Influence of Microplastics on Microbial Structure, Function, and Mechanical Properties of Stream Periphyton

Periphyton is a freshwater biofilm composed of prokaryotic and eukaryotic communities that occupy rocks and sediments, forming the base of the food web and playing a key role in nutrient cycling. Given the large surface that periphyton comprises, it may also act as a sink for a diverse range of man-made pollutants, including microplastics (MP). Here we investigated the effect of 1-4 mu m and 63-75 mu m sized, spherical polyethylene MP with native and ultraviolet (UV)-weathered surface on developing natural stream periphyton communities over 28 days. In order to ensure proper particle exposure, we first tested MP suspension in water or in water containing either Tween 80, extracellular polymeric substances - EPS, fulvic acids, or protein. We found the extract of EPS from natural periphyton to be most suitable to create MP suspensions in preparation of exposure. Upon exposure, all tested types of MP were found to be associated with the periphyton, independent of their size and other properties. While biomass accrual and phenotypic community structure of the photoautotrophs remained unchanged, the prokaryotic and eukaryotic communities experienced a significant change in composition and relative abundances. Moreover, alpha diversity was affected in eukaryotes, but not in prokaryotes. The observed changes were more prominent in periphyton exposed to UV-treated as compared with native surface MP. Mechanical properties, as assessed by compression rheology, showed that MP-exposed periphyton had longer filamentous streamers, higher stiffness, lower force recovery and a higher viscoelasticity than control periphyton. Despite the observed structural and mechanical changes of periphyton, functional parameters (i.e., photosynthetic yield, respiration and nutrient uptake efficiencies) were not altered by MP, indicating the absence of MP toxicity, and suggesting functional redundancy in the communities. Together, our results provide further proof that periphyton is a sink for MP and demonstrate that MP can impact local microbial community composition and mechanical properties of the biofilms. Consequences of these findings might be a change in dislodgement behavior of periphyton, a propagation through the food chains and impacts on nutrient cycling and energy transfer. Hence, taking the omnipresence, high persistence and material and size diversity of MP in the aquatic environment into account, their ecological consequences need further investigation.

Shifts in diversity and function of lake bacterial communities upon glacier retreat

Global climate change is causing a wastage of glaciers and threatening biodiversity in glacier-fed ecosystems. The high turbidity typically found in those ecosystems, which is caused by inorganic particles and result of the erosive activity of glaciers is a key environmental factor influencing temperature and light availability, as well as other factors in the water column. Once these lakes loose hydrological connectivity to glaciers and turn clear, the accompanying environmental changes could represent a potential bottleneck for the established local diversity with yet unknown functional consequences. Here, we study three lakes situated along a turbidity gradient as well as one clear unconnected lake and evaluate seasonal changes in their bacterial community composition and diversity. Further, we assess potential consequences for community functioning. Glacier runoff represented a diverse source community for the lakes and several taxa were able to colonize downstream turbid habitats, although they were not found in the clear lake. Operational taxonomic unit-based alpha diversity and phylogenetic diversity decreased along the turbidity gradient, but metabolic functional diversity was negatively related to turbidity. No evidence for multifunctional redundancy, which may allow communities to maintain functioning upon alterations in diversity, was found. Our study gives a first view on how glacier-fed lake bacterial communities are affected by the melting of glaciers and indicates that diversity and community composition significantly change when hydrological connectivity to the glacier is lost and lakes turn clear.

Contrasting responses of bacterial and fungal communities to plant litter diversity in a Mediterranean oak forest

Tree species diversity of forested ecosystems control the diversity of leaf litter inputs to the soil, with cascading effects on the microbial communities colonizing decomposing litter. However, the extent to which bacterial and fungal communities inhabiting the litter layer are affected by shifts in tree species diversity is not well understood. To investigate the role of litter species diversity, litter species identity and litter functional traits on bacterial and fungal communities of a typical Mediterranean oak forest, we set up a yearly field litterbag experiment that considered leaf litter mixtures of four abundant species: Quercus pubescens, Acer monspessulanum, Cotinus coggygria and Pinus halepensis. We found that both bacterial and fungal communities varied strongly during decomposition but showed distinct succession patterns. Both communities were also strongly influenced by litter species diversity, litter identity and litter functional traits. The intensity and the direction of these effects varied during decomposition. Litter diversity effects were mediated by litter species composition rather than litter species richness, highlighting the importance of litter species identity - and associated litter traits - as drivers of microbial communities. Both the "mass-ratio hypothesis", measured through the community weighted mean (CWM) litter traits, and the "niche complementarity hypothesis", measured through the functional dissimilarity (FD) of litter traits, contributed to litter diversity effects, with a greater relative importance of FD compared to CWM, and with an overall stronger impact on fungal-than on bacterial-communities. Interestingly, increasing FD was related to decreasing bacterial diversity, but increasing fungal diversity. Our findings provide clear evidence that any alteration of plant species diversity produces strong cascading effects on microbial communities inhabiting the litter layer in the studied Mediterranean oak forest.

Assessing the responses of Sphagnum micro-eukaryotes to climate changes using high throughput sequencing

Current projections suggest that climate warming will be accompanied by more frequent and severe drought events. Peatlands store ca. one third of the world's soil organic carbon. Warming and drought may cause peatlands to become carbon sources through stimulation of microbial activity increasing ecosystem respiration, with positive feedback effect on global warming. Micro-eukaryotes play a key role in the carbon cycle through food web interactions and therefore, alterations in their community structure and diversity may affect ecosystem functioning and could reflect these changes. We assessed the diversity and community composition of Sphagnum-associated eukaryotic microorganisms inhabiting peatlands and their response to experimental drought and warming using high throughput sequencing of environmental DNA. Under drier conditions, micro-eukaryotic diversity decreased, the relative abundance of autotrophs increased and that of osmotrophs (including Fungi and Peronosporomycetes) decreased. Furthermore, we identified climate change indicators that could be used as early indicators of change in peatland microbial communities and ecosystem functioning. The changes we observed indicate a shift towards a more "terrestrial'' community in response to drought, in line with observed changes in the functioning of the ecosystem.
