Preparation and characterization of Poly(Ethylene Glycol)/gamma-CD Polyrotaxanes using beta-CD derivatives as a capping reagent

As living systems understanding got better, as life could be modeled in a batch, using immortalized cells, researches in fundamental sciences found startling applications of advanced techniques on living systems. As the biomaterial field developed various molecules were found to have very interesting applications on living systems. Polyrotaxane is one of these particularly interesting molecules. It is a non covalently bonded molecular necklace produced by the threading of cyclic molecules onto a linear polymer in order to form a pseudopolyrotaxane, the capping of this pseudopolyrotaxane to avoid de-threading of the cyclic compounds forms a polyrotaxane. This compound has already proven its versatility through various application including drug delivery systems, DNA delivery agent, molecular machine, hydrogel building block, resin stabilizer and a lot more. Cyclodextrins (CDs) and Polyester are known to form a pseudopolyrotaxane. CDs are composed of 6 to 8 glucopyranose forming a toroid shaped molecule that can contain different molecules depending on the solvent, experimental conditions and the number of sugar subunits. In this project an attempt has been made to synthesize a special type of polyrotaxane that would allow the whole complex to bare a higher degree of freedom compared to the ones currently documented. In order to do so the widest CD has been used (Gamma-CD, GCD), the chosen host has been poly(ethylene glycol) (PEG) chains of varying molecular weight and the capping reagent used was beta-CD derivatives. Furthermore since current literature hasn't documented it so well and its properties are essential to this study, some more characterization of the PEG/GCD pseudopolyrotaxane has been made. The Macromolecules have been characterized by maldi tof mass spectroscopy(MALDI-T-MS), electro spray ionization (ESI) mass detection, X-ray diffraction(XRD), gel phase chromatography (GPC) and liquid Nuclear magnetic resonance (NMR). The beta CD and poly(ethylene glycol) derivatives have been characterized using Fourier transform infra red spectroscopy (FTIR), NMR, MALDI-Toff MS, ESI and thin layer Chromatography (TLC)

Design of amphiphilic nanocapsules based on hyperbranched star-block copolymers for the encapsulation and the controlled release of olfactory compounds

Effective encapsulation of small, volatile, weakly water-soluble molecules such as flavors and fragrances is necessary to protect them from degradation, to increase their lifetime, and to improve their water dispersion and fixation depending on the substrate of interest. The aim of this project has been to improve encapsulation and release in order to optimize the performance of fragrance molecules in water-based household (detergents, softeners, etc.) and body care applications (shampoos, lotions, etc.), and fine perfumery applications. More specifically, the aim has been to investigate the effectiveness of "unimolecular micelles" based on amphiphilic multi-arm star-block copolymers with a hyperbranched core with more than 26 functional groups, a hydrophobic inner and a hydrophilic outer shell. These have been prepared from a commercial hyperbranched polyester macroinitiator (HBP) by ring-opening polymerization of ε-caprolactone, followed by the atom transfer radical polymerization (ATRP) of tert-butyl acrylate (tBuA). Hydrolysis of the tert-butyl groups has then been used to convert the poly(tBuA) blocks to poly(acrylic acid) (PAA), resulting in HBP-(PCL)p-(PAA)q pH-dependent amphiphilic star-block copolymers with good control of the molecular weight distribution. These were shown to form stable nanocapsules with a well defined core-shell architecture, as confirmed by thermal and microstructural characterization. The necessity of the core-shell architecture for the effective encapsulation of fragrance molecules in aqueous dispersion has been demonstrated by NMR (nuclear magnetic resonance). The extent of encapsulation reflects the dynamic equilibrium between the free molecules and the fragrance/polymer complex and is dependent on the octanol/water partition coefficient (logP) of the fragrance compounds, as demonstrated with a similar non ionic core-shell architecture HBP-(PBMA)37-(PPEGMA)39 composed of a hydrophobic poly(butyl methacrylate) (PBMA) core and a hydrophilic poly(polyethylene glycol) methyl ether methacrylate (PPEGMA) shell (provided by the Polymer Laboratory of the EPFL). The fragrance loadings in the polymer (HBP-(PCL)p-(PAA)q and HBP-(PBMA)37-(PPEGMA)39), which reached up to 30 wt% depending on the type of the fragrance molecule, were argued to be linked to their solubility in the hydrophobic core of the star-block copolymer. Moreover, under conditions representative of real applications (fine perfumery and softener applications) the star-block copolymers significantly extended the time over which the concentration of certain volatiles remained above the human olfactory threshold. Effective encapsulation and delayed release of small hydrophobic molecules was hence demonstrated to be possible with the present systems. The straightforward synthesis, tailorable chemistry and globular architecture of the star-block copolymers investigated here therefore offer promise for the development of relatively cheap encapsulants with affinities for specific components in fragrance packages, and release triggered by selected substrates according to their surface chemistry.

