Enantiopure, supramolecular helices containing three-dimensional tetranuclear lanthanide(III) arrays: Synthesis, structure, properties, and solvent-driven Trinuclear/Tetranuclear interconversion

The enantiomerically pure pinene-bipyridine-based receptor, (-) or (+) L-, diastereoselectively self-assembles in dry acetonitrile in the presence of Ln(III) ions (Ln = La, Pr, Nd, Sm, Eu, Gd, and Tb) to give a C-3-symmetrical, pyramidal architecture with the general formula Ln(4)(L)(9)(mu(3)-OH)(2)) (abbreviated as tetra-Ln(4)L(9)). Three metal centers shape the base: an equilateral triangle surrounded by two sets of helically wrapping ligands with opposite configurations. This part of the structure is very similar to the species Ln(3)(L-6(mu(3)-OH)(H2O)(3)(2)) (recently reported by us and abbreviated as tris-LnL(2)) formed by the ligand and the Ln(III) ions when the reactions are performed in methanol. The tetranuclear structure is completed by a capping, helical unit LnL(3) whose chirality is also predetermined by the chirality of the ligand. A complete characterization of these isostructural, chiral compounds was performed in solid state (X-ray, IR) and in solution (ES-MS, NMR, CD, UV-vis and emission spectroscopies). The sign and the intensity of the CD bands in the region of the pi pi* transitions of the bipyridine (absolute Delta epsilon values at 327 nm are about 280 M-1.cm(-1)) are highly influenced by the helicity of the capping fragment LnL(3). The photophysical properties (lifetime, quantum yield) of the visible (Eu and Tb complexes) and NIR (Nd complex) emitters indicate a good energy transfer between the ligands and the metal centers. The two related superstructures tetra-Ln(4)L(9) and tris-LnL(2) can be interconverted in acetonitrile, the switching process depending on the amount of water present in the solvent, the size of the Ln(III) ion, and the concentration. The weak chiral recognition capabilities of the self-assembly leading to the formation of tetra-Ln(4)L(9) either by direct synthesis from a racemic mixture of the ligand and Ln(III) ions or by the conversion of a tris-Ln(+/-)-L racemate were likewise demonstrated.

The diastereoselective synthesis of chiral coordination compounds with d metals proved the effectiveness of the pinene-bipyridine approach in predetermining the chirality at the metal centre. The strong Lewis acidic character of lanthanide cations required the modification of this class of ligands in order to adapt their coordination properties to the binding affinity of Ln(III) toward hard donor atoms like oxygen. The pinene bipyridine carboxylic derivative (+)-L- (figure 1) has been designed and synthesized to form configurationally stable lanthanide complexes. This ligand proved its effectiveness as chiral building block for the synthesis of oligometallic supramolecular structures. Indeed a self-assembly process takes place with complete diastereoselectivity between the enantiomerically pure (-) or (+) L- and Ln(III) ions (Ln = La, Pr, Nd, Sm, Eu, Gd, Tb, Dy, Ho, Er) in methanol, giving rise to trinuclear enantiopure structures with general formula Ln3(L)6(µ3-OH)(H2O)32 , possessing predetermined helical chirality. The ligands in this case display a new version of supramolecular helical chirality since they are wrapping around a trimetallic core instead of one single metal centre. The same components afford a second polynuclear structure if the reaction is carried out in CH3CN. In this medium a three-dimensional tetranuclear array self-assembles (with general formula Ln4(L)9(µ3-OH)2). The nine ligands are forming three distinct helical domains, two supramolecular as in the case of the trinuclear array and one classical around a single metal centre, with predetermined configuration. The two classes of compounds have been structurally characterized in solid state (X-ray and IR) and in solution (ES-MS, NMR). Their chiroptical properties were investigated by means of Circular Dichroism and Circularly Polarized Luminescence (for the emitting compounds). Trinuclear and tetranuclear arrays are shown to interconvert in CH3CN, in a process mediated by water. This process was rationalized taking into account the nature of the Ln(III) ions, the initial concentration of the components and the addition or removal of water in solution. The availability of both the enantiomers of the ligand prompted us to study the self-recognition capabilities of the two systems. 1H-NMR and CD studies reveal an almost complete chiral recognition in the self-assembly leading to the trinuclear complex. The process leading to tetranuclear species appears instead perturbed, probably due to the higher number of fundamental components involved (4 metals and 9 ligands instead of 3 metals and 6 ligands). Figure 1: Reaction conditions leading to the synthesis of trinuclear and tetranuclear Ln(III) complexes with ligand (+)-L-. The introduction of a carboxylic moiety onto the 6' posititon of the (-)-5,6- and (-)-4,5- pinene bipyridine lead to the tridentate ligands (-)-HL1 and (-)-HL2, respectively. Mononuclear neutral complexes are obtained upon reaction with Ln(ClO4)3. Different extents of diastereoselectivity are pointed out in the two systems by X-ray analysis and CD spectroscopy. In particular the ligand (-)-HL1, in which the pinene moiety is next to the binding site, seems to be more effective in predetermining the configuration at the metal centre. A low diastereoselectivity is instead expressed in the reactions with (-)-HL2 as revealed by the X-ray structure of [Pr{(-)-L2}3] (both Δ and Λ isomers detected) and by the CD spectrum void of signal.

