Learning disability, learning disorder, or learning difficulty (British English) is a condition in the brain that causes difficulties comprehending or processing information and can be caused by several different factors. Given the "difficulty learning in a typical manner", this does not exclude the ability to learn in a different manner. Therefore, some people can be more accurately described as having a "learning difference", thus avoiding any misconception of being disabled with a possible lack of an ability to learn and possible negative stereotyping. In the United Kingdom, the term "learning disability" generally refers to an intellectual disability, while conditions such as dyslexia and dyspraxia are usually referred to as "learning difficulties". Theory of representation learning in cortical neural networks

Our brain continuously self-organizes to construct and maintain an internal representation of the world based on the information arriving through sensory stimuli. Remarkably, cortical areas related to different sensory modalities appear to share the same functional unit, the neuron, and develop through the same learning mechanism, synaptic plasticity. It motivates the conjecture of a unifying theory to explain cortical representational learning across sensory modalities. In this thesis we present theories and computational models of learning and optimization in neural networks, postulating functional properties of synaptic plasticity that support the apparent universal learning capacity of cortical networks. In the past decades, a variety of theories and models have been proposed to describe receptive field formation in sensory areas. They include normative models such as sparse coding, and bottom-up models such as spike-timing dependent plasticity. We bring together candidate explanations by demonstrating that in fact a single principle is sufficient to explain receptive field development. First, we show that many representative models of sensory development are in fact implementing variations of a common principle: nonlinear Hebbian learning. Second, we reveal that nonlinear Hebbian learning is sufficient for receptive field formation through sensory inputs. A surprising result is that our findings are independent of specific details, and allow for robust predictions of the learned receptive fields. Thus nonlinear Hebbian learning and natural statistics can account for many aspects of receptive field formation across models and sensory modalities. The Hebbian learning theory substantiates that synaptic plasticity can be interpreted as an optimization procedure, implementing stochastic gradient descent. In stochastic gradient descent inputs arrive sequentially, as in sensory streams. However, individual data samples have very little information about the correct learning signal, and it becomes a fundamental problem to know how many samples are required for reliable synaptic changes. Through estimation theory, we develop a novel adaptive learning rate model, that adapts the magnitude of synaptic changes based on the statistics of the learning signal, enabling an optimal use of data samples. Our model has a simple implementation and demonstrates improved learning speed, making this a promising candidate for large artificial neural network applications. The model also makes predictions on how cortical plasticity may modulate synaptic plasticity for optimal learning. The optimal sampling size for reliable learning allows us to estimate optimal learning times for a given model. We apply this theory to derive analytical bounds on times for the optimization of synaptic connections. First, we show this optimization problem to have exponentially many saddle-nodes, which lead to small gradients and slow learning. Second, we show that the number of input synapses to a neuron modulates the magnitude of the initial gradient, determining the duration of learning. Our final result reveals that the learning duration increases supra-linearly with the number of synapses, suggesting an effective limit on synaptic connections and receptive field sizes in developing neural networks.

Elucidating the role of neuron-astrocyte metabolic coupling in learning and memory