Water-soluble, unimolecular containers based on amphiphilic multiarm star block copolymers

A new class of water-soluble, amphiphilic star block copolymers with a large number of arms was prepared by sequential atom transfer radical polymerization (ATRP) of n-butyl methacrylate (BMA) and poly-(ethylene glycol) methyl ether methacrylate (PEGMA). As the macroinitiator for the ATRP, a 2-bromoisobutyric acid functionalized fourth-generation hyperbranched polyester (Boltorn H40) was used, which allowed the preparation of star polymers that contained on average 20 diblock copolymer arms. The synthetic concept was validated by AFM experiments, which allowed direct visualization of single molecules of the multiarm star block copolymers. DSC and SAXS experiments on bulk samples suggested a microphase-separated structure, in agreement with the core-shell architecture of the polymers. SAXS experiments on aqueous solutions indicated that the star block copolymers can be regarded as unimolecular micelles composed of a PBMA core and a diffuse PPEGMA corona. The ability of the polymers to encapsulate and release hydrophobic guests was evaluated using 1H NMR spectroscopy. In dilute aqueous solution, these polymers act as unimolecular containers that can be loaded with up to 27 wt % hydrophobic guest molecules. © 2006 American Chemical Society.

Cryo-AFM development for high resolution DNA imaging and spectroscopy

The thesis presents the development of a custom made atomic force microscope (AFM) conceived to image biological molecules at low temperatures in ultrahigh vacuum conditions, and its application to DNA single molecule studies. Starting from a first prototype working in contact mode, our aim was to implement non-contact AFM in order to reduce possible sample damaging due to strong tip interactions. The first chapter of the thesis discusses the technical solutions developed to improve the previous system. A complete setup for in situ exchange of samples and tips was designed and implemented. This permits a continuous operation of the microscope under the experimental conditions of UHV. Thermal instabilities and temperature differences between the tip and the sample were solved with the design of a movable shield to avoid heating by thermal radiation. The introduction of a new high voltage signal generator with variable frequency and the development of a user-friendly LabVIEW interface to communicate with this device notably improved the piezomotors driving. These piezomotors perform the micro-positioning of all the microscope parts and are driven through this interface. Finally, the previous tip-holder design was modified to incorporate a piezo actuator behind the cantilever, which provides the necessary excitation for the implementation of dynamic AFM modes. The installation of a Phase Lock Loop (PLL) circuit to lock the cantilever frequency and the replacement of old feedback electronics for a digital SPM Controller, with advanced options of frequency-shift feedback, was another requirement for the correct implementation of non-contact AFM. The second chapter deals with DNA sample preparation for high resolution imaging by AFM. Based on our studies, the advantages and drawbacks of the most commonly used techniques for DNA adsorption are discussed in detail. Our measurements in contact mode demonstrate the convenience of using hydrophobic substrates for high resolution DNA imaging. At hydrophilic surfaces, like mica, the presence of a thin water layer could be responsible of strong adhesion forces between the cantilever and the sample. In order to overcome this limitation we tested HOPG as hydrophobic substrate. HOPG, chemically modified with amphiphilic compounds, allowed us to deposit DNA without molecular damage or stress. The HOPG surface treatment was significantly improved. New protocols were developed to control the formation of dodecylamine disordered monolayers or lamella structures on the substrate. The lamella formation induces a subsequent self assembly of DNA single molecules. In the last chapter we discuss our results on DNA imaging obtained with the different operation modes implemented in our microscope: contact mode, tapping mode and frequency modulation (non-contact) AFM. We conclude that the last one provides higher signal stability and the necessary sensitivity to observe DNA details at a single-molecule level. We also present a study of frequency shift versus distance dependence (force spectroscopy experiments) performed in DNA samples. We discuss for the first time a plausible explanation to the origin of the atomic force contrast between the substrate and the DNA molecules. Force spectroscopy measurements performed along DNA molecules reveal two distinguishable types of interaction. This result suggests that the chemical composition of the minor-major groove and/or of the sugar-phosphate backbone along the molecule can be detected, opening numerous possibilities for further studies and applications of this technique.