Luminescent lanthanides bioprobes emitting in the visible and/or near-infrared ranges

This Ph.D. work deals with the development of lanthanide-based bioprobes for biomedical analyses and imaging. For this purpose, two different approaches have been followed, one focusing on visible-emitting complexes, while the second addresses the sensitization of NIR-emitting LnIII ions. The homoditopic ligands H2LC2 and H2LC3 have been tailored to self-assemble with lanthanide ions (LnIII) into neutral triple-stranded bimetallic helicates of overall composition [Ln2(L)3] and also to provide a versatile platform for further derivatization. The grafting of polyoxyethylene groups onto the pyridine (H2LC2) or the benzimidazole (H2LC3) units ensures water solubility, while maintaining large thermodynamic stability and adequate antenna effect for the excitation of LnIII ions. The conditional stability constants are large at physiological pH, with logβ23 reaching values in the range 22-25 for helicates with H2LC2 and H2LC3. The ligands triplet states feature adequate energy to efficiently sensitize the EuIII luminescence (QEu L = 21 and 11 % for L = H2LC2 and H2LC3, respectively) in aerated water at pH 7.4. Conversely, energy back transfer occurring in [Tb2(LC3)3] precludes a good sensitization of the TbIII luminescence, and only the TbIII helicate with H2LC2 displays sizeable quantum yield (QTbL = 11 %). Furthermore, it has been demonstrated that ligand H2LC2 sensitizes the emission of other visible- and NIR-emitting LnIII ions, such as SmIII or YbIII, for which in cellulo luminescence is evidenced, a rare observation. The EuIII emission spectra of both helicates arise from a main species with pseudo-D3 symmetry and without coordinated water. Cytotoxicity experiments reveal no significant effect of the helicates on the viability of several cancerous cell lines and cell imaging properties of the EuIII helicates are demonstrated for the HeLa cell line by luminescence microscopy. Bright EuIII emission is seen after relatively short time (15-30 min) and low helicate incubation concentrations (10 µM). Similar images were obtained for [Sm2(LC2)3] and [Tb2(LC2)3] using slightly higher loading concentration. The helicates stain the cytoplasm and the permeation mechanism is likely to be endocytosis. The described luminescent helical stains are robust chemical species which remain mostly undissociated in the cell medium and which resist to the presence of other complexing agents, as demonstrated in the case of H2LC2. All these properties make these helicates potential bioprobes for interacting with biological material and for cell imaging applications. As near-infrared (NIR) light, in addition to be less energetic than visible light, displays better penetrability into the human body, the development of bioprobes emitting in the near-infrared range is of prime importance for deeper tissue imaging, especially for early diagnosis of tumors. We therefore investigate the ability of new polydentate podands to sensitize the NIR luminescence of NdIII and YbIII ions. These ligands have been designed in a way to take advantage of the chelating effect of bidentate (Tsox, TsoxMe) or tridentate 8-hydroxyquinolinate (8-HQ) subunits (T2soxMe). The interaction between the ligands and LnIII ions in aqueous solution leads to the formation of stable 1:1 chelates (pLn in the range of 12-16), all the chromophoric 8-HQ units being coordinated to the metal center, exploiting the entropic effect generated by the anchor. At physiological pH, one major species is present for all the ligands regardless of the LnIII ion used. The low energy triplet states of the ligands are particularly well-suited for an efficient sensitization of the NdIII- or YbIII-centered luminescence (QNdL = 0.02-0.04 % and QYbL = 0.13-0.37 %) in buffered aqueous solution. ErIII NIR luminescence is also detected in water for the chelates with Tsox and TsoxMe. Lifetime determinations point in all cases to the absence of coordinated water molecules in the inner coordination sphere of the LnIII ion. It has also been demonstrated that methylation of the amide functions of Tsox removes the quenching mechanism induced by the proximate N-H groups and increases both the lifetimes and quantum yields of the TsoxMe chelates, with the YbIII complex being one of the most NIR luminescent lanthanide-based edifices described so far. Ligand TsoxMe thus appears to be the best candidate of the series regarding to the development of NIR probes for bioanalyses. Furthermore, other applications, for instance in telecommunications, are still conceivable since the incorporation of the Yb-TsoxMe chelate into sol-gel silica films has proven to be successful, the chelate withstanding the sol-gel process, while providing a slight enhancement of photophysical properties compared to the same chelate in aqueous solution.

