In Silico Voltage-Sensitive Dye Imaging: A Model-Based Approach for Bridging Scales of Cortical Activity

An understanding of how microscale voltage fluctuations interact to shape mesoscale cortical information processing is lacking. Voltage-sensitive dye imaging (VSDI) is a powerful technique for interrogating membrane potential dynamics in assemblies of cortical neurons, but with effective resolution limits that can confound interpretation. To address this limitation, we developed an in silico model of VSDI in a detailed digital reconstruction of rodent neocortical microcircuitry. Using this model, we clarify and extend previous experimental findings regarding the cellular origins and spatiotemporal dynamics of VSDI signals. Furthermore, we test the capacity of VSD image sequences to discriminate between afferent thalamic inputs at various spatial locations to estimate a lower bound on the functional resolution of VSDI. In addition, we explore experimentally inaccessible circuit properties to show that during periods of spontaneous activity, membrane potential fluctuations are anticorrelated with population firing rates. This relationship is dependent on recurrent connectivity, which we demonstrate by manipulating network connections. Finally, we investigate how the reciprocity between population spiking and membrane potential is affected by long-range, extrinsic synaptic input by adding artificial excitatory synapses to a subpopulation of layer 5 neurons. Based on our findings, we suggest that the lag time of peak anticorrelation between the VSD signal and population firing rate reflects the degree of perturbing external influence on the local microcircuitry. Our approach reveals that VSDI is a richer source of information than previously appreciated and underscores the power of a bottom-up computational approach for relating scales of cortical processing.

Sensory memory in a model of a cortical column

Sensory memory is the first step of a complex memorization process. Sensory information is buffered in a "sensory store" for a short period. Then part of it is transferred to working memory, where information can be actively processed and, eventually, through its rehearsal, can be memorized and stored in long-term memory. Sensory memory is characterized by a high capacity and a short temporal extension. In this work we show that such a transient buffering of information is well captured in models of cortical microcircuits. Sensory information stays for a short time in the perturbations it induced to the neural activity. Two fundamental characteristics of cortical columns appear to be useful to this purpose. The first one is an approximate balance of neuronal excitation and inhibition. The second one is the sparseness of cells recurrent connections. Together, these two discriminants ensure a broad set of complex dynamical behaviours of the neural network along with the presence of a phase of sustained non-oscillatory activity with very irregular spike trains in individual neurons. We show that at the vicinity of a phase transition from a quiescent state to a phase of chaotic spiking activity, global stimuli can be buffered in the "macroscopic" neuronal activity. However if the sensory input only excites a subpart of the network, efficient information buffering is made using the detailed spiking activity of every neuron in the network. The "microscopic" structure of the network is therefore needed in order to buffer sensory information. In most situations, addition of noise enhances the buffering performance and the computational power of the network. This effect is known in the physical literature as "stochastic resonance". If the network has to achieve complex transformation of its inputs, stochastic resonance is shown to be an emergent property of the population of neurons. Both the very irregular spiking activity present in vivo and the balanced excitation and inhibition measured in cortical columns have a new functional relevance in being the neuronal substrate of an efficient sensory memory .

Theory of representation learning in cortical neural networks

Our brain continuously self-organizes to construct and maintain an internal representation of the world based on the information arriving through sensory stimuli. Remarkably, cortical areas related to different sensory modalities appear to share the same functional unit, the neuron, and develop through the same learning mechanism, synaptic plasticity. It motivates the conjecture of a unifying theory to explain cortical representational learning across sensory modalities. In this thesis we present theories and computational models of learning and optimization in neural networks, postulating functional properties of synaptic plasticity that support the apparent universal learning capacity of cortical networks. In the past decades, a variety of theories and models have been proposed to describe receptive field formation in sensory areas. They include normative models such as sparse coding, and bottom-up models such as spike-timing dependent plasticity. We bring together candidate explanations by demonstrating that in fact a single principle is sufficient to explain receptive field development. First, we show that many representative models of sensory development are in fact implementing variations of a common principle: nonlinear Hebbian learning. Second, we reveal that nonlinear Hebbian learning is sufficient for receptive field formation through sensory inputs. A surprising result is that our findings are independent of specific details, and allow for robust predictions of the learned receptive fields. Thus nonlinear Hebbian learning and natural statistics can account for many aspects of receptive field formation across models and sensory modalities. The Hebbian learning theory substantiates that synaptic plasticity can be interpreted as an optimization procedure, implementing stochastic gradient descent. In stochastic gradient descent inputs arrive sequentially, as in sensory streams. However, individual data samples have very little information about the correct learning signal, and it becomes a fundamental problem to know how many samples are required for reliable synaptic changes. Through estimation theory, we develop a novel adaptive learning rate model, that adapts the magnitude of synaptic changes based on the statistics of the learning signal, enabling an optimal use of data samples. Our model has a simple implementation and demonstrates improved learning speed, making this a promising candidate for large artificial neural network applications. The model also makes predictions on how cortical plasticity may modulate synaptic plasticity for optimal learning. The optimal sampling size for reliable learning allows us to estimate optimal learning times for a given model. We apply this theory to derive analytical bounds on times for the optimization of synaptic connections. First, we show this optimization problem to have exponentially many saddle-nodes, which lead to small gradients and slow learning. Second, we show that the number of input synapses to a neuron modulates the magnitude of the initial gradient, determining the duration of learning. Our final result reveals that the learning duration increases supra-linearly with the number of synapses, suggesting an effective limit on synaptic connections and receptive field sizes in developing neural networks.
