametric aspect of the inter-cellular regulatory network. Studies are currently underway to validate the functional activity of VEGF and EGF on in vivo repopulating stem cells (NOD/SCID repopulating cells; SRC) using a clinical-grade, closed-system bioprocess. These results constitute a major step in the functional elucidation of the complex extracellular signaling networks that govern stem cell fate in vitro

The Cell Differentiation Status Influences the Outcome of Notch-Induced Malignancy

The Notch signaling cascade is a well conserved pathway that regulates many aspects of embryonic development and plays a crucial role in the differentiation process and tissue homeostasis in multiple organs of adult species. Several human pathological disorders, including cancer, arise as a consequence of the deregulation of the Notch signaling pathway. Notch deregulation, up to date, has been best characterized in acute T-cell lymphoblastic leukemia (T-ALL). Indeed, in T-ALL pediatric patients activating Notch1 mutations have been identified in more than 50% of the cases. Increased knowledge of the mechanisms underlying T-ALL development and maintenance is clearly needed as one out of 5 patients with T-ALL has a very poor prognosis. Moreover, the high toxicity of chemotherapeutic regimens has been associated with elevated risks of developing severe secondary health problems. In the present study, we characterized the role of Notch in T-ALL induction and maintenance. Using transgenic mouse models, we could demonstrate that the oncogenic potential of Notch is dependent on RBP-Jκ transcriptional activity. Indeed, the canonical Notch signaling pathway, mediated by the RBP-Jκ transcription factor, is essential for both Notch-mediated T-ALL induction and maintenance, as well as for the induction of a Notch-mediated lymphoproliferative disorder. Moreover, constitutive overexpression of Notch1 at discrete developmental stages within the hematopoietic system, led us to identify a correlation between the differentiation status of a cell, within the hierarchy of the hematopoietic tree, and its sensitivity to Notch1-induced transformation. We demonstrated that forced Notch1 expression in hematopoietic stem cells was not sufficient to induce T-ALL. Nevertheless, aberrant Notch1 expression in the bone marrow resulted in the development of a lethal lymphoproliferative disorder. Overexpression of Notch1 in thmus at the DN2 to DN3 stage of T-cell delvelopment generated an aggressive transplantable T-ALL with important metastatic potential. This suggested that progenitors already committed to the T-cell lineage are highly susceptible to Notch-mediated transformation, which is relevant to the human disease. Nevertheless, forced Notch1 expression after the β-selection checkpoint in the thymys, was no longer sufficient to induce transformation of late DN3 to DN4 thymocytes. We therefore established a murine genetic system that allowed us to dissect the mechanisms of aberrant Notch1 signaling in a lymphoproliferative disorder and a true T-ALL model. We were able to assess the genetic signature that is involved in the transplantability and metastatic potential of leukemic cells and generated a list of candidate genes implicated in these processes. In addition, we correlated genes identified in our murine models with genes associated as well in human T-ALL profiles to identify genes implicated in T-ALL pathogenesis in both mouse and human.

The Intestinal Epithelium

The mammalian intestine is a prototype of a self-renewing organ. The rapid cellular turnover is supported by proliferating progenitors located in the intestinal crypt, which is also the stem cell location. Homeostasis of the intestinal epithelium has to be under stringent control to ensure lifelong self-renewal. Processes like proliferation and differentiation (often deregulated in cancer) seem to be controlled by a relative small number of signaling pathways, including Wnt and Notch. Notch signaling is playing a crucial role in the maintenance of the intestinal homeostasis by controlling progenitors proliferation and their cell fate. In this thesis project we studied Notch signaling and its role in the murine intestine focusing on 1) Its downstream mediators; 2) The identification of the physiological Notch ligands; 3) Its role on the stem cell compartment, and 4) the potential cross talk between Notch and Wnt. Furthermore, 5) we investigated the role of Notch signaling in Wnt induced intestinal tumorigenesis. This study evidenced that Hes1 is not the unique Notch signaling mediator in the intestinal epithelium. Moreover the transcription factor Klf4 is not required for goblet cell differentiation. The in vivo role of the Notch ligands Dll1, Dll4 and Jag1 was assessed deriving inducible intestine specific single and double gene-targeted mice. Inactivation of individual ligands did not show any loss of Notch phenotype with the exception of Dll1 mutant mice. Only by deriving double mutants we identified Dll1 and Dll4 as the physiological Notch ligands in the intestine. We provided evidence for the first time that Notch1 signaling is active in intestinal stem cell. Furthermore we demonstrated that Notch signaling is required for the maintenance of intestinal stem cells, as well as for the transit-amplifying progenitor compartment. Using different genetic gain and loss-off function approaches we investigated the relation between the Notch and Wnt cascade for the maintenance of the proliferative crypt compartment. A synergistic relation was observed between Notch and Wnt in intestinal tumorigenesis. Collectively our data highlight Notch signaling as a key player for the maintenance of intestinal homeostasis.

Inhibition of Fibroblast Growth by Notch1 Signaling Is Mediated by Induction of Wnt11-Dependent WISP-1

Fibroblasts are an integral component of stroma and important source of growth factors and extracellular matrix (ECM). They play a prominent role in maintaining tissue homeostasis and in wound healing and tumor growth. Notch signaling regulates biological function in a variety of cells. To elucidate the physiological function of Notch signaling in fibroblasts, we ablated Notch1 in mouse (Notch1(Flox/Flox)) embryonic fibroblasts (MEFs). Notch1-deficient (Notch1(-/-)) MEFs displayed faster growth and motility rate compared to Notch1(Flox/Flox) MEFs. Such phenotypic changes, however, were reversible by reconstitution of Notch1 activation via overexpression of the intracellular domain of Notch1 (NICD1) in Notch1-deficient MEFs. In contrast, constitutive activation of Notch1 signaling by introducing NICD1 into primary human dermal fibroblasts (FF2441), which caused pan-Notch activation, inhibited cell growth and motility, whereas cellular inhibition was relievable when the Notch activation was countered with dominant-negative mutant of Master-mind like 1 (DN-MAML-1). Functionally, "Notch-activated" stromal fibroblasts could inhibit tumor cell growth/invasion. Moreover, Notch activation induced expression of Wnt-induced secreted proteins-1 (WISP-1/CCN4) in FF2441 cells while deletion of Notch1 in MEFs resulted in an opposite effect. Notably, WISP-1 suppressed fibroblast proliferation, and was responsible for mediating Notch1's inhibitory effect since siRNA-mediated blockade of WISP-1 expression could relieve cell growth inhibition. Notch1-induced WISP-1 expression appeared to be Wnt11-dependent, but Wnt1-independent. Blockade of Wnt11 expression resulted in decreased WISP-1 expression and liberated Notch-induced cell growth inhibition. These findings indicated that inhibition of fibroblast proliferation by Notch pathway activation is mediated, at least in part, through regulating Wnt1-independent, but Wnt11-dependent WISP-1 expression.