Supramolecular assembly, chirality, and electronic properties of rubrene studied by STM and STS

This thesis presents the first experimental results of a scanning tunneling microscopy (STM) and scanning tunneling spectroscopy (STS) investigation of rubrene at the supramolecular, molecular and submolecular level. Based on its semiconducting and fluorescent properties, this molecule is of particular interest in view of the emerging fields of molecular electronics and optoelectronics which could one day replace the conventional technology relying on semiconductors such as silicon and gallium arsenide. The goal is the substitution of these inorganic materials by cheap and flexible layers of semiconducting organic molecules for a new class of diodes and transistors, as well as the realization of electronic switches based on individual molecules. One fundamental approach is to take advantage of the molecular self-assembly behavior which results in the creation of well-ordered supramolecular structures. The investigations of the self-assembly of rubrene adsorbed on metal surfaces (Au(111), Au(100), Ag(111), and Ag(100)), which were carried out within the framework of this thesis, show a surprising diversity of supramolecular structures. Amongst other shapes, the molecules organize themselves into geometries of perfect hexagonal and pentagonal symmetry and create multifaceted patterns on the surface. A fascinating peculiarity consists in the spontaneous construction of nested structures which are built up by a hierarchical self-assembly of individual molecules into pentagonal supermolecules which form in a second step perfect supramolecular decagons. The geometric shape of rubrene is characterized by a structural asymmetry leading to the existence of two mirror imaged versions of the molecule which are not superimposable to each other, such as for instance our left and right hand or the helical DNA. The aspect of chirality is crucial for basic processes in living systems and calls for a fundamental understanding of the interaction mechanisms occurring between chiral molecules. The experiments on rubrene reveal that the intermolecular bonding differentiates between the two chiral types of the molecule (chiral recognition), yielding the self-organization into homochiral structures. These assemblies exhibit a geometry which is again chiral, demonstrating a propagation of chirality throughout the three stages of the supramolecular hierarchy. The semiconducting behavior of rubrene is furthermore probed by STS measurements detecting the energetic positions of the highest occupied molecular orbital (HOMO) and the lowest unoccupied molecular orbital (LUMO). The experimental data uncover that different adsorption conformations exhibit characteristic HOMO energies and reveal adsorption conformations of rubrene which preserve the intrinsic electronic structure of the free molecule. Furthermore, a switching of the molecular conformation and the electronic structure of one rubrene conformer is induced with the STM.
