This lecture covers the history and mechanisms of CRISPR-based tools and diagnostics, including the discovery of CRISPRs in prokaryotes, the development of cas genes, and the functional importance of CRISPR loci. It also delves into the CRISPR-cas adaptive immune system in prokaryotes, the adaptation process, crRNA maturation, interference mechanisms, and the use of Cas proteins for spacer acquisition. Advanced concepts such as the SHERLOCK detection assay and the integration of internal controls for RNAse contamination are discussed, along with the principles of isothermal amplification and the use of CRISPR for viral material detection.
