Structural mediation of human brain activity revealed by white-matter interpolation of fMRI

Understanding how the anatomy of the human brain constrains and influences the formation of large-scale functional networks remains a fundamental question in neuroscience. Here, given measured brain activity in gray matter, we interpolate these functional signals into the white matter on a structurally-informed high-resolution voxel-level brain grid. The interpolated volumes reflect the underlying anatomical information, revealing white matter structures that mediate the interaction between temporally coherent gray matter regions. Functional connectivity analyses of the interpolated volumes reveal an enriched picture of the default mode network (DMN) and its subcomponents, including the different white matter bundles that are implicated in their formation, thus extending currently known spatial patterns that are limited within the gray matter only. These subcomponents have distinct structure-function patterns, each of which are differentially observed during tasks, demonstrating plausible structural mechanisms for functional switching between task-positive and -negative components. This work opens new avenues for the integration of brain structure and function, and demonstrates the collective mediation of white matter pathways across short and long-distance functional connections.

Exploring dynamic functional connectivity by incorporating prior knowledge of brain structure

The synchronized firing of distant neuronal populations gives rise to a wide array of functional brain networks that underlie human brain function. Given the enormous perception, learning, and cognition potential of the human brain, it is not surprising that network-level representations of brain function reveal intrinsically rich organizational structure. What remains puzzling is how the human brain maintains its vast repertoire of functional connectivity (FC) states despite being constrained by the underlying fixed anatomical substrate. Advances in modern neuroimaging technologies, such as diffusion-weighted magnetic resonance imaging (DW-MRI), have made it possible to map the brain's anatomical scaffold, while functional MRI (fMRI) provides complementary information on neural activity. In this thesis, we develop and apply methods for combining DW-MRI and fMRI data into an integrated framework to analyze the interplay between brain structure and function. We first explored the dynamics of brain function during wakefulness and across the different non-rapid eye movement (NREM) sleep stages. We applied the innovation-driven co-activation pattern (iCAP) analysis to uncover the spatial and temporal organization of overlapping large-scale brain networks. Our results reveal new spatial patterns covering regions that support the physiological organization of sleep and arousal. Contrary to the previously observed decreasing FC that accompanies increasing sleep depth, we instead observe a surge of network activity and cross-network interactions during NREM stage 2, followed by an abrupt decrease in NREM stage 3. In the next step of the thesis, we propose a new method that models all brain voxels as nodes of a high-resolution voxel-level brain graph. We provide two ways to construct the graph and we characterize their properties by performing a spectral analysis of the graph Laplacian operator. Our findings show that despite the huge dimensionality of the proposed brain graphs, the majority of the structural information is captured by the Laplacian's lowest frequency eigenmodes. The lower end of the Laplacian spectra also captures about 85% of the energy content of functional MRI, suggesting that functional patterns are overall smooth over the structure, thus providing, for the first time, a direct and quantitative measure of how much brain function is shaped by the anatomy. Going beyond a scalar measure of the SC-FC link, we introduce a new framework that interpolates gray matter signals onto the white matter using the structure embedded in the voxel-level brain grid to guide the process. This enables visualization of key white matter structures that link temporally coherent gray matter areas. We found whole-brain structure-function networks that extend currently known spatial patterns that are limited within the gray matter only. Finally, we assessed the collective mediation of white matter pathways by giving a quantitative measure of the overall anatomical range between temporally coherent gray matter areas. We utilized a canonical model of graph diffusion to extract the anatomical range of functional network interactions. We find that this measure meaningfully differentiates brain regions according to a behaviorally relevant macroscale gradient that divides the cortex between low-level primary sensory areas and high-level cognitive functions.

