This lecture delves into metabolic signaling, focusing on glycolysis, anaerobic vs aerobic metabolism, and the Warburg Effect in cancer. It covers the typical workflow of extracellular flux assays, metabolomics, proteomics tools, and methods to monitor cell metabolic status. The lecture also explores the impact of glycolytic signaling switches in cancer and the methods to perturb the electron transport chain. A case study on breast tumor aggressiveness is presented, showcasing integrated quantitative proteomics and metabolomics interrogations to assess relative expression levels of proteins and subcellular localization. The study also investigates cellular ATP flux and the correlation between metabolic dysfunction and cancer progression.
