This lecture covers the design of novel enzymes through enzyme engineering, focusing on the creation of a novel enzyme from scratch, including the transition state model, optimization of catalytic side-chains, and ranking based on binding energy. It also explores the application of enzyme design for sustainable production. Examples of protein design for enzyme engineering and ligand biosensor design are presented, showcasing the use of computational protein design to build novel enzymes and selective bZIP-binding peptides. The lecture concludes with the de novo design of multi-state biosensors for various applications in synthetic biology.
