Adipose-specific knockout of raptor results in lean mice with enhanced mitochondrial respiration

raptor is a specific and essential component of mammalian TOR complex 1 (mTORC1), a key regulator of cell growth and metabolism. To investigate a role of adipose mTORC1 in regulation of adipose and whole-body metabolism, we generated mice with an adipose-specific knockout of raptor (raptor(ad-/-)). Compared to control littermates, raptor(ad-/-) mice had substantially less adipose tissue, were protected against diet-induced obesity and hypercholesterolemia, and exhibited improved insulin sensitivity. Leanness was in spite of reduced physical activity and unaffected caloric intake, lipolysis, and absorption of lipids from the food. White adipose tissue of raptor(ad-/-) mice displayed enhanced expression of genes encoding mitochondrial uncoupling proteins characteristic of brown fat. Leanness of the raptor(ad-/-) mice was attributed to elevated energy expenditure due to mitochondrial uncoupling. These results suggest that adipose mTORC1 is a regulator of adipose metabolism and, thereby, controls whole-body energy homeostasis.

Thermal microsensors for in vitro and in vivo monitoring of chemical and biological processes

Metabolism is a highly coordinated biochemical cellular activity indispensable for sustaining life, and is often related to mitochondrial functioning. Innovative technological approaches allowing the study of the thermal signature of chemical and metabolic processes are presented in this thesis. In particular, two solutions are proposed: (i) a nanocalorimetric platform enabling measurements of biochemical heat production in vitro, and (ii) a method for localized in vivo temperature measurements in mice using implantable miniaturized sensors. First, the design and development of the nanocalorimetric platform is presented. In particular, this platform is built around a commercial thermopile-based sensor chip, on which an open-well reservoir holding a sample with a volume of a few tens of microliter is directly positioned. Both components are embedded into an isothermal housing providing an excellent temperature stability (± 1 mK), which is a prerequisite for accurate heat power measurements. Furthermore, the platform is characterized by a fast thermalization time to the target temperature (less than 30 minutes) and a fast sensing response time (a few seconds). Numerical simulations, which allowed the optimization of the platform design, are also reported. Electrical calibration of the platform was carried out using resistive heaters positioned either on the sensor chip or in the sample reservoir. A heat power limit of detection of 70 nW was estimated. Then, two reactions for the validation of the nanocalorimetric platform were investigated: (i) the mixing of 1-propanol in water, and (ii) the oxidation of glucose catalyzed by glucose oxidase. The results showed good agreement with literature, confirming that our versatile platform may be applied to many thermochemical studies, including thermodynamic analysis and kinetic reaction analysis. Furthermore, localized in vivo temperature measurements in mice by means of implantable miniaturized sensors are presented. The aim was to monitor mouse metabolism during cold exposure, and to record possible temperature differences between the body temperature measured in the abdomen and the temperature of the brown adipose tissue (BAT) situated in the interscapular area. This approach is of biological interest as it may help unravelling the question whether biochemical activation of BAT is associated with local increase in metabolic heat production. Specifically, the preparation and calibration of miniaturized thermistor sensors are reported and the surgical procedure is also described. Temperature measurements, carried out on mice during cold exposure (6 °C) for a maximum duration of 6 hours, are presented and analyzed. Control measurements with a conventional probe confirmed the good performance of the implanted sensors. Moreover, analysis of the experimental results allowed the identification of two different mouse phenotypes, distinguishable in terms of their metabolic resistance to cold exposure. This difference was investigated from the thermal point of view by computational simulations. A simple physical model of the mouse body allowed to reproduce the global evolution of hypothermia and to explain qualitatively the temperature difference between abdomen and BAT locations. While with this approach, we have demonstrated the importance and feasibility of localized temperature measurements on mice, further optimization of this technique is foreseen to better identify local metabolism variations.

Differential roles of GDF15 and FGF21 in systemic metabolic adaptation to the mitochondrial integrated stress response

Perturbation of mitochondrial proteostasis provokes cell autonomous and cell non-autonomous responses that contribute to homeostatic adaptation. Here, we demonstrate distinct metabolic effects of hepatic metabokines as cell non-autonomous factors in mice with mitochondrial OxPhos dysfunction. Liver-specific mitochondrial stress induced by a loss-of-function mutation in Crif1 (LKO) leads to aberrant oxidative phosphorylation and promotes the mitochondrial unfolded protein response. LKO mice are highly insulin sensitive and resistant to diet-induced obesity. The hepatocytes of LKO mice secrete large quantities of metabokines, including GDF15 and FGF21, which confer metabolic benefits. We evaluated the metabolic phenotypes of LKO mice with global deficiency of GDF15 or FGF21 and show that GDF15 regulates body and fat mass and prevents diet-induced hepatic steatosis, whereas FGF21 upregulates insulin sensitivity, energy expenditure, and thermogenesis in white adipose tissue. This study reveals that the mitochondrial integrated stress response (ISRmt) in liver mediates metabolic adaptation through hepatic metabokines.

E2F transcription factor-1 regulates oxidative metabolism

Cells respond to stress by coordinating proliferative and metabolic pathways. Starvation restricts cell proliferative (glycolytic) and activates energy productive (oxidative) pathways. Conversely, cell growth and proliferation require increased glycolytic and decreased oxidative metabolism levels. E2F transcription factors regulate both proliferative and metabolic genes. E2Fs have been implicated in the G1/S cell-cycle transition, DNA repair, apoptosis, development and differentiation. In pancreatic β-cells, E2F1 gene regulation facilitated glucose-stimulated insulin secretion. Moreover, mice lacking E2F1 (E2f1(-/-)) were resistant to diet-induced obesity. Here, we show that E2F1 coordinates cellular responses by acting as a regulatory switch between cell proliferation and metabolism. In basal conditions, E2F1 repressed key genes that regulate energy homeostasis and mitochondrial functions in muscle and brown adipose tissue. Consequently, E2f1(-/-) mice had a marked oxidative phenotype. An association between E2F1 and pRB was required for repression of genes implicated in oxidative metabolism. This repression was alleviated in a constitutively active CDK4 (CDK4(R24C)) mouse model or when adaptation to energy demand was required. Thus, E2F1 represents a metabolic switch from oxidative to glycolytic metabolism that responds to stressful conditions.
