Methods and app. to study biol. mols. or biol. components at or in self-assembled monolayers (SAMs) on metal surfaces such as gold in aq. environments using attenuated total internal reflection Fourier transform infra-red (ATR-FTIR) spectroscopy are described. The method may be used for, e.g., the screening for effective substances suitable as agonists or antagonists or - if performed time resolved - for studying kinetics of interactions of, e.g., biol. mols. adsorption and desorption processes or for the development of materials and surfaces for biol. or medical applications. The binding reactions of an anti-lysozyme Fab fragment bearing a hexahistidine extension at the C-terminus of the heavy chain were studied using a nickel-charged CTA SAM on a gold-coated germanium ATR element.
