Impact of Carbon-Nanotube Substrate Coating in Neuronal Networks

The interest in new materials and their application in the neuroscience are growing incredibly fast. Current research interests are increasingly focusing on new artificial materials and their direct effects on the central nervous systems so that it would be possible use them to restore the original nervous function lost due to neuronal diseases or trauma. This thesis is part of the international Neuronano project and has been funded by the European Commission 6th Framework Programme. The main goal is to understand the interactions of carbon nanotubes (CNT) with the nervous tissues. Not only it focuses on how neuron stimulations and recordings instruments may be ameliorated by a CNT-neuron interface, but it also focuses on the endogenous effects of those artificial materials on neuronal activity. This thesis is divided into four themes: First, an integrated CNT-neuron system was developed to test whether electrical voltage stimulation, delivered via nanotubes, can induce neuronal signaling. It was also compared to neuronal responses evoked by voltage steps delivered via conventional patch-pipette. In the second part, the focus was on understanding the CNT-neuron interface and the tight contacts with the cell membranes that might favour electrical shortcuts between neuronal compartments. The third part is an overview of the effects of a CNT coated substrates when employed to grow long-term cell cultures. The activity was constantly monitored by an extracellular array of electrodes (MEA) for several weeks. The intimate relationship between nanotubes and cell membrane observed at single-neuron and network level, results in a more robust network activity. Finally, the MEA setup developed for cell cultures monitoring was used to investigate the effects of of BDNF on cortical cell culture model of Huntington disease.

Sensory memory in a model of a cortical column

Sensory memory is the first step of a complex memorization process. Sensory information is buffered in a "sensory store" for a short period. Then part of it is transferred to working memory, where information can be actively processed and, eventually, through its rehearsal, can be memorized and stored in long-term memory. Sensory memory is characterized by a high capacity and a short temporal extension. In this work we show that such a transient buffering of information is well captured in models of cortical microcircuits. Sensory information stays for a short time in the perturbations it induced to the neural activity. Two fundamental characteristics of cortical columns appear to be useful to this purpose. The first one is an approximate balance of neuronal excitation and inhibition. The second one is the sparseness of cells recurrent connections. Together, these two discriminants ensure a broad set of complex dynamical behaviours of the neural network along with the presence of a phase of sustained non-oscillatory activity with very irregular spike trains in individual neurons. We show that at the vicinity of a phase transition from a quiescent state to a phase of chaotic spiking activity, global stimuli can be buffered in the "macroscopic" neuronal activity. However if the sensory input only excites a subpart of the network, efficient information buffering is made using the detailed spiking activity of every neuron in the network. The "microscopic" structure of the network is therefore needed in order to buffer sensory information. In most situations, addition of noise enhances the buffering performance and the computational power of the network. This effect is known in the physical literature as "stochastic resonance". If the network has to achieve complex transformation of its inputs, stochastic resonance is shown to be an emergent property of the population of neurons. Both the very irregular spiking activity present in vivo and the balanced excitation and inhibition measured in cortical columns have a new functional relevance in being the neuronal substrate of an efficient sensory memory .

Interfacing neurons with carbon nanotubes: electrical signal transfer and synaptic stimulation in cultured brain circuits

The unique properties of single-wall carbon nanotubes (SWNTs) and the application of nanotechnology to the nervous system may have a tremendous impact in the future developments of microsystems for neural prosthetics as well as immediate benefits for basic research. Despite increasing interest in neuroscience nanotechnologies, little is known about the electrical interactions between nanomaterials and neurons. We developed an integrated SWNT-neuron system to test whether electrical stimulation delivered via SWNT can induce neuronal signaling. To that aim, hippocampal cells were grown on pure SWNT substrates and patch clamped. We compared neuronal responses to voltage steps delivered either via conductive SWNT substrates or via the patch pipette. Our experimental results, supported by mathematical models to describe the electrical interactions occurring in SWNT-neuron hybrid systems, clearly indicate that SWNTs can directly stimulate brain circuit activity.

Lentiviral Transgenesis as a Tool to Study the Role of Uncoupling Protein Isoforms in the Nervous System

A tight coupling exists between synaptic activity and glucose utilization by astrocytes. Metabolic cooperation between neurons and astrocytes mediates this coupling. During synaptic activation, glutamate that is released in the synaptic cleft as a neurotransmitter by neurons is rapidly cleared by an active uptake into astrocytes. One glutamate is co-transported with three Na+. Intracellular astrocytic Na+ homeostasis is re-established by the Na+/K+-ATPase which requires ATP provided mainly by glycolysis. The resulting lactate is released in extracellular space and contributes to the energetic balance of activated neurons. This coupling between astrocytes and neurons is known as the astrocyte-neuron lactate shuttle hypothesis (ANLSH). The role of mitochondria in this coupling remains to be determined and more specifically the role of uncoupling proteins (UCP) which have been recently identified in neural tissues. UCPs belong to a family of mitochondrial carriers which are present in the inner mitochondrial membrane. There are five isoforms named UCP1 to UCP5. The well-known characterized is UCP1, also named thermogenin, which is present in the brown adipose tissue (BAT) of the small rodents and is responsible of non-shivering thermogenesis. It dissipates the proton gradient across the inner mitochondrial membrane, thereby producing heat instead of ATP. UCP2 is ubiquitous and is implicated in protection against excessive reactive oxygen (ROS) production. The expression of UCP3 is restricted to skeletal muscle where it is linked to fatty acid metabolism. The role of the brain isoforms UCP4 and UCP5 is still poorly understood. Initially, we set out to evaluate their function in energy metabolism of astrocytes and neurons. We used the lentiviral strategy to overexpress or silence the UCPs isoforms in primary cultures of astrocytes and neurons. We found that only UCP4 and UCP5 had an uncoupling activity in brain cells. Indeed, we observed a reduced mitochondrial potential and a reduced ATP/ADP ratio in astrocytes as well as in neurons overexpressing UCP4 or UCP5. UCP4 reduced the oxidative pathway of astrocytes without modifying the basal glucose metabolism and without having a lethal effect on the cell. Oxygen consumption rate (OCR) of brain cells ovexpressing UCPs was reduced. Moreover, UCP4 increased lactate release by astrocytes, suggesting an implication in the astrocyte-neuron coupling. We also brought evidence that both UCP4 and UCP5 partially protect brain cells from oxidative damage. All these results were confirmed by experiments on UCPs silencing. In a second step, we used a co-culture model to study the impact of astrocytic uncoupling protein on neuronal survival and we demonstrated that the presence of uncoupling protein in astrocytes prevents neuronal death after glutamate excitotoxicity. Taken together, all these results support an important role of uncoupling proteins in the regulation of astrocytic energy production, thus promoting local export of lactate which can be used by neurons.