The monocarboxylate transporter MCT2 belongs to a large family of membrane proteins involved in the transport of lactate, pyruvate and ketone bodies. Although its expression in rodent brain has been well documented, the presence of MCT2 in the human brain has been questioned on the basis of low mRNA abundance. In this study, the distribution of the monocarboxylate transporter MCT2 has been investigated in the cortex of normal adult human brain using an immunohistochemical approach. Widespread neuropil staining in all cortical layers was observed by light microscopy. Such a distribution was very similar in three different cortical areas investigated. At the cellular level, the expression of MCT2 could be observed in a large number of neurons, in fibers both in grey and white matter, as well as in some astrocytes, mostly localized in layer I and in the white matter. Double staining experiments combined with confocal microscopy confirmed the neuronal expression but also suggested a preferential postsynaptic localization of synaptic MCT2 expression. A few astrocytes in the grey matter appeared to exhibit MCT2 labelling but at low levels. Electron microscopy revealed strong MCT2 expression at asymmetric synapses in the postsynaptic density and also within the spine head but not in the presynaptic terminal. These data not only demonstrate neuronal MCT2 expression in human, but since a portion of it exhibits a distinct synaptic localization, it further supports a putative role for MCT2 in adjustment of energy supply to levels of activity.

Introduction: Medial Temporal Lobe Epilepsy (mTLE) with hippocampal sclerosis is the most frequent cause of drug-resistant focal epilepsy in adults. These patients suffer from widespread subtle white matter abnormalities and abnormal functional connectivity extending beyond the affected lobe, as revealed by Diffusion Tensor Imaging and functional MRI studies. Diffusion spectrum imaging (DSI) is a new diffusion imaging technique with high angular resolution for improving the mapping of white matter tracts [1]. In this study, we used DSI to investigate the whole brain effect of TLE as reflected by topological measures of the structural connectivity network. Methods: Eight patients with right-sided mTLE and hippocampal sclerosis and 25 controls underwent our DSI protocol at 3T using a 32 channel coil: DSI (voxel size 2.2x2.2x3mm; 44 slices, 257 diffusion directions; max b-value = 6400 s/mm2), 3D T1-weighted MPRAGE and 3D T2-weighted images. The analysis was performed using the Connectome Mapper. The cortical and subcortical grey matters were parcellated into 86 Regions of Interest (ROI) with anatomical landmarks using Freesurfer 5.0. The ROIs were coregistered to the diffusion data by using a nonlinear registration between T1 and T2, then T2 to the diffusion space. The Diffusion Tool Kit was used for the reconstruction of the diffusion Orientation Distribution Function in each voxel. Streamline fibre-tracking was then performed from each voxel in the white matter areas using an in-house streamline-based algorithm. Two scalar maps, the Fractional Anisotropy and the Generalized Fractional Anisotropy were calculated. For each patient, the connectivity between every region pair was estimated was used to construct a connectivity matrix (the adjacency matrix of the structural network). Finally we used topological measures in order to reduce the data and obtain whole-brain characteristics of the network [2]: characteristic Path Length (L), Clustering Coefficient (C) and Small Worldness (S) where S=(C/Crand)/(L/ Lrand) with Crand and Lrand are computed for a random network. Significant differences between the mTLE and control groups were assessed using a Wilcoxon rank-sum test. Results: In patients, we found a higher characteristic path length (0.0288), with a lower clustering coefficient (0.0079) and reduced small-worldness (p=0.000058) compared to controls. Network nodes that contributed significantly to these alterations were located within as well as outside the temporal lobe in both hemispheres, although with a predominance for the ipsilateral hemisphere and bilateral limbic structures. The pattern of significant nodes showed concordance with brain regions known to be involved in epileptic networks in patients with mTLE. Conclusions: Our results show a reduced efficiency of structural brain networks and altered connectivity patterns that are concordant with the mapping of functional epileptic networks [3] and altered functional connectivity [4] in patients with mTLE. Therefore, the network in mTLE is less segregated than in controls with reduced global interactions between the nodes and reduced specialised communities. Further studies are needed to establish the relevance of these findings with respect to the propagation of epileptic activity, cognitive deficits in mTLE and outcome of epilepsy surgery in individual patients.
