Confocal microscopyConfocal microscopy, most frequently confocal laser scanning microscopy (CLSM) or laser scanning confocal microscopy (LSCM), is an optical imaging technique for increasing optical resolution and contrast of a micrograph by means of using a spatial pinhole to block out-of-focus light in image formation. Capturing multiple two-dimensional images at different depths in a sample enables the reconstruction of three-dimensional structures (a process known as optical sectioning) within an object.
Cardinal directionThe four cardinal directions, or cardinal points, are the four main compass directions: north, south, east, and west, commonly denoted by their initials N, S, E, and W respectively. Relative to north, the directions east, south, and west are at 90 degree intervals in the clockwise direction. The ordinal directions (also called the intercardinal directions) are northeast (NE), southeast (SE), southwest (SW), and northwest (NW). The intermediate direction of every set of intercardinal and cardinal direction is called a secondary intercardinal direction.
Pinhole cameraA pinhole camera is a simple camera without a lens but with a tiny aperture (the so-called pinhole)—effectively a light-proof box with a small hole in one side. Light from a scene passes through the aperture and projects an inverted image on the opposite side of the box, which is known as the camera obscura effect. The size of the images depends on the distance between the object and the pinhole. The camera obscura or pinhole image is a natural optical phenomenon.
Optical microscopeThe optical microscope, also referred to as a light microscope, is a type of microscope that commonly uses visible light and a system of lenses to generate magnified images of small objects. Optical microscopes are the oldest design of microscope and were possibly invented in their present compound form in the 17th century. Basic optical microscopes can be very simple, although many complex designs aim to improve resolution and sample contrast. The object is placed on a stage and may be directly viewed through one or two eyepieces on the microscope.
NorthNorth is one of the four compass points or cardinal directions. It is the opposite of south and is perpendicular to east and west. The word north is related to the Old High German nord, both descending from the Proto-Indo-European unit *ner-, meaning "left; below" as north is to left when facing the rising sun. Binary relationIn mathematics, a binary relation associates elements of one set, called the domain, with elements of another set, called the codomain. A binary relation over sets X and Y is a new set of ordered pairs (x, y) consisting of elements x in X and y in Y. It is a generalization of the more widely understood idea of a unary function. It encodes the common concept of relation: an element x is related to an element y, if and only if the pair (x, y) belongs to the set of ordered pairs that defines the binary relation.
Optical heterodyne detectionOptical heterodyne detection is a method of extracting information encoded as modulation of the phase, frequency or both of electromagnetic radiation in the wavelength band of visible or infrared light. The light signal is compared with standard or reference light from a "local oscillator" (LO) that would have a fixed offset in frequency and phase from the signal if the latter carried null information. "Heterodyne" signifies more than one frequency, in contrast to the single frequency employed in homodyne detection.
Dark-field microscopyDark-field microscopy (also called dark-ground microscopy) describes microscopy methods, in both light and electron microscopy, which exclude the unscattered beam from the image. As a result, the field around the specimen (i.e., where there is no specimen to scatter the beam) is generally dark. In optical microscopes a darkfield condenser lens must be used, which directs a cone of light away from the objective lens. To maximize the scattered light-gathering power of the objective lens, oil immersion is used and the numerical aperture (NA) of the objective lens must be less than 1.