The brain has very high energy demands that are mainly met by the circulating blood glucose to ensure its proper functioning. Thus, it is not surprising that though the human brain weighs only 2- 3% of the body weight, it consumes approximately 25% of total body glucose. “Neuro-energetic coupling" is a unique feature of the brain and refers to the existence of the tight coupling between (neuronal) synaptic brain activity and energy metabolism. The main excitatory neurotransmitter in the brain is glutamate and most of the energy requirements at the cortical level are met through glutamate-mediated neurotransmission, suggesting that there is a close relationship between brain activity, glutamatergic neurotransmission and energy metabolism. Until the 80s, blood-borne glucose was thought to be the sole energy substrate of the brain cells and it was assumed that glucose is metabolized by neurons and astrocytes (a type of glial cell) independently. It is only during the last two decades that the concept of compartmentalization of energy metabolism between neurons and astrocytes has become more obvious. The transfer of lactate from astrocytes to neurons termed as the “astrocyte-neuron lactate shuttle (ANLS)” model proposed by Dr. Pellerin and Prof. Magistretti posits that glutamate released during brain activation activates glycolysis (one of glucose metabolic pathways) in astrocytes leading to lactate (a monocarboxylate) release in the extracellular space, and this astrocytic lactate is then captured and used as an energy substrate by the activated neurons to meet their energy requirements. It includes a complex chain of events involved in neuro-metabolic coupling and supports the preferential use of lactate by activated neurons under conditions of increased energy demands. Higher brain functions such as learning a new task are associated with structural changes in brain and are metabolically demanding necessitating the need of an additional energy supply to meet the neurons increased energy requirement. In this context, the current thesis project aimed to elucidate the contribution of metabolic coupling between astrocytes and neurons, known as “neuron-astrocyte metabolic coupling” to learning and memory formation. Using an in vivo approach combined with behavioral, molecular and gene manipulation techniques in mice, we set out to investigate the role of neuron-astrocyte metabolic coupling, particularly ANLS in cognition. The project was divided into two main parts: In the first part, the expression of glucose metabolism-related genes was investigated during long term memory (LTM) formation in fear-motivated inhibitory avoidance (IA) learning paradigm. Using (14C) 2-Deoxyglucose (2-DG) technique, we first defined the brain areas that are involved in IA LTM formation. This technique provides an indirect measurement of brain activity by measuring glucose uptake in different brain areas during the task. The results of brain metabolic mapping show an increase in glucose utilization in the hippocampus, amygdala, anterior cingulate cortex and pre-limbic and infra-limbic cortical areas. Results from the quantitative analysis of mRNA levels in the dorsal hippocampus at different time point’s following IA task highlight a time dependent, learning specific change in the expression of genes related to glucose metabolism. Specific set of genes involved in the synthesis and degradation of glycogen (brain glucose reserve present only in astrocytes), pyruvate metabolism and pentose phosphate shunt as well as the ANLS were induced following the IA task. These early observations indicate that the metabolic interactions between astrocytes and neurons undergo modifications during a learning process and suggest that these adaptations may play a key role in the establishment of long-term memory. In the second part of research project, we focused on investigating the behavioral consequences of down regulating the expression of Monocarboxylate Transporter 1 (MCT1), a key ANLS related gene, on cognition and higher brain functions. Mice heterozygous for the MCT1 gene (MCT1+/-) were characterized in numerous behavioral paradigms such as general wellbeing, reflexive and motor capabilities. After having established that MCT1+/- mice have no alterations in general emotional state or in locomotion, a battery of behavioral tests was performed to study cognition in these mice. Compared with wild-type control mice, the MCT1+/- mice display normal muscle strength and motor coordination. However, when tested for higher brain functions, the MCT1+/- mice exhibit learning and memory deficits in several learning paradigms/tasks that are hippocampus and / or amygdala dependent. The cognitive impairment observed in the MCT1 heterozygous mice further highlights the importance of ANLS in memory and suggest that disruption of lactate transfer from astrocytes to neurons via the MCT1 results in cognitive impairment. The overall results in this thesis demonstrate that cerebral energy metabolism, and in particular the transfer of lactate from astrocytes to neurons not only provides temporal adaptations in the learning process, but it also plays a fundamental role in memory formation.

Analysis of EEG Correlates of Perceived Difficulty in Dynamically Changing Flying Tasks

Real-time workload estimation can be an important tool to improve human-machine interaction. Despite multiple efforts in this sense, most studies focused on tasks designed to have extreme conditions of workload (high vs low), and these levels remain rather constant throughout task execution. However, some applications may induce changing levels of workload and it is not clear how decoders trained in the extreme conditions will perform. In this study we study this scenario in a simulated drone task where participants had to fly through several waypoints. The task difficulty was modulated by the size of waypoints; in one of the studied conditions, the size was modulated in real time by the EEG-based decoding of the perceived difficulty. We show that this protocol can effectively induce different levels of workload and perceived difficulty. Furthermore, post-experiment analysis using a sparse-regularized technique supports the feasibility of decoding perceived difficulty in this dynamically changing task above chance level (average class-balanced accuracy across subjects 67% +/- 7%). Interestingly, an analysis of the selected features in independent recording sessions showed that roughly 20% of the features were stable across multiple sessions.
