An essential Aurora-related kinase transiently associates with spindle pole bodies during Plasmodium falciparum erythrocytic schizogony

Aurora kinases compose a family of conserved Ser/Thr protein kinases playing essential roles in eukaryotic cell division. To date, Aurora homologues remain uncharacterized in the protozoan phylum Apicomplexa. In malaria parasites, the characterization of Aurora kinases may help understand the cell cycle control during erythrocytic schizogony where asynchronous nuclear divisions occur. In this study, we revisited the kinome of Plasmodium falciparum and identified three Aurora-related kinases, Pfark-1, -2, -3. Among these, Pfark-1 is highly conserved in malaria parasites and also appears to be conserved across Apicomplexa. By tagging the endogenous Pfark-1 gene with the green fluorescent protein (GFP) in live parasites, we show that the Pfark-1-GFP protein forms paired dots associated with only a subset of nuclei within individual schizonts. Immunofluorescence analysis using an anti-a-tubulin antibody strongly suggests a recruitment of Pfark-1 at duplicated spindle pole bodies at the entry of the M phase of the cell cycle. Unsuccessful attempts at disrupting the Pfark-1 gene with a knockout construct further indicate that Pfark-1 is required for parasite growth in red blood cells. Our study provides new insights into the cell cycle control of malaria parasites and reports the importance of Aurora kinases as potential targets for new antimalarials.

During infection in humans, P. falciparum invades and refurbishes the red blood cells (RBCs) in order to persist and proliferate. Parasite survival depends on expression of a parasite encoded cytoadherence ligand at the surface of the infected RBC called PfEMP1 (P. falciparum erythrocyte membrane protein 1). PfEMP1 comprises a N-terminal extracellular part, which mediates binding to receptors at the host endothelium. It also contains a C-terminal part which is conserved among all PfEMP1 molecules. This acidic terminal segment (ATS) anchors PfEMP1 to the membrane of infected RBCs and has a role for trafficking. To bring proteins to the RBC surface, P. falciparum builds its own trafficking machinery de novo since the erythrocytes do not contain secretory organelles. These new structures include parasite derived compartments called Maurer's clefts (MCs) located at the RBC periphery. The resident MCs protein MAHRP1 (membrane-associated histidine-rich protein 1) has been shown to be critical for PfEMP1 trafficking. A MAHRP1 KO strain had no PfEMP1 on the RBC surface but accumulated PfEMP1 within the parasite. MAHRP1 is a 28.9kDa protein with 249 amino acids containing a N-terminal, a transmembrane, and a C-terminal domain comprising histidine-rich repeats. To re-establish PfEMP1 trafficking, the MAHRP1 KO parasite line was complemented with various truncated HA-tagged fragments of MAHRP1. Immunofluorescent assays (IFAs) showed that the Cterminal domain with the histidine-rich repeats was not essential for PfEMP1 trafficking.

were exported to MCs and restored trafficking of PfEMP1. Shorter MAHRP1 fragments were not exported to MCs and PfEMP1 transport remained impaired. New transfection constructs were designed including the C-terminal part to identify important domain(s) of MAHRP1 for PfEMP1 trafficking. To identify other proteins interacting with PfEMP1, the ATS domain of PfEMP1 was recombinantly expressed and used in pull-down experiments. In these experiments using parasite lysates, PF14_0377 was identified as a potential interaction partner. This protein has been described as putative, vesicular-associated membrane protein. To subsequently assess the interaction(s) and the localization(s), PF14_0377 was cloned and transfected in 3D7 for IFAs. Due to time restrictions all these new transfectants could not be analyzed completely

Malaria Parasite-Infected Erythrocytes Secrete PfCK1, the Plasmodium Homologue of the Pleiotropic Protein Kinase Casein Kinase 1

Casein kinase 1 (CK1) is a pleiotropic protein kinase implicated in several fundamental processes of eukaryotic cell biology. Plasmodium falciparum encodes a single CK1 isoform, PfCK1, that is expressed at all stages of the parasite's life cycle. We have previously shown that the pfck1 gene cannot be disrupted, but that the locus can be modified if no loss-of-function is incurred, suggesting an important role for this kinase in intra-erythrocytic asexual proliferation. Here, we report on the use of parasite lines expressing GFP- or His-tagged PfCK1 from the endogenous locus to investigate (i) the dynamics of PfCK1 localisation during the asexual cycle in red blood cells, and (ii) potential interactors of PfCK1, so as to gain insight into the involvement of the enzyme in specific cellular processes. Immunofluorescence analysis reveals a dynamic localisation of PfCK1, with evidence for a pool of the enzyme being directed to the membrane of the host erythrocyte in the early stages of infection, followed by a predominantly intra-parasite localisation in trophozoites and schizonts and association with micronemes in merozoites. Furthermore, we present strong evidence that a pool of enzymatically active PfCK1 is secreted into the culture supernatant, demonstrating that PfCK1 is an ectokinase. Our interactome experiments and ensuing kinase assays using recombinant PfCK1 to phosphorylate putative interactors in vitro suggest an involvement of PfCK1 in many cellular processes such as mRNA splicing, protein trafficking, ribosomal, and host cell invasion.

Pre-replication complex organization in the atypical DNA replication cycle of Plasmodium falciparum: characterization of the mini-chromosome maintenance (MCM) complex formation

The overall organization of cell division in Plasmodium is unique compared to that observed in model organisms because DNA replicates more than once per cell cycle at several points of its life cycle. The sequencing of the Plasmodium genome has also revealed the apparent absence of many key components (e.g. Cdt1, DDK and Cdc45) of the eukaryotic cell cycle machinery that are responsible for the formation of the pre-replication complex (pre-RC). We have characterized the Plasmodium falciparum minichromosome maintenance complex (MCM) that plays a key role in the transition of pre-RC to the RC. Similar to other eukaryotes, the Plasmodium genome encodes six MCM subunits. Here, we show that expression levels of at least three of the PfMCM subunits, the homologues of MCM2, MCM6 and MCM7, change during the intraerythrocytic development cycle, peaking in schizont and decreasing in the ring and trophozoite stages. PfMCM2, 6 and 7 subunits interact with each other to form a developmentally regulated complex: these interactions are detectable in rings and schizonts, but not in trophozoites. PfMCM2, 6 and 7 subunits are localized in both cytosolic and nucleosolic fractions during all intraerythrocytic stages of P. falciparum development, with increased nuclear localization in schizonts. Only PfMCM6 is associated with the chromatin fraction at all stages of growth. No phosphorylation of PfMCM2, 6 and 7 was detected, but two as yet unidentified threonine-phosphosphorylated proteins were present in the complex, whose pattern of phosphorylation varied during parasite development.
