Protéine

Protéine de fusion

Une protéine de fusion est une protéine artificielle obtenue par la combinaison de différentes protéines, ou partie de protéines. Elle est obtenue à la suite de la création par recombinaison de l'ADN d'un gène comportant les cadres de lecture ouverts correspondant aux protéines ou parties de protéines désirées. Les protéines de fusion peuvent également être appelées protéines chimères. Une des applications les plus connues des protéines de fusion est la fusion d'une protéine d'intérêt à une protéine fluorescente.

The invention relates to new proteins called alkylcytosine transferases (ACTs) derived from O6-alkylguanine-DNA alkyltransferase, and to substrates for ACTs specifically transferring a label to these ACTs and to fusion proteins comprising these. The substrates according of the invention are substituted cytosines of formula (I) wherein R1 is an aromatic or a heteroaromatic group, or an optionally substituted unsaturated alkyl, cycloalkyl or heterocyclyl group with the double bond connected to OCH2-; R2 is a linker; and L is a label or a plurality of same or different labels. The invention further relates to methods of transferring label L from these substrates of formula (I) to ACTs and ACT fusion proteins. The system of ACT-compound of formula (I) is particularly suitable for double labelling studies together with the known system O6-alkylguanine-DNA alkyltransferase (AGT)-benzylguanines.

O6-alkylguanine-DNA alkyltransferases for efficient labeling of fusion proteins

The present invention relates to methods of transferring a label from suitable substrates to O6-alkylguanine-DNA alkyltransferase (AGT) fusion proteins, to suitable fusion proteins, to suitable variants of AGT, and to novel labeled fusion proteins obtained. A protein of interest is incorporated into an AGT fusion protein, the AGT fusion protein is contacted with an AGT substrate carrying a label, and the AGT fusion protein is detected and/or manipulated using the label in a system designed for recognizing and/or handling the label. This allows the detection and/or manipulating of the fusion protein, both in vitro and in vivo, by attaching mols. to the fusion proteins that introduce a new phys. or chem. property to the fusion protein. Examples of such mols. are, among others, spectroscopic probes or radioactively labeled mols., affinity tags, mols. generating reactive or hydroxyl radicals, cross-linkers and lipid. AGT in fusion protein is a human variant wherein Asn157 is replaced by Gly and Ser159 by Glu, and the one wherein Gly160 is replaced by Ala or Trp. AGT in fusion protein is a human variant wherein Asn157 is replaced by Ser, Ser159 by His, and Gly160 by Asn. [on SciFinder (R)]
