This lecture covers the application of microarray technology in genomic data analysis, focusing on Affymetrix GeneChip Probe Arrays. Topics include signal quantification, array design, image analysis, artifacts in microarrays, and data preprocessing techniques such as exploratory data analysis and normalization. The instructor explains the process of measuring gene expression, background correction, and quantile normalization. Different methods for differential expression analysis, such as MAS 5.0, Li-Wong, and RMA, are discussed, along with the bias-variance tradeoff. Conclusions from studies comparing various methods are presented, highlighting the importance of choosing an appropriate analysis approach.
