Facilitating sensory responses in developing mouse somatosensory barrel cortex

The sensory responses in the barrel cortex of mice aged postnatal day (P)7-P12 evoked by a single whisker deflection are smaller in amplitude and spread over a smaller area than those measured in P13-P21 mice. However, repetitive 10-Hz stimulation or paired pulse whisker stimulation in P7-P12 mice evoked facilitating sensory responses, contrasting with the depressing sensory responses observed in P13-P21 mice. This facilitation occurred during an interval ranging 300-1,000 ms after the first stimulus and was measured using whole cell recordings, voltage-sensitive dye imaging, and calcium-sensitive dye imaging. The facilitated responses were not only larger in amplitude but also propagated over a larger cortical area. The facilitation could be blocked by local application of pharmacological agents reducing cortical excitability. Local cortical microstimulation could substitute for the first whisker stimulus to produce a facilitated sensory response. The enhanced sensory responses evoked by repetitive sensory stimuli in P7-P12 mice may contribute to the activity-dependent specification of the developing cortical circuits. In addition, the facilitating sensory responses allow long integration times for sensory processing compatible with the slow behavior of mice during early postnatal development.

Visualizing the cortical representation of whisker touch: voltage-sensitive dye imaging in freely moving mice

Voltage-sensitive dye imaging resolves the spatiotemporal dynamics of supragranular subthreshold cortical activity with millisecond temporal resolution and subcolumnar spatial resolution. We used a flexible fiber optic image bundle to visualize voltage-sensitive dye dynamics in the barrel cortex of freely moving mice while simultaneously filming whisker-related behavior to generate two movies matched frame-by-frame with a temporal resolution of up to 2 ms. Sensory responses evoked by passive whisker stimulation lasted longer and spread further across the barrel cortex in awake mice compared to anesthetized mice. Passively evoked sensory responses were large during behaviorally quiet periods and small during active whisking. However, as an exploring mouse approached an object while whisking, large-amplitude, propagating cortical sensory activity was evoked by active whisker-touch. These experiments demonstrate that fiber optics can be used to image cortical sensory activity with high resolution in freely moving animals. The results demonstrate differential processing of sensory input depending upon behavior.

Interaction of sensory responses with spontaneous depolarization in layer 2/3 barrel cortex

The rodent primary somatosensory cortex is spontaneously active in the form of locally synchronous membrane depolarizations (UP states) separated by quiescent hyperpolarized periods (DOWN states) both under anesthesia and during quiet wakefulness. In vivo whole-cell recordings and tetrode unit recordings were combined with voltage-sensitive dye imaging to analyze the relationship of the activity of individual pyramidal neurons in layer 2/3 to the ensemble spatiotemporal dynamics of the spontaneous depolarizations. These were either brief and localized to an area of a barrel column or occurred as propagating waves dependent on local glutamatergic synaptic transmission in layer 2/3. Spontaneous activity inhibited the sensory responses evoked by whisker deflection, accounting almost entirely for the large trial-to-trial variability of sensory-evoked postsynaptic potentials and action potentials. Subthreshold sensory synaptic responses evoked while a cortical area was spontaneously depolarized were smaller, briefer and spatially more confined. Surprisingly, whisker deflections evoked fewer action potentials during the spontaneous depolarizations despite neurons being closer to threshold. The ongoing spontaneous activity thus regulates the amplitude and the time-dependent spread of the sensory response in layer 2/3 barrel cortex.

Two-photon calcium imaging of neocortical projection neurons in whisker somatosensory cortex during goal-directed sensorimotor learning

Abstract Excitatory projection neurons of the neocortex are thought to play important roles in per-ceptual and cognitive functions of the brain by directly connecting diverse cortical and subcortical areas. However, many aspects of the anatomical and functional organization of these inter-areal connections are unknown. The mouse primary somatosensory whisk-er barrel cortex (S1) serves as an important model for investigating the mammalian neo-cortex, and, here, I firstly investigate the structure and secondly the function of a specific subset of S1 cortico-cortical long-range projection neurons. In the first part of my thesis, I studied long-range axonal projections of excitatory layer 2/3 neurons with cell bodies located in S1. As a population, these neurons densely projected to secondary whisker somatosensory cortex (S2) and primary/secondary whisker motor cortex (M1/2), with additional axon in the dysgranular zone surrounding the barrel field, perirhinal temporal association cortex and striatum. The execution of a goal-directed behavior requires the brain to process incoming sensory information from the environment in a context-, learning- and motivation-dependent manner in order to perform specific motor actions. Cortico-cortical communica-tion in the context of goal-directed sensorimotor transformation has begun to be studied, but little is known about how signaling between interconnected cortical areas is modified by sensorimotor learning, as well as in response to changes in reward contingencies. Hence, in the second part of my thesis, I studied cortico-cortical dynamics in primary whisker somatosensory barrel cortex (S1) of mice during a combined whisker and audito-ry task. Using transgenic mice expressing GCaMP6f combined with two-photon micros-copy and retrograde labeling techniques, I chronically monitored the activity of excitatory layer 2/3 neurons in S1 projecting to M1 or S2, while mice learned the behavioral switch task. The results demonstrated that both classes of neurons responded after whisker and auditory stimulation. However, the whisker stimulus evoked response was stronger than the auditory stimulus evoked response. Neurons projecting to S2 exhibited stronger re-sponses compared to neurons projecting to M1 neurons. Those responses remained rela-tively stable across training sessions and under different reward conditions. Furthermore, both classes of neurons responded during spontaneous licking, but neurons projecting to S2 had larger licking-related responses compared to neurons projecting to M1. This work therefore furthers our knowledge of the structure and function of specific types of cortical projection neurons, which is a necessary step towards detailed under-standing of how sensory information might be signaled from primary sensory areas to downstream brain regions for further processing.
